    Analysis of monitoring results of foodborne pathogens in infant food on the market of Guangxi Zhuang Autonomous Region in 2011-2016

    Objective To understand the contamination status and trend of foodborne pathogens in infant food in Guangxi Zhuang Autonomous Region, and provide reference to prevent and control the foodborne disease effectively. Methods Collecting infant food from 14 cities of Guangxi Zhuang Autonomous Region in 2011-2016. Four kinds of pathogenic bacteria were tested according to the food safety standard microbiological test (GB 4789) and the national food pollution and hazardous factors risk monitoring manual (2014). Results A total of 7 956 samples were detected. The positive rate of supplementary cereal food for infants and young children was 6.1% (249/4 084), and that of infant formula was 4.7% (183/3 872). The positive rates of Enterobacter sakazakii, Bacillus cereus (not in 2011 and 2015) and Staphylococcus aureus were 2.7%-5.6%, 2.3%-8.5% and 0.0%-0.4% respectively in supplementary cereal food for infants and young children. In the infant formula, the positive rate of Enterobacter sakazakii was from 0.2% to 1.8%. Bacillus cereus was 10.2%-12.4% (not in 2011 and 2015), and Staphylococcus aureus was 0.0%-0.3%. Salmonella was detected in one sample, and the origin was Inner Mongolia Autonomous Region. The positive rate of supplementary cereal food for infants and young children was 6.1% (248/4 077) in domestic products, while there was 1 positive in 7 foreign samples. In formula food, the positive rate was 4.7% (177/3 758) in domestic products and 5.3% (6/114) in imported products. Conclusion The contamination of Enterobacter sakazakii and Bacillus cereus in infant food sold in Guangxi Zhuang Autonomous Region posed a threat to infants. Therefore, it was necessary to strengthen the health supervision of domestic production enterprises

    Effects of Poncirin, a Citrus Flavonoid and Its Aglycone, Isosakuranetin, on the Gut Microbial Diversity and Metabolomics in Mice

    Poncirin (PC) and its aglycone, isosakuranetin (IR), occur naturally in citrus fruits. This study aimed to explore the pathways behind the different health benefits of PC and IR by evaluating the effect of these two bioactive flavonoids on the gut microbial diversity and metabolomics of mice. The 16S rRNA gene sequencing was used to analyze the alteration of gut microbiota in mice after PC and IR intervention. The metabolic impact of PC and IR in mice were studied using a metabolomics approach based on LC-MS analysis. Results showed that, after 7 days intervention, PC and IR multiplied the abundance of Parabacteroides in mice’s intestinal tracts by 1.2 and 1.0 times, respectively. PC increased the abundance of Bacteroides by 2.4 times. IR reduced the Allobaculum abundance by 1.0 time and increased Alloprevotella abundance by 1.5 times. When mice were given PC, their fecal acetic acid level increased by 1.8 times, while their isobutyric and isovaleric acid content increased by 1.2 and 1.3 times, respectively. Supplementation with IR had no significant effect on the content of short-chain fatty acids (SCFAs) in the feces of mice. The potential urine biomarkers of mice in the PC group were involved in the digestion and absorption of protein and carbohydrate, as well as the metabolism of amino acids, such as glycine, serine, threonine, tryptophan, D-arginine, D-ornithine, etc. IR mainly affected the amino acid metabolic pathways in mice, including taurine and hypotaurine metabolism, glutathione metabolism, histidine metabolism, D-glutamate metabolism, etc. This study provided valuable clues for future research on the health promoting mechanisms of PC and IR

    Derlin1 functions as an oncogene in cervical cancer via AKT/mTOR signaling pathway

    Abstract Background Cervical cancer (CC) ranks third in the morbidity and mortality of female cancer around the world. Derlin1 has been found to be overexpressed in several human cancers. However, it is still unclear about its roles in CC. The research aims to explore the relationship between Derlin1 and CC. Methods We purchased a human CC tissues microarray, which contained CC tissues and corresponding para-cancerous tissues from 93 patients with primary cervical squamous cell carcinoma. Immunohistochemical staining was used to confirm the expression of Derlin1 in these tissues. And we detected the differential expression of Derlin1 in cervical cancer cell lines and normal cervical epithelial cells (H8). Further, the cervical cancer cell lines SiHa and C33A were used as an in vitro model, which was down-regulated the expression of Derlin1 using siRNA interference technology. The effects of Derlin1 down-regulating in CC cell lines on cell proliferation and migration were detected by CCK8 assay and transwell assay, respectively. The effect of Derlin1 down-regulating on apoptosis was analyzed by flow cytometry, and apoptosis-related proteins were detected using western blotting. In-depth mechanisms were studied using western blotting. In addition, the effects of Derlin1 up-regulating in normal cervical epithelial cells also were exposed. Results Derlin1 was significantly elevated in CC tissues (81.7%, 76/93), and the expression of Derlin1 was positively correlated with the tumor size, pathological grade, and lymph node metastasis in CC patients. And Derlin1 was high expressed in cervical cancer cell lines compared to H8 cells. Knockdown of Derlin1 in cervical cancer cell lines inhibited cell proliferation and migration. Moreover, knockdown of Derlin1 induced apoptosis and affected the expression of apoptosis-related proteins, including Bcl-2, Bax, Bim, caspase3 and caspase9. Further experiments showed that AKT/mTOR signal pathway might be involve in this processes that knockdown of Derlin1 inhibited the expression of p-AKT and p-mTOR. Over-expression of Derlin1 in H8 cells promoted cell proliferation and migration via up-regulated the expression of p-AKT and p-mTOR. Conclusion Derlin1 is an oncogene in CC via AKT/mTOR pathway. It might be a potential therapeutic target for CC

    Clinical Value of Serum HMGB1 Levels in Early Detection of Recurrent Squamous Cell Carcinoma of Uterine Cervix: Comparison with Serum SCCA, CYFRA21-1, and CEA Levels

    Aim To evaluate the clinical value of serum high mobility group box chromosomal protein 1 (HMGB1) levels in making the early diagnosis of recurrent cervical squamous cell carcinomas (CSCC) and compare it with the value of serum squamous cell carcinoma antigen (SCCA), cytokeratin fragment (CYFRA) 21-1, and carcinoembryonic antigen (CEA) levels. Methods Immunohistochemical staining of tissue from 64 patients with recurrent CSCCs, 72 patients with non-recurrent carcinoma, and 28 healthy participants was performed to determine the expression of HMGB1 protein. The serum levels of the 4 markers in 112 patients with recurrent CSCC, 174 patients with non-recurrent disease, and 128 healthy participants were measured by enzyme-linked immunosorbent assay. The receiver operating characteristic (ROC) curves were constructed and the area under the curve (AUC) was calculated. Results Higher immunostaining score was found in recurrent CSCC tissue sections than in non-recurrent CSCC sections. Serum HMGB1 levels in patients with recurrent CSCC were significantly higher than in patients with non-recurrent disease and healthy controls. The AUC of HMGB1, SCCA, CYFRA21- 1, and CEA was 0.816, 0.768, 0.703, and 0.625, respectively. HMGB1 had the best specificity and positive likelihood ratio (78.0% and 3.25, respectively), whereas SCCA had the best sensitivity and negative likelihood ratio (76.3% and 0.34, respectively). Parallel combined measurements increased the diagnostic sensitivity and serial combination increased the specificity. High serum HMGB1 levels were inversely correlated with disease-free survival (P = 0.009, Pearson χ2 test) and overall survival (P = 0.018). Conclusion HMGB1 was overexpressed in recurrent CSCCs. Serum HMGB1 level could be a useful and specific marker for evaluating the disease recurrence and predicting prognosis in patients with CSCC. Serial combined measurements of serum HMGB1, SCCA, and CYFRA21-1 increased the diagnostic specificity, and parallel combined testing increased the diagnostic sensitivity

    Simplified residence time prediction models for constructed wetland water recycling systems

    The experimental farmland–channel–wetland systems (FCWS) in Guilin, China have been recently designed based on wetland water recycling systems in Midwest USA. The present article develops a methodology for simplifying the prediction of residence time as a function of the flow rate and physical shape of these contaminant removal systems. A series of two-dimensional simulation studies on surface flow through FCWS wetland of different shapes are performed. Parameters influencing hydraulic characteristics such as empirical values of inlet and outlet flow conditions, and wetland shapes are utilized as inputs to the study. Roughness coefficient was assumed to be constant across the different wetland designs discussed in this article. The mean velocity values within the wetland decreases with increase in ratio of variant inlet widths and wetland inflow rates. The results from the simulation are used as inputs for performing a multivariate multiparameter regression algorithm. This framework models the residence time within the wetland independently as a function of shape, mass inflow, and inlet geometry. This simplified model can be used with ease to evaluate existing as well as new wetland system designs for potential improvement in its function of desalting and filtering waters.Published versio

    Genome-wide characterization and expression profiling of the MAPKKK genes in Gossypium arboreum L.

    Mitogen-activated protein kinase kinase kinases (MAPKKKs) are important components of MAPK cascades, which have different functions during developmental processes and stress responses. To date, there has been no systematic investigation of this gene family in the diploid cotton Gossypium arboreum L. In this study, a genome-wide survey was performed which identified 78 MAPKKK genes in G. arboreum. Phylogenetic analysis classified these genes into three subgroups: 14 belonged to ZIK, 20 to MEKK, and 44 to Raf. Chromosome location, phylogeny, and the conserved protein motifs of the MAPKKK gene family in G. arboreum were analyzed. The MAPKKK genes had a scattered genomic distribution across 13 chromosomes. The members in the same subfamily shared similar conserved motifs. The MAPKKK expression patterns were analyzed in mature leaves, stems, roots, and at different ovule developmental stages, as well as under salt, and drought stresses. Transcriptome analysis showed that 76 MAPKKK genes had different transcript accumulation patterns in the tested tissues and 38 MAPKKK genes were differentially expressed in response to salt and drought stresses. These results lay the foundation for understanding the complex mechanisms behind MAPKKK-mediated developmental processes and abiotic stress-signaling transduction pathways in cotton.The accepted manuscript in pdf format is listed with the files at the bottom of this page.     Grapefruit has attracted much attention as a functional fruit, of which “Cocktail” is a special variety with low acidity. The present study aimed to investigate the effects of alcoholic fermentation on the non-volatile and volatile compounds of “Cocktail” grapefruit juice. To analyze, a non-targeted metabolomics method based on UPLC-MS/MS and volatiles analysis using GC-IMS were performed. A total of 1015 phytochemicals were identified, including 296 flavonoids and 145 phenolic acids, with noticeably increasing varieties and abundance following the fermentation. Also 57 volatile compounds were detected, and alcoholic fermentation was effective in modulating aromatic profiles of grapefruit juice, with terpenes and ketones decreasing, and alcohols increasing together with esters. Citraconic acid and ethyl butanoate were the most variable non-volatile and volatile substances, respectively. The results provide a wealth of information for the study of “Cocktail” grapefruit and will serve as a valuable reference for the large-scale production of grapefruit fermented juice in the future.</p
