Flame-Retardant and Thermal Degradation Mechanism of Caged Phosphate Charring Agent with Melamine Pyrophosphate for Polypropylene
An efficient caged phosphate charring agent named PEPA was synthesized and combined with melamine pyrophosphate (MPP) to flame-retard polypropylene (PP). The effects of MPP/PEPA on the flame retardancy and thermal degradation of PP were investigated by limiting oxygen index (LOI), vertical burning test (UL-94), cone calorimetric test (CCT), and thermogravimetric analysis (TGA). It was found that PEPA showed an outstanding synergistic effect with MPP in flame retardant PP. When the content of PEPA was 13.3 wt% and MPP was 6.7 wt%, the LOI value of the flame retardant PP was 33.0% and the UL-94 test was classed as a V-0 rating. Meanwhile, the peak heat release rate (PHRR), average heat release rate (AV-HRR), and average mass loss rate (AV-MLR) of the mixture were significantly reduced. The flame-retardant and thermal degradation mechanism of MPP/PEPA was investigated by TGA, Fourier transform infrared spectroscopy (FTIR), TG-FTIR, and scanning electron microscopy-energy dispersive X-ray spectrometry (SEM-EDXS). It revealed that MPP/PEPA could generate the triazine oligomer and phosphorus-containing compound radicals which changed the thermal degradation behavior of PP. Meanwhile, a compact and thermostable intumescent char was formed and covered on the matrix surface to prevent PP from degrading and burning
Predicting In Vivo Anti-Hepatofibrotic Drug Efficacy Based on In Vitro High-Content Analysis
Background/Aims
Many anti-fibrotic drugs with high in vitro efficacies fail to produce significant effects in vivo. The aim of this work is to use a statistical approach to design a numerical predictor that correlates better with in vivo outcomes.
Methods
High-content analysis (HCA) was performed with 49 drugs on hepatic stellate cells (HSCs) LX-2 stained with 10 fibrotic markers. ~0.3 billion feature values from all cells in >150,000 images were quantified to reflect the drug effects. A systematic literature search on the in vivo effects of all 49 drugs on hepatofibrotic rats yields 28 papers with histological scores. The in vivo and in vitro datasets were used to compute a single efficacy predictor (Epredict).
Results
We used in vivo data from one context (CCl4 rats with drug treatments) to optimize the computation of Epredict. This optimized relationship was independently validated using in vivo data from two different contexts (treatment of DMN rats and prevention of CCl4 induction). A linear in vitro-in vivo correlation was consistently observed in all the three contexts. We used Epredict values to cluster drugs according to efficacy; and found that high-efficacy drugs tended to target proliferation, apoptosis and contractility of HSCs.
Conclusions
The Epredict statistic, based on a prioritized combination of in vitro features, provides a better correlation between in vitro and in vivo drug response than any of the traditional in vitro markers considered.Institute of Bioengineering and Nanotechnology (Singapore)Singapore. Meta-analysis Followed by Replication Identifies Loci in or near CDKN1B, TET3, CD80, DRAM1, and ARID5B as Associated with Systemic Lupus Erythematosus in Asians
Systemic lupus erythematosus (SLE) is a prototype autoimmune disease with a strong genetic involvement and ethnic differences. Susceptibility genes identified so far only explain a small portion of the genetic heritability of SLE, suggesting that many more loci are yet to be uncovered for this disease. In this study, we performed a meta-analysis of genome-wide association studies on SLE in Chinese Han populations and followed up the findings by replication in four additional Asian cohorts with a total of 5,365 cases and 10,054 corresponding controls. We identified genetic variants in or near CDKN1B, TET3, CD80, DRAM1, and ARID5B as associated with the disease. These findings point to potential roles of cell-cycle regulation, autophagy, and DNA demethylation in SLE pathogenesis. For the region involving TET3 and that involving CDKN1B, multiple independent SNPs were identified, highlighting a phenomenon that might partially explain the missing heritability of complex diseases

Among the nine mammalian protein arginine methyltransferases (PRMTs), PRMT8 is unusual because it has restricted expression in the nervous system and is the only membrane-bound PRMT. Emerging studies have demonstrated that this enzyme plays multifaceted roles in diverse processes in neurons. Here we will summarize the unique structural features of PRMT8 and describe how it participates in various neuronal functions such as dendritic growth, synapse maturation, and synaptic plasticity. Recent evidence suggesting the potential role of PRMT8 function in neurological diseases will also be discussed

Synthesis of Zirconium-Containing Polyhedral Oligometallasilsesquioxane as an Efficient Thermal Stabilizer for Silicone Rubber
Free radicals play a negative role during the thermal degradation of silicone rubber (SR). Quenching free radicals is proposed to be an efficient way to improve the thermal-oxidative stability of SR. In this work, a novel zirconium-containing polyhedral oligometallasilsesquioxane (Zr-POSS) with free-radical quenching capability was synthesized and characterized. The incorporation of Zr-POSS effectively improved the thermal-oxidative stability of SR. The T5 (temperature at 5% weight loss) of SR/Zr-POSS significantly increased by 31.7 °C when compared to the unmodified SR. Notably, after aging 12 h at 280 °C, SR/Zr-POSS was still retaining about 65%, 60%, 75%, and 100% of the tensile strength, tear strength, elongation at break, and hardness before aging, respectively, while the mechanical properties of the unmodified SR were significantly decreased. The possible mechanism of Zr-POSS for improving the thermal-oxidative stability of SR was intensively studied and it was revealed that the POSS structure could act as a limiting point to suppress the random scission reaction of backbone. Furthermore, Zr could quench the free radicals by its empty orbital and transformation of valence states. Therefore, it effectively suppressed the thermal-oxidative degradation and crosslinking reaction of the side chains
Forest Resource Assets Departure Audit Considering Ecological Sustainable Development: A Case Study
The Forest Resources Assets Departure Audit (FRADA) is an important measure for the Chinese government to protect and develop forest resources, as well as to promote carbon peaking and carbon neutrality. Explicit audit content and scientific audit method are the basic premises for conducting an accurate audit. In this study, landscape ecology methods were employed to learn the spatiotemporal properties of forest resources, with the support of spatial information technology such as Geography Information System (GIS) and Remote Sensing (RS). A FRADA system from the perspective of landscape pattern index and ecological vulnerability was constructed. A case study was conducted for the evaluation of the proposed FRADA System. Lichuan, a county-level city in Hubei Province, China that is rich in forest resources was selected as the audit sample area, and the tenure period from 2015 to 2019 was regarded as the tenure cycle. The results showed that: (1) Before and after the tenure cycle, the number of patches (NP), patch density (PD) and largest patch index (LPI) of forest landscape showed a downward trend. Meanwhile, the decreased quantity and quality of forest landscapes showed inadequate protection of large forest patches with significant ecological value. In general, it was not beneficial for the sustainable development of forest resources in the view of ecology. (2) The landscape shape index (LSI) and connectivity index (CONNECT) showed an upward trend, which indicates that the shape of the forest landscape is changing in a complex way, and the connectivity has been improved. While, fractal dimension (FD) and fragmentation index (FN) showed a downward trend, indicating that the fragmentation trend of the forest landscape has been alleviated and the forest resources have a continuous and concentrated distribution. (3) The vulnerability of the forest landscape was significantly aggravated. Regarding administrative areas under the jurisdictions of Nanping Township, Jiannan Town and Fobaoshan Development Zone were particularly disturbed, and required special attention. The research results enrich and develop the theories and methods of FRADA and provide suggestions for the sustainable development of forest resources in Lichuan
Preparation and Characterization of UV-Curable Cyclohexanone-Formaldehyde Resin and Its Cured Film Properties
UV-curable cyclohexanone-formaldehyde (UVCF) resin was prepared with cyclohexanone-formaldehyde (CF) resin, isophorone diisocyanate (IPDI), and pentaerythritol triacrylate (PETA) as base substance, bridging agent, and functional monomer, respectively. The structure of UVCF was characterized by Fourier transform infrared spectroscopy (FT-IR), 1H-nuclear magnetic resonance spectroscopy (1H-NMR), and gel permeation chromatography (GPC). The viscosity and photopolymerization behavior of the UV-curable formulations were studied. The thermal stability and mechanical properties of the cured films were also investigated. The results showed that UVCF resin was successfully prepared, the number of average molecular weight was about 2010, and its molecular weight distribution index was 2.8. With the increase of UVCF resin content, the viscosity of the UV-curable formulations increased. After exposure to UV irradiation for 230 s, the photopolymerization conversion of the UV-curable formulations was above 80%. Moreover, when the UVCF content was 60%, the formulations had high photopolymerization rate, and the cured UVCF films showed good thermal stability and mechanical properties
