Lentivirus-meditated frataxin gene delivery reverses genome instability in Friedreich ataxia patient and mouse model fibroblasts
Friedreich ataxia (FRDA) is a progressive neurodegenerative disease caused by deficiency of frataxin protein, with the primary sites of pathology being the large sensory neurons of the dorsal root ganglia and the cerebellum. FRDA is also often accompanied by severe cardiomyopathy and diabetes mellitus. Frataxin is important in mitochondrial iron–sulfur cluster (ISC) biogenesis and low-frataxin expression is due to a GAA repeat expansion in intron 1 of the FXN gene. FRDA cells are genomically unstable, with increased levels of reactive oxygen species and sensitivity to oxidative stress. Here we report the identification of elevated levels of DNA double strand breaks (DSBs) in FRDA patient and YG8sR FRDA mouse model fibroblasts compared to normal fibroblasts. Using lentivirus FXN gene delivery to FRDA patient and YG8sR cells, we obtained long-term overexpression of FXN mRNA and frataxin protein levels with reduced DSB levels towards normal. Furthermore, γ-irradiation of FRDA patient and YG8sR cells revealed impaired DSB repair that was recovered on FXN gene transfer. This suggests that frataxin may be involved in DSB repair, either directly by an unknown mechanism, or indirectly via ISC biogenesis for DNA repair enzymes, which may be essential for the prevention of neurodegeneration.Ataxia UK, FARA Australasia and FARA US
Gestion des déchets et des effluents radioactifs générés lors de la prise en charge d’un blessé radiocontaminé dans une structure hospitalière
La gestion des déchets et des effluents radioactifs générés à la suite de la prise en
charge d’un blessé radiocontaminé dans une chaîne de décontamination hospitalière doit
être anticipée pour respecter la règlementation qui s’y rattache. Cette gestion spécifique
sera d’autant plus complexe que le volume des déchets sera important. La première étape
consiste à contrôler la présence effective d’une contamination par mesure de l’activité
radiologique, afin de réaliser un tri en amont, dans le but de réduire le volume de
déchets radiocontaminés générés. La filière d’élimination est ensuite adaptée aux
caractéristiques physiques du radionucléide, en particulier sa période. Si cette période
est inférieure à 100 jours, la gestion se fait en décroissance pendant au moins 10
périodes avant élimination dans les filières applicables aux déchets hospitaliers. Pour
une période supérieure à 100 jours, des mesures spécifiques de pré-traitement et de mise
en fûts doivent être mises en œuvre avant colisage pour respecter les spécificités
requises par l’Agence Nationale pour la gestion des Déchets RadioActifs (ANDRA), seul
organisme identifié pour la reprise des déchets de ce type. Le traitement des effluents
liquides nécessite l’intervention d’une entreprise spécialisée. La gestion post
accidentelle des déchets hospitaliers radiocontaminés soulève également le problème de la
responsabilité financière et de la détention hospitalière de sources non médicales
Histamine food poisonings: A systematic review and meta-analysis
ABSTRACTObjective: The aim of this study was to assess the mean of histamine concentration in food poisoning.Design: Systematic review and meta-analysis of reports published between 1959 and 2013.Study selection: Main criteria for inclusion of studies were: all report types that present outbreaks of "histamine poisoning' or "scombroid syndrome" from food, including histamine content and type of food. Health status of people involved must be nonpathological.Results: Fifty-five (55) reports were included, these studies reported 103 incidents. All pooled analyses were based on random effect model; histamine mean concentration in poisoning samples was 1107.21 mg/kg with confidence interval for the meta-mean of 422.62–2900.78 mg/kg; heterogeneity index (I2) was 100% (P < 0.0001); prediction interval was 24.12–50822.78 mg/kg. Fish involved in histamine poisoning was mainly tuna or Istiophoridae species. No clues of association between concomitant conditions (female sex, alcohol consumption, previous medication,..
