    Keratinocyte Responsive Element 3: Analysis of a Keratinocyte-specific Regulatory Sequence in the 230-kDa Bullous Pemphigoid Antigen Gene Promoter

    The 230-kDa bullous pemphigoid antigen gene is expressed primarily, if not exclusively, in basal keratinocytes of the epidermis. Keratinocyte responsive element 3, a cis-element at position ā€“216 to ā€“197 of the human 230-kDa bullous pemphigoid antigen gene promoter, confers tissue-specific expression to this gene (Tamai et al: J Biol Chem 270:7609ā€“7614, 1995). In this study, we investigated the functional characteristics of keratinocyte responsive element 3 on the 230-kDa bullous pemphigoid antigen gene core promoter by transient transfections of cultured normal human keratinocytes and normal human fibroblasts, as well as of lung carcinoma (A549), osteosarcoma (OST), and gastric adenocarcinoma (GT3TKB) cell lines. A 230-kDa bullous pemphigoid antigen gene core promoter/luciferase reporter gene plasmid construct, pBPL, was modified to develop a series of constructs (pKBPLā€“p4KBPL), which have insertions of one, two, three, or four tandem repeats of keratinocyte responsive element 3, and these plasmids were used in transient transfections of the cultured cells. The promoter activities of pKBPLā€“p4KBPL constructs, relative to pBPL, in normal human keratinocytes were 7.6-, 15.5-, 4.6-, and 2.7-fold higher, respectively, whereas no upregulatory effect by keratinocyte responsive element 3 insertion was observed in other cell lines tested. prKBPL, a plasmid constructed with keratinocyte responsive element 3 in reverse orientation, showed essentially no activity in normal human keratinocytes. Insertion of a random 20 bp sequence between keratinocyte responsive element 3 and the 230-kDa bullous pemphigoid antigen gene core promoter resulted in about 40% reduction of luciferase activity in normal human keratinocytes. These data suggest that keratinocyte responsive element 3 functions as a position-, copy number-, and orientation-dependent cis-element contributing to tissue-specific regulation of the 230-kDa bullous pemphigoid antigen gene

    High-Energy Electron Transfer Dissociation (HE-ETD) Using Alkali Metal Targets for Sequence Analysis of Post-Translational Peptides

    Post-translational modifications (PTMs) of proteins are important in the activation, localization, and regulation of protein function in vivo. The usefulness of electron capture dissociation (ECD) and electron-transfer dissociation (ETD) in tandem mass spectrometry (MS/MS) using low-energy (LE) trap type mass spectrometer is associated with no loss of a labile PTM group regarding peptide and protein sequencing. The experimental results of high-energy (HE) collision induced dissociation (CID) using the Xe and Cs targets and LE-ETD were compared for doubly-phosphorylated peptides TGFLT(p)EY(p)VATR (1). Although HE-CID using the Xe target did not provide information on the amino acid sequence, HE-CID using the Cs target provided all the z-type ions without loss of the phosphate groups as a result of HE-ETD process, while LE-ETD using fluoranthene anion gave only z-type ions from z5 to z11. The difference in the results of HE-CID between the Xe and Cs targets demonstrated that HE-ETD process with the Cs target took place much more dominantly than collisional activation. The difference between HE-ETD using Cs targets and LE-ETD using the anion demonstrated that mass discrimination was much weaker in the high-energy process. HE-ETD was also applied to three other phosphopeptides YGGMHRQEX(p)VDC (2: X = S, 3: X = T, 4: X = Y). The HE-CID spectra of the doubly-protonated phosphopeptides (= [M + 2H]2+) of 2, 3, and 4 using the Cs target showed a very similar feature that the c-type ions from c7 to c11 and the z-type ions from z7 to z11 were formed via Nā€“CĪ± bond cleavage without a loss of the phosphate group

    ABSTRACT Anti-symmetrically laminated composites have coupling effects between tensile stress and twisting deformation, and are very attractive as fan blade materials of aircraft engines. Blades fabricated by anti-symmetrically laminated composites can automatically adjust the stagger angle to better aerodynamic conditions with change of axial force or rotational speed owing to the coupling effects. Thus, the anti-symmetrically laminated composite blades are expected to improve aerodynamic efficiency and the stability of aircraft engines.     Anti-IL-6 Receptor Antibody Causes Less Promotion of Tuberculosis Infection than Anti-TNF-Ī± Antibody in Mice

    Objective. Our aim was to investigate the effects of IL-6 blockade on the progression of Mycobacterium tuberculosis (TB) and compare them with those of TNF-Ī± blockade in mice. Methods. Mice were intravenously infected with TB and injected with antibodies. Survival was monitored and histological and immunological studies were carried out. Results. All anti-IL-6R Ab-treated mice and 8 of 10 control mice survived until sacrificed 224 days after TB challenge, whereas anti-TNF-Ī± Ab-treated mice all died between 120 and 181 days. Anti-IL-6R Ab-treated mice exhibited no significant differences in TB CFU in organs, including the lungs, and no deterioration in histopathology compared to control mice at 4 weeks. In contrast, anti-TNF-Ī± Ab-treated mice exhibited increased TB CFU and greater progression of histopathological findings in organs than control mice. Spleen cells from anti-TNF-Ī± Ab-treated mice had decreased antigen-specific response in IFN-Ī³ release and proliferation assays. The results in anti-IL-6R Ab-treated mice suggest that spleen cell responses were decreased to a lesser degree. Similar results were obtained in IL-6 knockout (KO) mice, compared with TNF receptor 1 (TNFR1) KO and TNFR1/IL-6 double KO (DKO) mice. Conclusion. IL-6R blockade promotes the progression of TB infection in mice far less than TNF-Ī± blockade

    Near-Infrared Imaging Polarimetry of S106 Cluster-Forming Region with SIRPOL

    We present the results of wide-field JHKs polarimetry toward the HII region S106 using the IRSF (Infrared Survey Facility) telescope. Our polarimetry data revealed an extended (up to ~ 5') polarized nebula over S106. We confirmed the position of the illuminating source of most of the nebula as consistent with S106 IRS4 through an analysis of polarization vectors. The bright portion of the polarized intensity is consistent with the red wing component of the molecular gas. Diffuse polarized intensity emission is distributed along the north--south molecular gas lanes. We found the interaction region between the radiation from S106 IRS4 and the dense gas. In addition, we also discovered two small polarization nebulae, SIRN1 and SIRN2, associated with a young stellar objects (YSO). Aperture polarimetry of point-like sources in this region was carried out for the first time. The regional magnetic field structures were derived using point-like source aperture polarimetry, and the magnetic field structure position angle around the cluster region in S106 was found to be ~ 120\arcdeg. The magnetic fields in the cluster region, however, have three type position angles: ~ 20\arcdeg, ~ 80\arcdeg, and ~ 120\arcdeg. The present magnetic field structures are consistent with results obtained by submillimeter continuum observations. We found that the magnetic field direction in the dense gas region is not consistent with that of the low density gas region.Comment: 22 pages, 11 figures, accepted for publication in A
