    Pre-elementary Children With Imperfect Letter-Name Knowledge Are at Great Risk of Reading Difficulty in First Grade : One-Year Longitudinal Study in Japanese Hiragana

    While letter-naming ability is a well-known preschool predictor of the later acquisition of literacy, little is known about an appropriate benchmark (i.e., how many letter names children must know at a given age) and how it may vary among different writing systems. The present study aimed to establish a letter-naming benchmark in Japanese Hiragana for pre-elementary children (age 5 to 6 years) and examined whether this benchmark predicts risk or success in later reading development via a one-year longitudinal survey. Children (N = 291) were assessed once in their pre-elementary year for Hiragana-naming accuracy and once in their first-grade year for oral reading fluency. As a result, the ability to name 40 of 45 letters was determined to be an optimal cut-off, and failure to meet it strongly predicted a risk of deficient reading fluency in first grade. These findings support the notion that Japanese children without near-perfect mastery of Hiragana-naming in their pre-elementary year are at great risk of reading difficulty in first grade. In addition, possible contrasts between Hiragana- and alphabet-naming indicated a need for further research in different languages and scripts to establish appropriate goals and policies for this foundational skill of reading in early education.Peer reviewe

    Dicer is an RNase III enzyme with two catalytic subunits, which catalyzes the cleavage of double-stranded RNA to small interfering RNAs and micro-RNAs, which are mainly involved in invasive nucleic acid defense and endogenous genes regulation. Dicer is abundantly expressed in embryos, indicating the importance of the protein in early embryonic development. In addition, Dicer is thought to be involved in defense mechanism against foreign nucleic acids such as viruses. This paper will mainly focus on the recent progress of Dicer-related research and discuss potential RNA interference pathways in aquatic species

    The Role of Apelin on the Alleviative Effect of Angiotensin Receptor Blocker in Unilateral Ureteral Obstruction-Induced Renal Fibrosis

    Background: Apelin is a selective endogenous ligand of the APJ receptor, which genetically has closest identity to the angiotensin II type 1 receptor (AT-1). The effects of the apelin/APJ system on renal fibrosis still remain unclear. Methods: We examined the effects of the apelin/APJ system on renal fibrosis during AT-1 blockade in a mouse unilateral ureteral obstruction (UUO) model. Results: We obtained the following results: (1) At UUO day 7, mRNA expressions of apelin/APJ and phosphorylations of Akt/endothelial nitric oxide synthase (eNOS) in the UUO kidney were increased compared to those in the nonobstructed kidney. (2) AT-1 blockade by the treatment with losartan resulted in a further increase of apelin mRNA as well as phosphorylations of Akt/eNOS proteins, and this was accompanied by alleviated renal interstitial fibrosis, decreased myofibroblast accumulation, and a decreased number of interstitial macrophages. (3) Blockade of the APJ receptor by the treatment with F13A during losartan administration completely abrogated the effects of losartan in the activation of the Akt/eNOS pathway and the amelioration of renal fibrosis. (4) Inhibition of NOS by the treatment with L-NAME also resulted in a further increase in renal fibrosis compared to the control group. Conclusion: These results suggest that increased nitric oxide production through the apelin/APJ/Akt/eNOS pathway may, at least in part, contribute to the alleviative effect of losartan in UUO-induced renal fibrosis

    子どものいるがん患者の家族への看護: 必要とされるケアについての看護者の認識

    本研究は、がん患者の子どもへの看護を行う看護師の認識を明らかにすることを目的とし た。  調査方法は、子育て期にあるがん患者の子どもへの看護についての経験を持つ看護師4 名 を対象に、実際のケアの経緯やケアの根拠、考えや思いについて半構成的面接を実施した。 本研究は事例研究とした。  分析は、女性のがん患者の子どもへの実際のケアとケアに至るまでの考えを文脈ごと抜き 出し、「ケアの経緯や根拠の看護の認識」と「これまでの実際のケア」の視点から解釈しカ テゴライズした。  結果として以下の4 点が明らかになった。1. 患者や家族のライフステージを見通した関わ り2. 病院外での子どもと他者の関係性をイメージした関わり3. 子どもからの視点を持った 関わり4. 専門看護師としての高い役割意識と内省を生かした関わりであった。看護師は、が ん患者の子どもにまでケアの視点を広げることによって、患者としての困りごとだけではな く、患者が持つ母親という役割、またその子どもが必要とする家族や学校などの社会の状況 まで視野に入れて関わっていた。  本研究の結果から、がん患者の家族の一員である子どもへの関わりとして、急性期、慢性期、 ターミナル期と治療期をある期間で区切った関わりではなく、母親ががんと告知されたその 時から様々な進行状況を想定した見通しのある関わりをいかに看護者が想定できるかが必要 であると考えられた。がん患者は、家族の中で様々な役割を持っており、その人の家族での 役割にまで思いを及ばせることによって、看護師の一方方向の看護でなく、患者や家族と協 力することができ、様々に起こりうる患者や家族の問題について良い方向に導くことに繋が ると考えられる。  以上より、がん患者の看護として患者の家族である子どもにも見通しをもって関わってい くことが重要である

    ERK Regulates Renal Cell Proliferation and Renal Cyst Expansion in inv Mutant Mice

    Nephronophthisis (NPHP) is the most frequent genetic cause of end-stage kidney disease in children and young adults. Inv mice are a model for human nephronophthisis type 2 (NPHP2) and characterized by multiple renal cysts and situs inversus. Renal epithelial cells in inv cystic kidneys show increased cell proliferation. We studied the ERK pathway to understand the mechanisms that induce cell proliferation and renal cyst progression in inv kidneys. We studied the effects of ERK suppression by administering PD184352, an oral mitogen-activated protein kinase kinase (MEK) inhibitor on renal cyst expansion, extracellular signal-regulated protein kinase (ERK) activity, bromo-deoxyuridine (BrdU) incorporation and expression of cell-cycle regulators in invΔC kidneys. Phosphorylated ERK (p-ERK) level increased along with renal cyst enlargement. Cell-cycle regulators showed a high level of expression in invΔC kidneys. PD184352 successfully decreased p-ERK level and inhibited renal cyst enlargement. The inhibitor also decreased expression of cell-cycle regulators and BrdU incorporation in renal epithelial cells. The present results showed that ERK regulated renal cell proliferation and cyst expansion in inv mutants

    Jumping to conclusion bias in adolescents with ASD

    Background and Purpose: Jumping to conclusion (JTC)—a cognitive bias in thinking processes—leads to drawing conclusions based on little information, and could be related to psychosis and paranoia. While it has recently been pointed out that it could accompany the autism spectrum disorder (ASD), no interventions targeting this bias in adolescents with ASD have been reported. Therefore, this exploratory study investigated the effects of a group social cognition program on JTC bias in adolescents with ASD. Patients and Methods : Group rehabilitation using social cognition and interaction training (SCIT) was conducted for 12- to 18-year-old adolescents with ASD. An SCIT program comprehensively targets social cognitive functions, including interventions for JTC bias, and examines changes before and after the SCIT intervention, social cognitive functioning tasks, and subjective quality of life (QOL). Results : Thirteen adolescents with ASD participated in this program ; 10 (76.9%) stayed through it. The proportion of participants with JTC bias decreased significantly before and after SCIT (before : 7 / 10 ; after : 1 / 10 ; p = 0.041), and subjective QOL increased significantly (p = 0.014). Conclusion : The results show that a group social cognition program with a JTC bias approach improves the JTC bias and increases subjective QOL in adolescents with ASD

    Training Attention to the Other’s Need Improves Helping Behaviors in Children with Autism Spectrum Disorder: A Behavioral Analysis Using a Two-dimensional Motion Capture System

    While children with autism spectrum disorder (ASD) can acquire helping behaviors through appropriate interventions, changes in behaviors prior to helping (pre-helping behaviors) remain unclear. In the present study, we examined the effects of social skills training (SST) on helping and pre-helping behaviors in two children with ASD by using a two-dimensional motion capture system. During the SST, the children learned one helping behavior that they lent their items to their partners, and we measured their head movements before initiating the helping behavior (i.e., pre-helping behavior). As a result of SST, the participants became able to help others in response to less-explicit social stimuli after the intervention. Regarding pre-helping behaviors, the children with ASD before the intervention looked straight at the helpee (i.e., recipient of the help) more often than did typically developing peers, and such a behavior was shown to increase after SST. These results indicate that although spontaneous attention to social stimuli may be reduced in children with ASD, success in attending to a helpee could lead to the emergence of helping behaviors. Moreover, the changes in pre-helping behavior indicate an increase in children’s attention to the helpee after the intervention, which may have enhanced their sensitivity to persons in need

    Applicability of the Movement Assessment Battery for Children-Second Edition (MABC-2) for Japanese Children Aged 3–6 Years: A Preliminary Investigation Emphasizing Internal Consistency and Factorial Validity

    This study investigated the applicability of the Movement Assessment Battery for Children – Second Edition (MABC-2) for 3- to 6-year-old Japanese children, particularly addressing its internal consistency and factorial validity. The MABC-2 test set for 3- to 6-year-old children was administered to 252 children. Differences between Japanese children and those of the original normative sample (i.e. United Kingdom children) were investigated along with sex differences. The Japanese children aged 3–6 years were found to have higher Manual Dexterity and Balance component scores than children of the normative sample. Girls scored higher than boys on the Balance component. Results of several analyses showed good internal consistency of the MABC-2. Confirmatory factor analysis revealed that a theoretical three-component model of the MABC-2 was not fitted to Japanese children aged 3–6 years. Instead, a new three-component model was postulated and discussed. The new three-component model of the MABC-2, with Manual Dexterity, Static Balance and Ball Skills, and Dynamic Balance, has high factorial validity in Japanese children aged 3–6 years

    CNVs in Three Psychiatric Disorders

    BACKGROUND: We aimed to determine the similarities and differences in the roles of genic and regulatory copy number variations (CNVs) in bipolar disorder (BD), schizophrenia (SCZ), and autism spectrum disorder (ASD). METHODS: Based on high-resolution CNV data from 8708 Japanese samples, we performed to our knowledge the largest cross-disorder analysis of genic and regulatory CNVs in BD, SCZ, and ASD. RESULTS: In genic CNVs, we found an increased burden of smaller (500 kb) exonic CNVs in SCZ/ASD. Pathogenic CNVs linked to neurodevelopmental disorders were significantly associated with the risk for each disorder, but BD and SCZ/ASD differed in terms of the effect size (smaller in BD) and subtype distribution of CNVs linked to neurodevelopmental disorders. We identified 3 synaptic genes (DLG2, PCDH15, and ASTN2) as risk factors for BD. Whereas gene set analysis showed that BD-associated pathways were restricted to chromatin biology, SCZ and ASD involved more extensive and similar pathways. Nevertheless, a correlation analysis of gene set results indicated weak but significant pathway similarities between BD and SCZ or ASD (r = 0.25–0.31). In SCZ and ASD, but not BD, CNVs were significantly enriched in enhancers and promoters in brain tissue. CONCLUSIONS: BD and SCZ/ASD differ in terms of CNV burden, characteristics of CNVs linked to neurodevelopmental disorders, and regulatory CNVs. On the other hand, they have shared molecular mechanisms, including chromatin biology. The BD risk genes identified here could provide insight into the pathogenesis of BD
