    Haptoglobin genotyping by allele-specific polymerase chain reaction amplification

    We performed haptoglobin (Hp) genotyping by polymerase chain reaction using allele-specific primer-pairs. The major six genotypes of Hp were identified using this method. Among Japanese individuals living in Ehime and Okayama Prefectures, the allele frequencies were estimated to be Hp2 = 0.723 and Hp1s = 0.277. Genotyping of Hp was possible with 0.3 ng of DNA and with 0.125 microliter of blood. It was also possible with whole blood left at room temperature for a month and also with the bloodstains left at room temperature for three years. In the heated blood samples, both alleles, Hp2 and Hp1s, were detected in those heated at 100 degrees C for 2 h. In bloodstains, Hp2 and Hp1s were detected in samples heated at 100 degrees C for 2 h and 120 degrees C for 30 min. In addition, the genotype could be detected in samples other than blood such as saliva, hair roots, tissue sections and dental pulps. The present method for Hp genotyping is expected to become a useful method in forensic analysis.</p

    Effects of Aquatic Pole Walking on the Reduction of Spastic Hypertonia in a Patient with Hemiplegia: A Case Study

    Here we report an acute effect of aquatic pole walking (PW) training intervention on a 64-year-old male patient with chronic hemiparesis and symptoms of spasticity in the right lower limb. A comparison of over ground walking before and after 20 minutes of aquatic PW training revealed a significant improvement in gait performance. As a main result, the average speed of walking after the intervention was 0.16 m/s after the intervention as compared to 0.04 m/s in the initial condition. The time taken for each stride cycle was drastically decreased, mainly due to shortening of the stance time. Underlying the improved gait performance was the emergence of functional muscle activity in the paralyzed and spastic leg muscles. The result observed in this patient should be further tested among a large population of patients presenting similar symptoms. Moreover, the basic mechanisms underlying aquatic PW intervention should be further elucidated

    In the present experiments attempts were made to identify semen from various specimens such as the semen itself, spots of semen on clothes, putrefied semen or semen contaminated with blood, menstrual blood, vaginal fluid, according to the techniques of LEVONEN. As the result it has been clarified that in every instance it is possible to isolate and detect the spots of choline by spraying Dragehdorff's reagent.</p

    A Case of Paragonimus westermani Infection by Eating Imperfectly Cooked Wild Boar Flesh

    A 19 year old woman was admitted because of abnormal chest X-ray showing smoothly outlined cystic lesion. The eggs of Paragonimus westermani was identified in the broncho-alveolar lavage fluid. Anti-Paragonimus westermani antibody was positive in the serum by the ELISA method. Six eggs were observed in one gram of feces before the administration of praziquantel. Praziquantel (75mg/kg) was administered for two day, the egg of Paragonimus westermani disappeared in the feces and the size of cystic lesion in the chest X-ray decreased

    Genomic regions involved in yield potential detected by genome-wide association analysis in Japanese high-yielding rice cultivars

    BACKGROUND: High-yielding cultivars of rice (Oryza sativa L.) have been developed in Japan from crosses between overseas indica and domestic japonica cultivars. Recently, next-generation sequencing technology and high-throughput genotyping systems have shown many single-nucleotide polymorphisms (SNPs) that are proving useful for detailed analysis of genome composition. These SNPs can be used in genome-wide association studies to detect candidate genome regions associated with economically important traits. In this study, we used a custom SNP set to identify introgressed chromosomal regions in a set of high-yielding Japanese rice cultivars, and we performed an association study to identify genome regions associated with yield. RESULTS: An informative set of 1152 SNPs was established by screening 14 high-yielding or primary ancestral cultivars for 5760 validated SNPs. Analysis of the population structure of high-yielding cultivars showed three genome types: japonica-type, indica-type and a mixture of the two. SNP allele frequencies showed several regions derived predominantly from one of the two parental genome types. Distinct regions skewed for the presence of parental alleles were observed on chromosomes 1, 2, 7, 8, 11 and 12 (indica) and on chromosomes 1, 2 and 6 (japonica). A possible relationship between these introgressed regions and six yield traits (blast susceptibility, heading date, length of unhusked seeds, number of panicles, surface area of unhusked seeds and 1000-grain weight) was detected in eight genome regions dominated by alleles of one parental origin. Two of these regions were near Ghd7, a heading date locus, and Pi-ta, a blast resistance locus. The allele types (i.e., japonica or indica) of significant SNPs coincided with those previously reported for candidate genes Ghd7 and Pi-ta. CONCLUSIONS: Introgression breeding is an established strategy for the accumulation of QTLs and genes controlling high yield. Our custom SNP set is an effective tool for the identification of introgressed genome regions from a particular genetic background. This study demonstrates that changes in genome structure occurred during artificial selection for high yield, and provides information on several genomic regions associated with yield performance.     Antarctic micrometeorites collected at the Dome Fuji Station

    Antarctic micrometeorites (AMMs) were found among the precipitated fine particles recovered from a water tank in the Dome Fuji Station. These AMMs had been contained in the recent fallen snow around the station. Initial processing of the precipitated particles revealed that they were dominated by natural and artificial terrestrial materials, thus a series of processes were developed to separate AMMs from terrestrial particles. The recovery rate of AMMs by the processes was approximately 45% in weight, which was determined from a weight ratio of recovered/accreted AMMs. The micro-morphology and major-element concentration of the recovered AMMs were characterized. They appear to have been heated upon atmospheric entry to varying temperatures and can be classified into two major types based on the degree of heating : (1) fine-grained, irregular-shaped, partial-melted micrometeorites with chondritic composition, and (2) total-melted spherical micrometeorites with chondritic composition except for volatile elements. A digital catalog for the AMMs identified in this study was established on the web site [URL : http : //dust. cc. gakushuin. ac. jp/], in which optical characteristics, high-resolution images, and chemical compositions of individual AMMs are presented. The AMMs listed in the catalog are the first Japanese collection of extraterrestrial dust. The criterion and techniques developed for the selection and initial analysis of AMMs are applicable for the dust samples that are being collected by the 39th Japanese Antarctic Research Expedition team
