    Shedding light on the base-pair opening dynamics of nucleic acids in living human cells

    生細胞中の核酸のダイナミクスに光を当てる --生細胞中における挙動は試験管中とは異なる--. 京都大学プレスリリース. 2022-11-30.Base-pair opening is a fundamental property of nucleic acids that plays important roles in biological functions. However, studying the base-pair opening dynamics inside living cells has remained challenging. Here, to determine the base-pair opening kinetics inside living human cells, the exchange rate constant (kex) of the imino proton with the proton of solvent water involved in hairpin and G-quadruplex (GQ) structures is determined by the in-cell NMR technique. It is deduced on determination of kex values that at least some G-C base pairs of the hairpin structure and all G-G base-pairs of the GQ structure open more frequently in living human cells than in vitro. It is suggested that interactions with endogenous proteins could be responsible for the increase in frequency of base-pair opening. Our studies demonstrate a difference in dynamics of nucleic acids between in-cell and in vitro conditions

    This study is to report a case of elementary school which applied the school organizational development theory(=SOD theory, Sako & Nakagawa 2005)to an approach to improving pupils\u27 achievement.Our SOD theory is based on two basic assumptions about a model of teaches\u27 spontaneous improvement activity cycle and about a model of process of collaboration among teachers. This SOD theory is composed of the following three components of organizational development of school.(1)rearrangement of organizational structure or control : installation of \u27core-system\u27 that is a field where stimulate teachers\u27 communication based on actual conditions of pupils and the fact of the teachersY\u27 teaching,(2)change of organizational process : improvement of quality and style of teachers\u27 communication,and(3)installation of teachers\u27 team that facilitate teachers\u27 collaboration process.The school organizational development methods based on the above components, was introduced into an elementary school with task of raising achievement of pupils.Main results of school organizational development to the school were as follows,(1)the information about actual conditions of less able pupil and teachers\u27 instructional activities was opened and shared among the teachers were facilitated,(2)sharing practical knowledges of instruction were facilitated,(3)teachers\u27 attitude and activities towards spontaneous improvement of instruction became obvious,and(4)pupils\u27 achievement was steadily improved

    Accepted 13 Feb 2015.Two second-generation quadruplex hammerhead ribozymes, whose activity enhances in response to K(+)via quadruplex formation of embedded r(GGA)3GG, were developed. Different strategies were applied to suppress basal activity when K(+) is absent. As a result, the activity enhancement upon the addition of K(+) has reached as high as 21-fold

    Microbial Control in the Primary Packaging of Pills Using Ionizing Radiation and Its Effect on Characteristic Constituents for Quality Control in Irradiated Pills

    Pharmaceutical products that mix natural raw materials are subject to unavoidable contamination with microorganisms from the environment and animals. This study focused on the effect of radiation on the quality of primary packaged pills, which are crude drug products. The pills, which were sealed in a sack for primary packaging laminated with polyethylene terephthalate, polyethylene, and aluminum foil, were irradiated by gamma rays or electron beam (EB). The survival counts of bacteria were reduced to 103 CFU/g or less by 6 kGy of irradiation. The counts of the spore-forming bacteria Bacillus megaterium, B. cereus, and Brevibacillus brevis in the pills were reduced to not over 100 CFU/g after 10 kGy irradiation. Although some of the cinnamaldehyde in the pills was oxidized to cinnamic acid, the decomposition of swertiamarin, berberine, glycyrrhizin, and cinnamaldehyde in the pills after 10 kGy irradiation were within the analytical accuracy by high-performance liquid chromatography. Gamma-ray or EB treatment at the final production of crude drug preparations was within the permissible standard value for the non-aqueous preparations for oral administration, with no statistically significant change in the indicator ingredients of crude drugs

    Combination Therapy with Empagliflozin and Insulin Results in Successful Glycemic Control: A Case Report of Uncontrolled Diabetes Caused by Autoimmune Pancreatitis and Subsequent Steroid Treatment

    A 66-year-old Japanese male presented with thirst, polyuria, and hemoglobin A1c and postprandial glucose levels (13.1% and 529 mg/dL, respectively) that indicated severe hyperglycemia. Based on his high immunoglobulin G4 level and the results of magnetic resonance imaging and magnetic resonance cholangiopancreatography, we diagnosed him with autoimmune pancreatitis. Insulin was initiated to control his diabetes. One month later, the patient commenced on prednisolone therapy for the treatment of autoimmune pancreatitis, after which his total insulin dosage increased to a maximum of 52 units/day. When the prednisolone dosage was later tapered, the patient’s total dosage of insulin was reduced to 42 units/day. However, he had gained 3.6 kg from the start of prednisolone therapy, and 42 units/day was insufficient for maintaining glycemic control. Thus, empagliflozin, a sodium-dependent glucose transporter 2 (SGLT2) inhibitor, was added. Thereafter, we were able to reduce the patient’s total dosage of insulin; it was eventually discontinued with good glycemic control and weight loss. Such results suggest that the combination of insulin with an SGLT2 inhibitor may be a viable option for the treatment of diabetic patients on prednisolone therapy
