    Study on Safety Control of Composite Roof in Deep Roadway based on Energy Balance Theory

    Improving the safety and stability of composite roof in deep roadway is the strong guarantee for safe mining and sustainable development of coal mines. With three roadways of different composite roofs in Hulusu Coal Mine and Menkeqing Coal Mine as the research background, this paper explores the mechanical properties and energy dissipation law of coal-rock structures with different height ratios from the perspective of energy release and dissipation through lab experiments. The results indicate that the key to the stability of coal-rock structures lies in maintaining relatively low dissipation energy. Based on experimental results and the energy balance theory, two support principles were put forward and applied to experimental roadways. The field monitoring results show that the anchoring force on different composite roof displays different characteristics, proving that the work done by the support can adjust timely to the energy release and conversion so as to improve the safety and stability of roadways with different composite roofs. This study provides a reference for the deformation control in deep roadways with composite roofs under similar conditions

    In vitro cultivation of rat bone marrow mesenchymal stem cells and establishment of pEGFP/Ang-1 transfection method

    ABSTRACTObjectiveTo obtain the bone marrow mesenchymal stem cells (BMSCs), complete phenotypic identification and successfully transfect rat BMSCs by recombinant plasmid pEGFP/Ang-1.MethodsBMSCs were isolated from bone marrow using density gradient centrifugation method and adherence screening method, and purified. Then the recombinant plasmid pEGFP/Ang-1 was used to transfect BMSCs and the positive clones were obtained by the screen of G418 and observed under light microscopy inversely. Green fluorescent exhibited by protein was enhanced to measure the change time of the expression amount of Ang-1.ResultsBMSCs cell lines were obtained successfully by adherence screening method and density gradient centrifugation. Ang-1 recombinant plasmid was transfected smoothly into rat BMSCs, which can express Ang-1 for 3 d and decreased after 7 d.ConclusionsAdherence screening method and density gradient centrifugation can be effective methods to obtain BMSCs with high purity and rapid proliferation. Besides, the expression of transfected recombinant plasmid pEGFP/Ang-1 in rat BMSCs is satisfactory

    改良穴位图谱在中医护理操作中运用效果评价

    Objective To find an effective study method to master the nursing procedures of acupoints that will be applied in the clinical nursing work. Methods Based on the Chinese medicine hospital of traditional Chinese medicine nursing guidelines (trial version) published by the bureau of traditional Chinese medicine and given full expression to the advantages of the Traditional Chinese Medicine, acupoints map was improved and was applied in clinical work in August 2011 in our hospital. The nursing operation workload and learning effect in the continuity two years were compared by retrospective investigation combined with questionnaire survey. Results 1, The operation workload increases obviously and the overall growth rate in five work targets is 88.66%, after improving acupoints map of TCM nursing. 2, There are significant differences (P < 0.001) on learning interest, learning degree, memory speed, knowledge of diseas, method of operation and clinical application in our comparison.Conclusion The improved acupoints map can inspire the learning interest of the nursing stuff, help them to master common nursing acupoints of traditional Chinese medicine quickly and apply it to clinical disease. There are good social and economic benefits of this method, so it is worth promoting.目的 寻找一种有效掌握中医护理穴位的学习方法并运用于临床护理工作中。 方法 以中医药局印发《中医医院中医护理工作指南(试行)》文件的内容为指导思想,全面体现中医特色和优势。本院积极开展中医护理操作,于2011年8月设计改良穴位图谱并运用于临床。采用回顾性调查并结合问卷式调查,比较实施前后一年期间中医护理操作工作量及学习效果。结果 (1)2011年8月采用改良穴位图谱后中医护理操作工作量明显增长,五项工作量的总体增长率达88.66%;(2)采用改良穴位图谱前后护士在学习兴趣、易学程度、记忆速度、疾病腧穴知识、操作方法、临床应用等方面比较均有显著性差异(P<0.001)。 结论 采取改良穴位图谱学习方法能提高护理人员对穴位的学习兴趣,快速掌握中医护理疾病的常用腧穴并应用于临床,赢得了较好的社会效益和经济效益,值得推广

    Deciphering of interactions between platinated DNA and HMGB1 by hydrogen/deuterium exchange mass spectrometry

    A high mobility group box 1 (HMGB1) protein has been reported to recognize both 1,2-intrastrand crosslinked DNA by cisplatin (1,2-cis-Pt-DNA) and monofunctional platinated DNA using trans-[PtCl2(NH3)(thiazole)] (1-trans-PtTz-DNA). However, the molecular basis of recognition between the trans-PtTz-DNA and HMGB1 remains unclear. In the present work, we described a hydrogen/deuterium exchange mass spectrometry (HDX-MS) method in combination with docking simulation to decipher the interactions of platinated DNA with domain A of HMGB1. The global deuterium uptake results indicated that 1-trans-PtTz-DNA bound to HMGB1a slightly tighter than the 1,2-cis-Pt-DNA. The local deuterium uptake at the peptide level revealed that the helices I and II, and loop 1 of HMGB1a were involved in the interactions with both platinated DNA adducts. However, docking simulation disclosed different H-bonding networks and distinct DNA-backbone orientations in the two Pt-DNA-HMGB1a complexes. Moreover, the Phe37 residue of HMGB1a was shown to play a key role in the recognition between HMGB1a and the platinated DNAs. In the cis-Pt-DNA-HMGB1a complex, the phenyl ring of Phe37 intercalates into a hydrophobic notch created by the two platinated guanines, while in the trans-PtTz-DNA-HMGB1a complex the phenyl ring appears to intercalate into a hydrophobic crevice formed by the platinated guanine and the opposite adenine in the complementary strand, forming a penta-layer π–π stacking associated with the adjacent thymine and the thiazole ligand. This work demonstrates that HDX-MS associated with docking simulation is a powerful tool to elucidate the interactions between platinated DNAs and proteins

    PREPARATION OF MACROPOROUS TIO2 BY STARCH MICROSPHERES TEMPLATE WITH ASSISTANCE OF SUPERCRITICAL CO2

    In this work a green route is reported to prepare a TiO2 macroporous network using corn starch microspheres flake as a bio-template. The starch microspheres prepared by emulsion technology were used as a template into which precursor tetrabutyl titanate (TBOT) was permeated using supercritical carbon dioxide (scCO2) as a forceful carrier or infiltration media, resulting in the formation of an organic/inorganic hybrid material; then the coated template was gelled and dried during the scCO2-coating and the depressurization processes, followed by removal of the template by calcination at 700°C; finally, TiO2 inverse-opals-like material reversely replicating the starch microspheres template was obtained. Scanning electron microscopy (SEM), nitrogen sorption measurements, and X-ray diffraction (XRD) indicated that the products were the inverse replicas from their templates. The obtained TiO2 inverse opals-like material showed a wide dispersion of pore sizes from mesopores to macropores – a few nanometers to several micrometers –with the BET surface area up to 103 m2/g, and a predominantly anatase crystalline phase. In addition, the wall thickness of the macropores varied with tunable pressure for closed cells or open-cell foams. So this facile and environmentally friendly process for the preparation of high-surface area, thermally-stable, metal-oxide catalysts and supports by a starch microsphere templating approach may have widespread potential applications in catalysis, absorbents, photoelectric materials, and so on

    A preliminary study on the immune responses of HPV16-E7 by combined intranasal immunization with lymphotoxin

    Objectives: Human papillomavirus (HPV) ranks the first cause of cervical cancer. Cervical cancer has high prevalence ratesin women around the world. The HPV-E7 oncoprotein is expressed in cervical cancer and is a target of developing immunotherapiesagainst HPV-associated tumors. However, the antigenicity of this protein is low. Due to this reason, potentadjuvants are required to enhance its therapeutic efficacy. This preliminary study aims to evaluate whether lymphotoxin(LT) could act as an effective immune adjuvant for HPV infection in mice models.Material and methods: Intranasal immunization was used to explore the effect of HPV-E7 and/or LT immune response.After the third intranasal immunization, the titer for the HPV-E7 antibody was detected in serum and vaginal washing fluid.Also, we assessed the expression of chemokine ligand 13 (CXCL13) and Peripheral Node Addressin (PNAd) in the lymphnodes after intranasal immunization with immunohistochemical analysis.Results: compared to HPV-E7 immunization, intranasal immunization with HPV-E7 plus LT significantly increased HPV-E7-specificserum IgG and vaginal IgA titers. Furthermore, the combined use of HPV-E7 and LT strongly induced E7-specific CTLresponses.Conclusions: LT can be effective for intranasal immunized HPV-E7 to improve E7-specific immune responses to HPV infection.It is new approach to eradicate chronic HPV infection capable of inducing an effective anti-infection method
