    Trifluoromethyl-phenyl-triazolyl derivative of beta-bisabolol induces cell death in ME-180 cervical cancer cells through induction of apoptosis and ROS generation

    The aim of the current investigation was to design, synthesize and demonstrate the anticancer and apoptotic activity of trifluoromethyl-phenyl-triazolyl derivative of beta-bisabolol (TTB) in ME-180 human cervical cancer cells.  MTT and clonogenic assays were used to evaluate the cell viability and colony formation tendencies of the cells respectively. Phase contrast and fluorescence microscopic investigations were used to evaluate the effect of TTB on cellular morphology and apoptosis. Flow cytometric analysis using fluorescent CM-DCFH2-DA were used to study the effect of TTB on reactive oxygen species (ROS) formation. The results revealed that TTB significantly inhibited the proliferation of ME-180 human cervical cancer cells in a time-dependent as well as dose-dependent manner. TTB has the capacity to inhibit both anchorage dependent as well as anchorage independent growth of ME-180 cervical cancer cells. TTB-treated cells revealed chromatin condensation, fragmented nuclei and nuclear shrinkage. A 3-fold increase of ROS production was seen after 72 ?M TTB treatment.     Phylogenetic distribution and predominant genotype of the avian infectious bronchitis virus in China during 2008-2009

    <p>Abstract</p> <p>Background</p> <p>The nephropathogenic avian infectious bronchitis (IB) caused unprecedented economic losses to the commercial chicken industry of China in 2008-2009. To investigate the prevalence of nephropathogenic IB in China, eighty IBV isolates from different provinces during 2008-2009 were identified by dwarf embryo test and RT-PCR.</p> <p>Results</p> <p>The strains were mostly isolated in winter and spring with a wide age range of IB outbreaks, from 4 to 69 days. By the virus recovery trials, 70/80 of the strains resulted in the deaths or distresses of birds from nephritis. To learn more about the molecular evolutionary characteristics of the circulating field strains, the coding region of major spike 1 (S1) protein gene of these strains was RT-PCR amplified and sequenced. Compared to the published representative strains, nucleotides and amino acids sequence analysis indicated that the S1 genes of these strains and the reference strains displayed homologies ranging from 75.1% to 99.8% and from 73.1% to 99.8% respectively. S1 protein of the major pandemic strains contained 540 or 542 amino acids with the cleavage site of HRRRR or RRFRR. Phylogenetic analysis revealed that recent field isolates of IBV in China were mostly belonged to A2-branch (QXIBV-branch) and HN08-branch, only one isolate was belonged to Gray-branch and M41-branch respectively. Most of the 80 strains showed evolutionarily distant from vaccine strains.</p> <p>Conclusions</p> <p>The results of this study suggested that nephropathogenic IBVs were mainly A2-like strains in China during 2008-2009.</p

    A new score system for predicting response to cardiac resynchronization therapy

    Background: The aim of this study was to establish a score system derived from clinical, echocardiographic and electrocardiographic indexes and evaluate its clinical value for cardiac resynchronization therapy (CRT) patient selection. Methods: Ninety-three patients receiving CRT were enrolled. A patient selection score system was generated by the clinical, echocardiographic and electrocardiographic parameters achieving a significant level by univariate and multivariate Cox regression model. The positive response to CRT was a left ventricular end systolic volume decrease of ≥ 15% and not reaching primary clinical endpoint (death or re-hospitalization for heart failure) at the end of follow-up. Results: Thirty-nine patients were CRT non-responders (41.94%) and 54 were responders (58.06%). A 4-point score system was generated based on tricuspid annular plane systolic ex­cursion (TAPSE), longitudinal strain (LS), and complete left bundle branch block (CLBBB) combined with a wide QRS duration (QRSd). The sensitivity and specificity for prediction of a positive response to CRT at a score &gt; 2 were 0.823 and 0.850, respectively (AUC: 0.92295% CI 0.691–0.916, p&lt; 0.001). Conclusions: A patient selection score system based on the integration of TAPSE, LS and CLBBB combined with a wide QRSd can help to predict positive response to CRT effectively and reliably

    Effects of Dioscorea polystachya \u27yam gruel\u27 on the cognitive function of diabetic rats with focal cerebral ischemia-reperfusion injury via the gut-brain axis

Focal cerebral ischemia-reperfusion injury is closely related to hyperglycemia and gut microbiota imbalance, while gut microbiota contributes to the regulation of brain function through the gut-brain axis. Previous studies in patients with diabetes have found that \u27yam gruel\u27 is a classic medicated diet made from Dioscorea polystachya, increases the content of Bifidobacterium, regulates oxidative stress, and reduces fasting blood glucose levels. The research reported here investigated the effects of \u27yam gruel\u27 on the cognitive function of streptozotocin-induced diabetic rats with focal cerebral ischemia-reperfusion injury and explored the mechanism underlying the role of the gut-brain axis in this process. \u27Yam gruel\u27 was shown to improve cognitive function as indicated by increased relative content of probiotic bacteria, and short-chain fatty acids in the intestinal tract and cerebral cortex reduced oxidative stress and inflammatory response and promotion of the expression of neurotransmitters and brain-derived neurotrophic factor. Thus, it is concluded that \u27yam gruel\u27 has a protective effect on cognitive function via a mechanism related to the gut-brain axis

    Proteomic analysis of sex conversion induced by CPPU in male grapevine of Vitis amurensis

    If N-(2-chloro-4-pyridyl)-N′-phenylurea (CPPU) could induce sex conversion in male plants of Vitis amurensis Rupr., this would reduce blindness of selection for male parents according to the fruit characters in cold-tolerant and disease-tolerant grape crossbreeding. Flower bud samples of male plants were treated with 100 mg∙L-1 CPPU at 15 days before anthesis. Two-dimensional gel electrophoresis (2-DE) was used to analyze the proteins related to sex conversion at different development time points. More than 600 protein spots were detected. Among them, 31 differentially expressed proteins were identified by MALDI-TOF/TOF, and 24 protein spots could be assigned to a probable function. Seventeen proteins participated in the sex conversion and with complex interaction. Sex conversion might receive the ROS signal in the beginning, and then pollen tube proteins were proposed to down-regulate to repress the stamen development, while the up-regulated cell elongation protein might promote the development of pistil. Adenine phosphoribosyl transferase 3 was proposed as the key protein in the sex organ conversion that was up-regulated by CPPU in the male V. amurensis achieving the ability to fruit in the end.

    Knowledge, attitude, and practice of medication and its influencing factors among residents in western China: a large-scale cross-sectional study

    BackgroundThis study aimed to understand the knowledge, attitude, and practice (KAP) of drug use among residents in western China and its influencing factors for accurately designing the knowledge, contents, and methods of popular science activities for safe drug use among residents to provide a reference for conducting rational drug use educational activities and improving residents’ level of safe drug use.MethodsA cross-sectional questionnaire survey was conducted to investigate the KAP of medication among western China residents and its influencing factors from March to April 2023. Each question option was assigned a score according to logic, and the risk factors for resident medication safety KAP were explored through univariate and logistic regression analyses.ResultsA total of 7,557 valid questionnaires were collected, with an effective recovery rate of 96.7%. The average scores of KAP were 72.77 ± 22.91, 32.89 ± 10.64, and 71.27 ± 19.09, respectively. In the evaluation criteria of the questionnaire, the score of medication knowledge reached “good,” and the score of attitude and practice was “average.” Multiple linear regression analysis indicated that male sex and low education level were significant factors affecting the lack of drug knowledge among residents. Old age and low education level were the factors of poor attitude toward medication. The low condition of medical security was a factor in residents’ irregular drug use behavior.ConclusionThe overall level of rational drug use among residents in western China is good, but there are still some inconsistencies. Rational drug use education should be conducted according to the risk points of residents in drug safety KAP to further improve the level of rational drug use of residents

    A possible 250-second X-ray quasi-periodicity in the fast blue optical transient AT2018cow

    The fast blue optical transients (FBOTs) are a new population of extragalactic transients of unclear physical origin. A variety of mechanisms have been proposed including failed supernova explosion, shock interaction with a dense medium, young magnetar, accretion onto a compact object, and stellar tidal disruption event, but none is conclusive. Here we report the discovery of a possible X-ray quasi-periodicity signal with a period of \sim250 second (at a significance level of 99.76%) in the brightest FBOT AT2018cow through the analysis of XMM-Newton/PN data. The signal is independently detected at the same frequency in the average power density spectrum from data taken from the Swift telescope, with observations covering from 6 to 37 days after the optical discovery, though the significance level is lower (94.26%). This suggests that the QPO frequency may be stable over at least 1.1×\times 104^{4} cycles. Assuming the \sim250 second QPO to be a scaled-down analogue of that typically seen in stellar mass black holes, a black hole mass of 103105\sim10^{3}-10^{5} solar masses could be inferred. The overall X-ray luminosity evolution could be modeled with the stellar tidal disruption by a black hole of 104\sim10^4 solar masses, providing a viable mechanism to produce AT2018cow. Our findings suggest that other bright FBOTs may also harbor intermediate-mass black holes.Comment: 18 pages, 10 figures. Accepted for publication in Research in Astronomy and Astrophysic

    The cross-reactivity of the enterovirus 71 to human brain tissue and identification of the cross-reactivity related fragments

    <p>Abstract</p> <p>Background</p> <p>EV71 occasionally cause a series of severe neurological symptoms, including aseptic meningitis, encephalitis, and poliomyelitis-like paralysis. However, the neurological destruction mechanism was remained to be clarified. This study described the cross reaction between EV71 induced IgG and human brain tissue.</p> <p>Results</p> <p>Cross reaction of the IgG from 30 EV71 infected patients' sera to human tissues of cerebra was observed, which suggested that some EV71 antigens could induce IgG cross-reactivity to human cerebra. To identify the regions of EV71 virus that containing above antigens, the polypeptide of virus was divided into 19 peptides by expression in prokaryotes cell. Mouse anti-sera of these peptides was prepared and applied in immunohistochemical staining with human adult and fetus brain tissue, respectively. The result indicated the 19 peptides can be classified into three groups: strong cross-reactivity, weak cross-reactivity and no cross-reactivity with human brain tissue according the cross reaction activity. Then, the increased Blood Brain Barrier (BBB) permeability and permits IgG entry in neonatal mice after EV71 infection was determined.</p> <p>Conclusion</p> <p>EV71 induced IgG could enter BBB and cross-reacted with brain tissue in EV71 infected neonatal mice, and then the peptides of EV71 that could induce cross-reactivity with brain tissue were identified, which should be avoided in future vaccine designing.</p

    Predicting progression of white matter hyperintensity using coronary artery calcium score based on coronary CT angiography—feasibility and accuracy

    ObjectiveCoronary artery disease (CAD) usually coexists with subclinical cerebrovascular diseases given the systematic nature of atherosclerosis. In this study, our objective was to predict the progression of white matter hyperintensity (WMH) and find its risk factors in CAD patients using the coronary artery calcium (CAC) score. We also investigated the relationship between the CAC score and the WMH volume in different brain regions.MethodsWe evaluated 137 CAD patients with WMH who underwent coronary computed tomography angiography (CCTA) and two magnetic resonance imaging (MRI) scans from March 2018 to February 2023. Patients were categorized into progressive (n = 66) and nonprogressive groups (n = 71) by the change in WMH volume from the first to the second MRI. We collected demographic, clinical, and imaging data for analysis. Independent risk factors for WMH progression were identified using logistic regression. Three models predicting WMH progression were developed and assessed. Finally, patients were divided into groups based on their total CAC score (0 to &lt;100, 100 to 400, and &gt; 400) to compare their WMH changes in nine brain regions.ResultsAlcohol abuse, maximum pericoronary fat attenuation index (pFAI), CT-fractional flow reserve (CT-FFR), and CAC risk grade independently predicted WMH progression (p &lt; 0.05). The logistic regression model with all four variables performed best (training: AUC = 0.878, 95% CI: 0.790, 0.938; validation: AUC = 0.845, 95% CI: 0.734, 0.953). An increased CAC risk grade came with significantly higher WMH volume in the total brain, corpus callosum, and frontal, parietal and occipital lobes (p &lt; 0.05).ConclusionThis study demonstrated the application of the CCTA-derived CAC score to predict WMH progression in elderly people (≥60 years) with CAD
