    Developmental and Anatomical Patterns Of IL-2 Gene Expression in Vivo in The Murine Thymus

    Interleukin-2 (IL-2) is a potent growth factor that mature T lymphocytes synthesize and use as a proliferation signal. Much controversy has arisen concerning whether it is used to drive the extensive proliferation of immature pre-T cells in the thymus. Immature thymocytes acquire the competence to express IL-2 at an early stage, but it has remained uncertain whether they are activated to exercise this competence in vivo. Therefore, we have used in situ hybridization and immunohistochemistry on serial sections obtained from fetal and adult thymuses of normal C57BL/6 mice and of mice bearing the scid defect to determine where, when, and whether IL-2 is expressed in vivo. Our results show a striking spatial and temporal pattern of IL-2 expression in the normal fetal thymus. We detected a burst of IL-2 mRNA accumulation at day 14.5 of gestation, which rapidly decreased by day 15. At day 15, we observed maximal IL-2 protein production that subsequently decreased by day 16 of gestation. Both in situ hybridization and immunohistochemical staining revealed an unexpectedly strict localization of IL-2 expressing cells to patches around the periphery of the fetal thymus, creating a previously unrecognized compartment of high IL-2 protein content. IL-2 production in the day-15 fetal thymus appeared to be unaffected by the scid mutation, indicating that this response is likely to be T-cell receptor (TcR)-independent. Several features distinguish the IL-2 induction pattern in the adult thymus from that in the fetal thymus. In the normal adult thymus, IL-2-expressing cells are extremely rare (found at a frequency of 10^(-7)), but they are reproducibly detectable as isolated cells in the outer cortex and subcapsular region of the thymus. Unlike the fetal thymic IL-2 producers, the IL-2 producers in the adult thymus are completely eliminated in mice homozygous for the scid mutation. This suggests that the IL-2-expressing cells in the normal adult thymus are of a more mature phenotype than the immature, TcR-negative cells that accumulate in the scid adult thymus. Thus, our work demonstrates that two developmentally distinct types of cell interactions induce IL-2 expression in vivo: one, a broadly localized interaction in day 14-15 fetal thymus that is unaffected by the scid mutation; the other, a rare event that occurs asynchronously from late fetal through adult life, but which is completely eliminated by the scid defect. These results imply that significant differences exist between the physiological processing of thymocytes in the fetal and postnatal thymic microenvironments

    Spontaneous Expression of Interleukin-2 In Vivo in Specific Tissues of Young Mice

    In situ hybridization and immunohistochemistry were used to determine the spectrum of tissues in which interleukin-2 (IL-2) mRNA and protein are found in healthy, normal young mice. In neonatal animals, IL-2 is expressed specifically by distinct, isolated cells at three major sites: the thymus, skin, and gut. Based on morphology and distribution, the IL-2-expressing cells resemble CD3ε + T cells that are also present in all these locations. Within the thymus of postweanling animals, both TcRαβ and TcRγδ lineage cells secrete "haloes" of the cytokine that diffuse over many cell diameters. Within the skin, isolated cells expressing IL-2 are seen at birth in the mesenchyme, and large numbers of IL-2-expressing cells are localized around hair follicles in the epidermis in 3-week-old animals. At this age, a substantial subset of CD3ε + cells is similarly localized in the skin. Significantly, by 5 weeks of age and later when the CD3ε + cells are evenly distributed throughout the epidermis, IL-2 RNA and protein expression are no longer detectable. Finally, within the intestine, IL-2 protein is first detected in association with a few discrete, isolated cells at day 16 of gestation and the number of IL-2 reactive cells increases in frequency through El9 and remains abundant in adult life. In postnatal animals, the frequency of IL- 2-positive cells in villi exceeds by greater than fivefold that found in mesenteric lymph node or Peyer's patches. Overall, these temporal and spatial patterns of expression provide insight into the regulation of IL-2 in vivo and suggest a role for IL-2 expression distinct from immunological responses to antigen

    Fabrication and Preliminary Characterization of Hydrophobic Silica Aerogel Films for Oil Remediation Studies

    Due to the potential of oil sorbents for oil spill clean-up, hydrophobic silica-based aerogel thin films have been synthesized and characterized by various methods. Aerogels were prepared using a two-step acid/base sol-gel process and functionalized with trimethylchlorosilane in hexane, followed by drying in ambient conditions. After film deposition by spin-coating, samples were characterized by scanning electron microscopy, nitrogen adsorption analysis (BET surface area = 377.66 m2 /g), and variable-angle spectroscopic ellipsometry (thickness of 287.00 ± 0.85 nm, refractive index of 1.08, porosity of 80.9%). Unlike previous studies with aerogel films, quartz crystal microbalance with dissipation monitoring (QCM-D) was applied to preliminarily measure the oil absorbing capacity of the fabricated aerogels. Upon exposure to hexadecane, aerogel films retained the model oil with an estimated Sauerbrey mass of ~970.6 ng/cm2 .     An Axiomatic Analysis of Diversity Evaluation Metrics: Introducing the Rank-Biased Utility Metric

    Many evaluation metrics have been defined to evaluate the effectiveness ad-hoc retrieval and search result diversification systems. However, it is often unclear which evaluation metric should be used to analyze the performance of retrieval systems given a specific task. Axiomatic analysis is an informative mechanism to understand the fundamentals of metrics and their suitability for particular scenarios. In this paper, we define a constraint-based axiomatic framework to study the suitability of existing metrics in search result diversification scenarios. The analysis informed the definition of Rank-Biased Utility (RBU) -- an adaptation of the well-known Rank-Biased Precision metric -- that takes into account redundancy and the user effort associated to the inspection of documents in the ranking. Our experiments over standard diversity evaluation campaigns show that the proposed metric captures quality criteria reflected by different metrics, being suitable in the absence of knowledge about particular features of the scenario under study.Comment: Original version: 10 pages.     Neuronal imaging with ultrahigh dynamic range multiphoton microscopy

    Multiphoton microscopes are hampered by limited dynamic range, preventing weak sample features from being detected in the presence of strong features, or preventing the capture of unpredictable bursts in sample strength. We present a digital electronic add-on technique that vastly improves the dynamic range of a multiphoton microscope while limiting potential photodamage. The add-on provides real-time negative feedback to regulate the laser power delivered to the sample, and a log representation of the sample strength to accommodate ultrahigh dynamic range without loss of information. No microscope hardware modifications are required, making the technique readily compatible with commercial instruments.     Value-Added Meat: Measuring Past Successes and Predicting Future Winners

    Livestock industries are significantly affected by changes in consumer behaviour. In order to add value to meat and livestock production, many firms and farms are supporting the development of new products – these products can differ by credence attribute, by degree of processing and by marketing strategies. The literature suggests that one of the most important determinants of success in product innovation is an understanding of the market the product is introduced into. In this report consumer preferences for meat products, by animal species including minor meats, and by type of processing are examined. Responses to economic variables such as price, advertising and income are identified as are responses to food safety and meat related health issues. Interesting results include the fact that income elasticities of demand for meat products purchased at grocery stores are negative in this study and that own and cross price elasticities for certain meats, across processing levels for example, show strong substitutions. Thus introducing new meat products may not result in increased sales by animal species but may only result in substitution of one meat type product for another product of the same meat type. Successful new product introductions or changes in product quality are shown, in this report, to be accompanied by significant marketing/advertising investment and, either by design or by serendipity, to have credence attributes in line with consumer's changing concerns. An example of this is the response in branded chicken sales, at the time of BSE in Canada, for a brand that could advertise itself as being 100% grain fed. Marketing strategies, such as working with the Health Check™ program of the Heart and Stroke Foundation of Canada are shown to have an impact on firm level sales (as well as on sales at the individual product level), a halo effect, that may be of importance in the firm's development of other value-added products. Significant heterogeneity exists in consumer behaviour and it is important to recognize this heterogeneity in the development of value-added meat products. Added to the consumer heterogeneity, in general, is the heterogeneity in responses by meat type.     Impulsivity, Rejection Sensitivity, and Reactions to Stressors in Borderline Personality Disorder

    This research investigated baseline impulsivity, rejection sensitivity, and reactions to stressors in individuals with borderline personality disorder compared to healthy individuals and those with avoidant personality disorder . The borderline group showed greater impulsivity than the avoidant and healthy groups both in a delay-discounting task with real monetary rewards and in self-reported reactions to stressors; moreover, these findings could not be explained by co-occurring substance use disorders. Distress reactions to stressors were equally elevated in both personality disorder groups (relative to the healthy group). The borderline and avoidant groups also reported more maladaptive reactions to a stressor of an interpersonal versus non-interpersonal nature, whereas the healthy group did not. Finally, self-reported impulsive reactions to stressors were associated with baseline impulsivity in the delay-discounting task, and greater self-reported reactivity to interpersonal than non-interpersonal stressors was associated with rejection sensitivity.     Competition and Burn Severity Determine Post-Fire Sapling Recovery in a Nationally Protected Boreal Forest of China: An Analysis from Very High-Resolution Satellite Imagery

    Anticipating how boreal forest landscapes will change in response to changing fire regime requires disentangling the effects of various spatial controls on the recovery process of tree saplings. Spatially explicit monitoring of post-fire vegetation recovery through moderate resolution Landsat imagery is a popular technique but is filled with ambiguous information due to mixed pixel effects. On the other hand, very-high resolution (VHR) satellite imagery accurately measures crown size of tree saplings but has gained little attention and its utility for estimating leaf area index (LAI, m2/m2) and tree sapling abundance (TSA, seedlings/ha) in post-fire landscape remains untested. We compared the explanatory power of 30 m Landsat satellite imagery with 0.5-m WorldView-2 VHR imagery for LAI and TSA based on field sampling data, and subsequently mapped the distribution of LAI and TSA based on the most predictive relationships. A random forest (RF) model was applied to assess the relative importance and causal mechanisms of spatial controls on tree sapling recovery. The results showed that pixel percentage of canopy trees (PPCT) derived from VHR imagery outperform all Landsat-derived spectral indices for explaining variance of LAI (R2VHR = 0.676 vs. R2Landsat = 0.427) and TSA (R2VHR = 0.508 vs. R2Landsat = 0.499). The RF model explained an average of 55.5% (SD = 3.0%, MSE = 0.382, N = 50) of the variation of estimated LAI. Understory vegetation coverage (competition) and post-fire surviving mature trees (seed sources) were the most important spatial controls for LAI recovery, followed by burn severity (legacy effect), topographic factors (environmental filter) and nearest distance to unburned area (edge effect). These analyses allow us to conclude that in our study area, mitigating wildfire severity and size may increase forest resilience to wildfire damage. Given the easily-damaged seed banks and relatively short seed dispersal distance of coniferous trees, reasonable human help to natural recovery of coniferous forests is necessary for severe burns with a large patch size, particularly in certain areas. Our research shows the VHR WorldView-2 imagery better resolves key characteristics of forest landscapes like LAI and TSA than Landsat imagery, providing a valuable tool for land managers and researchers alike
