    Bioactivating a bone substitute accelerates graft incorporation in a murine model of vertical ridge augmentation

    OBJECTIVE: Compared to autologous bone grafts, allogeneic bone grafts integrate slowly, which can adversely affect clinical outcomes. Here, our goal was to understand the molecular mechanisms underlying graft incorporation, and then test clinically feasible methods to accelerate this process. METHODS: Wild-type and transgenic Wnt reporter mice were used in a vertical ridge augmentation procedure. The surgery consisted of tunneling procedure to elevate the maxillary edentulous ridge periosteum, followed by the insertion of bone graft. Micro-computed tomographic imaging, and molecular/cellular analyses were used to follow the bone graft over time. Sclerostin null mice, and mice carrying an activated form of β-catenin were evaluated to understand how elevated Wnt signaling impacted edentulous ridge height and based on these data, a biomimetic strategy was employed to combine bone graft particles with a formulation of recombinant WNT protein. Thereafter, the rate of graft incorporation was evaluated. RESULTS: Tunneling activated osteoprogenitor cell proliferation from the periosteum. If graft particles were present, then osteoprogenitor cells attached to the matrix and gave rise to new bone that augmented edentulous ridge height. Graft particles alone did not stimulate osteoprogenitor cell proliferation. Based on the thicker edentulous ridges in mice with amplified Wnt signaling, a strategy was undertaken to load bone graft particles with WNT; this combination was sufficient to accelerate the initial step of graft incorporation. SIGNIFICANCE: Local delivery of a WNT protein therapeutic has the potential to accelerate graft incorporation, and thus shorten the time to when the graft can support a dental implant

    A Keypoint Based Enhancement Method for Audio Driven Free View Talking Head Synthesis

    Audio driven talking head synthesis is a challenging task that attracts increasing attention in recent years. Although existing methods based on 2D landmarks or 3D face models can synthesize accurate lip synchronization and rhythmic head pose for arbitrary identity, they still have limitations, such as the cut feeling in the mouth mapping and the lack of skin highlights. The morphed region is blurry compared to the surrounding face. A Keypoint Based Enhancement (KPBE) method is proposed for audio driven free view talking head synthesis to improve the naturalness of the generated video. Firstly, existing methods were used as the backend to synthesize intermediate results. Then we used keypoint decomposition to extract video synthesis controlling parameters from the backend output and the source image. After that, the controlling parameters were composited to the source keypoints and the driving keypoints. A motion field based method was used to generate the final image from the keypoint representation. With keypoint representation, we overcame the cut feeling in the mouth mapping and the lack of skin highlights. Experiments show that our proposed enhancement method improved the quality of talking-head videos in terms of mean opinion score

    Novel NR5A1 Missense Mutation in Premature Ovarian Failure: Detection in Han Chinese Indicates Causation in Different Ethnic Groups

    Background The etiology of most premature ovarian failure (POF) cases is usually elusive. Although genetic causes clearly exist and a likely susceptible region of 8q22.3 has been discovered, no predominant explanation exists for POF. More recently, evidences have indicated that mutations in NR5A1 gene could be causative for POF. We therefore screened for mutations in the NR5A1 gene in a large cohort of Chinese women with non-syndromic POF. Methods Mutation screening of NR5A1 gene was performed in 400 Han Chinese women with well-defined 46,XX idiopathic non-syndromic POF and 400 controls. Subsequently, functional characterization of the novel mutation identified was evaluated in vitro. Results A novel heterozygous missense mutation [c.13T\u3eG (p.Tyr5Asp)] in NR5A1 was identified in 1 of 384 patients (0.26%). This mutation impaired transcriptional activation on Amh, Inhibin-a, Cyp11a1and Cyp19a1 gene, as shown by transactivation assays. However, no dominant negative effect was observed, nor was there impact on protein expression and nuclear localization. Conclusions This novel mutation p.Tyr5Asp, in a novel non-domain region, is presumed to result in haploinsufficiency.     Electron transport through dipyrimidinyl-diphenyl diblock molecular wire: protonation effect

    Recently, rectifying direction inversion has been observed in dipyrimidinyl-diphenyl (PMPH) diblock molecular wire [J. Am. Chem. Soc. (2005) 127, 10456], and a protonation mechanism was suggested to explain this interesting phenomena. In this paper, we study the protonation effect on transport properties of PMPH molecule by first principles calculations. No significant rectification is found for the pristine diblock molecular wire. Protonation leads to conductance enhancement and rectification. However, for all considered junctions with rectifying effect, the preferential current directions are samely from dipyrimidinyl side to diphenyl side. Effect of molecule-electrode anchoring geometry is studied, and it is not responsible for the discrepancy between experiment and theory.Comment: 17 pages, 8 figure

    Predicting depressive symptom by cardiometabolic indicators in mid-aged and older adults in China: a population-based cross-sectional study

    ObjectiveDepressive symptom is a serious mental illness often accompanied by physical and emotional problems. The prevalence of depressive symptom in older adults has become an increasingly important public health priority. Our study used cardiometabolic indicators to predict depressive symptom in middle-aged and older adults in China.MethodsThe data came from the China Health and Retirement Longitudinal Study 2011 (CHARLS2011), which was a cross-sectional study. The analytic sample included 8,942 participants aged 45 years or above. The study evaluated the relationship between cardiometabolic indicators and depression by measuring 13 indicators, including body mass index (BMI), waist circumference, waist-height ratio (WHtR), conicity index, visceral adiposity index (VAI), Chinese visceral adiposity index (CVAI), lipid accumulation product (LAP), a body shape index (ABSI), body roundness index (BRI), triglyceride glucose index (TyG-index) and its correlation index (TyG-BMI, TyG-waist circumference, TyG-WHtR). Binary logistic regression analysis was used to examine the association between thirteen cardiometabolic indicators and depressive symptom. In addition, the receiver operating characteristic (ROC) curve analysis and area under curve (AUC) were used to evaluate the predictive anthropometric index and to determine the optimum cut-off value.ResultsThe study included 8,942 participants, of whom 4,146 (46.37%) and 4,796 (53.63%) were male and female. The prevalence of depressive symptom in mid-aged and older adults in China was 41.12% in males and 55.05% in females. The results revealed that BMI [AUC = 0.440, 95%CI: 0.422–0.457], waist circumference [AUC = 0.443, 95%CI: 0.425–0.460], WHtR [AUC = 0.459, 95%CI: 0.441–0.476], LAP [AUC = 0.455, 95%CI: 0.437–0.472], BRI [AUC = 0.459, 95%CI: 0.441–0.476], CVAI [AUC = 0.449, 95%CI: 0.432–0.467], TyG-BMI [AUC = 0.447, 95%CI: 0.429–0.465], and TyG-waist circumference [AUC =0.452, 95%CI: 0.434–0.470] were weak predictors of depressive symptom (p < 0.05) in males. In females, BMI [AUC = 0.470, 95%CI: 0.453–0.486], LAP [AUC = 0.484, 95%CI: 0.467–0.500], TyG-BMI [AUC = 0.470, 95%CI: 0.454–0.487], and TyG-waist circumference [AUC =0.481, 95%CI: 0.465–0.498] were weak predictors of depressive symptom (p < 0.05). On the other side, VAI, ABSI, conicity index and TyG index could not predict depressive symptom in middle-aged and older adults.ConclusionMost cardiometabolic indicators have important value in predicting depressive symptom. Our results can provide measures for the early identification of depressive symptom in middle-aged and older adults in China to reduce the prevalence of depressive symptom and improve health

    Transient evolution of non-metallic inclusions in molten high aluminum and high manganese steel contacting with slag and crucible: experimental investigation and FactSage macros modeling

    In this work, high temperature experiments of high Al molten steel contacting with crucible and slag were performed to investigate the effect of crucible–slag–steel reaction on transient evolution of inclusions in the high Al molten steel. Processing samples were taken for the chemical compositions of steel and also the inclusions have been characterized by using SEM and EDS AZtec. FactSage macros have been applied to simulate the evolution of molten steel composition and inclusions in molten steel during steel-slag reaction and comparing to the experimental results. Due to the steel–crucible–slag reactions especially under the condition of the high Al molten steel, the Ca and Mg transferred from slag and crucible to the molten steel and then subsequently led to the evolution of Mg and Ca-containing inclusions. The Mg inclusions transferred from MgAl2O4 to MgO inclusions while Ca transferred to steel and modified MgAl2O4 at crucible interface to form CaMgAl2O5 phase. The FactSage macros modeling did agree reasonably well with the experimental results

    ViroProfiler: a containerized bioinformatics pipeline for viral metagenomic data analysis

    ABSTRACTBacteriophages play central roles in the maintenance and function of most ecosystems by regulating bacterial communities. Yet, our understanding of their diversity remains limited due to the lack of robust bioinformatics standards. Here we present ViroProfiler, an in-silico workflow for analyzing shotgun viral metagenomic data. ViroProfiler can be executed on a local Linux computer or cloud computing environments. It uses the containerization technique to ensure computational reproducibility and facilitate collaborative research. ViroProfiler is freely available at https://github.com/deng-lab/viroprofiler

    Pole-to-Ground Fault Analysis and Fast Protection Scheme for HVDC Based on Overhead Transmission Lines

    Flexible direct current (DC) transmission network technology is an effective method for large capacity clean energy access to power grids, but the DC short-circuit fault detection for it is a difficult problem. In this paper, the pole-to-ground fault transient characteristics in a multi-terminal DC power grid, based on overhead transmission lines and DC circuit breakers, are analyzed firstly. Then, a fast protection scheme is proposed according to the fault transient characteristics. Only local information is utilized for fault detection and location in the proposed scheme. Moreover, the scheme is verified to have the advantages of fast action speed, high reliability and the ability to resist the transition resistance. A four terminal DC power grid model based on actual engineering parameters is established in PSCAD/EMTDC, and the validity of the protection scheme under different fault conditions is verified by simulation results
