Pharmacodynamic Evaluation and PK/PD-Based Dose Prediction of Tulathromycin: A Potential New Indication for Streptococcus suis Infection
Tulathromycin is the first member of the triamilide antimicrobial drugs that has been registered in more than 30 countries. The goal of this study is to provide a potential new indication of tulathromycin for Streptococcus suis infections. We investigated the pharmacokinetic and ex vivo pharmacodynamics of tulathromycin against experimental S. suis infection in piglets. Tulathromycin demonstrated a relatively long elimination half-life (74.1 h) and a mean residence time of 97.6 h after a single intramuscular administration. The minimal inhibitory concentration (MIC) and bactericidal concentration in serum were markedly lower than those in broth culture, with Mueller–Hinton broth/serum ratios of 40.3 and 11.4, respectively. The post-antibiotic effects were at 1.27 h (1× MIC) and 2.03 h (4× MIC) and the post-antibiotic sub-MIC effect values ranged from 2.47 to 3.10 h. The ratio of the area under the concentration–time curve divided by the MIC (AUC/MIC) correlated well with the ex vivo antimicrobial effectiveness of tulathromycin (R2 = 0.9711). The calculated AUC12h/MIC ratios in serum required to produce the net bacterial stasis, 1-log10 and 2-log10 killing activities were 9.62, 18.9, and 32.7, respectively. Based on the results of Monte Carlo simulation, a dosage regimen of 3.56 mg/kg tulathromycin was estimated to be effective, achieving for a bacteriostatic activity against S. suis infection over 5 days period. Tulathromycin may become a potential option for the treatment of S. suis infections
Multiple imaging modality-guided radiofrequency ablation combined with transarterial chemoembolization for hepatocellular carcinoma in special locations
PURPOSEWe aimed to evaluate the safety and effectiveness of radiofrequency ablation (RFA) combined with transarterial chemoembolization (TACE) guided by multiple imaging modalities for hepatocellular carcinomas (HCCs) in special (i.e., high-risk or unfavorable) locations compared with those in conventional locations.METHODSA total of 122 HCC patients were enrolled, including 85 patients (69.7%) with HCC in conventional locations and 37 (30.3%) with HCC in special locations. The clinical data, overall survival (OS), progression-free survival (PFS), and procedure-related adverse events were analyzed.RESULTSRFA combined with TACE was successfully performed in all patients. Three complications (2.5%) occurred, with no significant difference between the conventional (n=1, 1.2%) and special (n=2, 5.4%) locations (P = 0.218). Complete tumor necrosis rate was not significantly different between the conventional (n=73, 85.9%) and special (n=34, 91.9%) locations at one-month imaging (P = 0.353). After a follow-up of 3–48 months, the PFS was 17 months for patients with HCC in conventional locations and 14 months for patients with HCC in special locations; one-year PFS rate was 68.1% in the conventional location group, not significantly (P = 0.741) different from 59.1% in the special location group. The OS was 28 months in the conventional location group while 32 months in the special location group. The cumulative one- and two-year OS rates were 89.9% and 63.3%, respectively, in the conventional location group, not significantly different from 96.3% and 65% in the special location group (P = 0.273). Age (P = 0.043) and tumor size (P < 0.001) were significant prognostic factors for OS, and tumor size (P < 0.001) was the only significant prognostic factor for PFS.CONCLUSIONRFA guided by multiple imaging modalities combined with TACE may be safe and effective for treating HCCs in special locations
Circ-NOLC1 promotes epithelial ovarian cancer tumorigenesis and progression by binding ESRP1 and modulating CDK1 and RhoA expression
Circular RNAs (circRNAs) play important roles in cancer tumorigenesis and progression, representing prognostic biomarkers and therapeutic targets. In this case, we demonstrated the role of circ-NOLC1 in epithelial ovarian cancer (EOC). Our results have shown that Circ-NOLC1 expression was higher in EOC tissues than in normal tissues, and was positively associated with FIGO stage, differentiation. Among ovarian cancer cell lines, circ-NOLC1 expression was the highest in A2780, and lowest in CAOV3. Overexpression of circ-NOLC1 in CAOV3 cells increased cell proliferation, migration, and invasion ability, whereas silencing of circ-NOLC1 in A2780 cells had the opposite effect: however, neither circ-NOLC1 downregulation nor overexpression influenced NOLC1 mRNA expression. In nude mice with subcutaneous tumors, circ-NOLC1 downregulation decreased tumor growth. Bioinformatic analysis and RNA-binding protein immunoprecipitation showed that circ-NOLC1 could bind to ESRP1. In addition, the overexpression of circ-NOLC1 significantly increased ESRP1, RhoA, and CDK1 protein and mRNA expression level; circ-NOLC1 downregulation had the opposite effects. The tumor-promoting effect of circ-NOLC1 was inhibited by knockdown of ESRP1, CDK1, or RhoA expression in circ-NOLC1-overexpressing cells, which might act by modulating RhoA and CDK1 expression. In conclusion, our study demonstrated that Circ-NOLC1 might promote EOC tumorigenesis and development by binding ESRP1 and modulating CDK1 and RhoA expression
An Updated Search of Steady TeV Ray Point Sources in Northern Hemisphere Using the Tibet Air Shower Array
Using the data taken from Tibet II High Density (HD) Array (1997
February-1999 September) and Tibet-III array (1999 November-2005 November), our
previous northern sky survey for TeV ray point sources has now been
updated by a factor of 2.8 improved statistics. From to
in declination (Dec) range, no new TeV ray point
sources with sufficiently high significance were identified while the
well-known Crab Nebula and Mrk421 remain to be the brightest TeV ray
sources within the field of view of the Tibet air shower array. Based on the
currently available data and at the 90% confidence level (C.L.), the flux upper
limits for different power law index assumption are re-derived, which are
approximately improved by 1.7 times as compared with our previous reported

Proton-Boron Fusion Yield Increased by Orders of Magnitude with Foam Targets
A novel intense beam-driven scheme for high yield of the tri-alpha reaction
11B(p,{\alpha})2{\alpha} was investigated. We used a foam target made of
cellulose triacetate (TAC, C_9H_{16}O_8) doped with boron. It was then heated
volumetrically by soft X-ray radiation from a laser heated hohlraum and turned
into a homogenous, and long living plasma. We employed a picosecond laser pulse
to generate a high-intensity energetic proton beam via the well-known Target
Normal Sheath Acceleration (TNSA) mechanism. We observed up to 10^{10}/sr
{\alpha} particles per laser shot. This constitutes presently the highest yield
value normalized to the laser energy on target. The measured fusion yield per
proton exceeds the classical expectation of beam-target reactions by up to four
orders of magnitude under high proton intensities. This enhancement is
attributed to the strong electric fields and nonequilibrium thermonuclear
fusion reactions as a result of the new method. Our approach shows

Chromogranin A (CgA) as Poor Prognostic Factor in Patients with Small Cell Carcinoma of the Cervix: Results of a Retrospective Study of 293 Patients
BACKGROUND: Small cell carcinoma of the cervix (SCCC) is a very rare tumor. Due to its rarity and the long time period, there is a paucity of information pertaining to prognostic factors associated with survival. The objective of this study was to determine whether clinicopathologic finings or immunohistochemical presence of molecular markers predictive of clinical outcome in patients with SCCC. METHODOLOGY AND FINDINGS: We retrospectively reviewed a total of 293 patients with SCCC (47 patients from Cancer Center of Sun Yat-sen University in china, 71 patients from case report of china journal, 175 patients from case report in PubMed database). Of those 293 patients with SCCC, the median survival time is 23 months. The 3-year overall survival rates (OS) and 3-year disease-free survival rates (DFS) for all patients were 34.5% and 31.1%, respectively. Univariate and multivariate analysis showed that FIGO stage (IIb-IV VS I-IIa, Hazard Ratio (HR) = 3.08, 95% confidence interval (CI) of ratio = [2.05, 4.63], P<0.001), tumor mass size (≥ 4 cm VS <4 cm, HR = 2.37, 95% CI = [1.28, 4.36], P = 0.006) and chromogranin A (CgA) (Positive VS Negative, HR = 1.81, 95% CI = [1.12, 2.91], P = 0.015) were predictive of poor prognosis. CgA stained positive was found to be highly predictive of death in early-stage (FIGO I-IIa) patient specifically. CONCLUSIONS: Patients with SCCC have poor prognosis. FIGO stage, tumor mass size and CgA stained positive may act as a surrogate for factors prognostic of survival. CgA may serve as a useful marker in prognostic evaluation for early-stage patients with SCCC
Simulation Studies for the First Pathfinder of the CATCH Space Mission
The Chasing All Transients Constellation Hunters (CATCH) space mission is an
intelligent constellation consisting of 126 micro-satellites in three types (A,
B, and C), designed for X-ray observation with the objective of studying the
dynamic universe. Currently, we are actively developing the first Pathfinder
(CATCH-1) for the CATCH mission, specifically for type-A satellites. CATCH-1 is
equipped with Micro Pore Optics (MPO) and a 4-pixel Silicon Drift Detector
(SDD) array. To assess its scientific performance, including the effective area
of the optical system, on-orbit background, and telescope sensitivity, we
employ the Monte Carlo software Geant4 for simulation in this study. The MPO
optics exhibit an effective area of cm at the focal spot for 1 keV
X-rays, while the entire telescope system achieves an effective area of
cm at 1 keV when taking into account the SDD detector's detection
efficiency. The primary contribution to the background is found to be from the
Cosmic X-ray Background. Assuming a 625 km orbit with an inclination of
, the total background for CATCH-1 is estimated to be
counts s in the energy range of 0.5--4 keV. Based on
the background within the central detector and assuming a Crab-like source
spectrum, the estimated ideal sensitivity could achieve erg
cm s for an exposure of 10 s in the energy band of 0.5--4
keV. Furthermore, after simulating the background caused by low-energy charged
particles near the geomagnetic equator, we have determined that there is no
need to install a magnetic deflector

Large-scale genetic study in East Asians identifies six new loci associated with colorectal cancer risk
Known genetic loci explain only a small proportion of the familial relative risk of colorectal cancer (CRC). We conducted the largest genome-wide association study in East Asians with 14,963 CRC cases and 31,945 controls and identified six new loci associated with CRC risk (P = 3.42 × 10−8 to 9.22 × 10−21) at 10q22.3, 10q25.2, 11q12.2, 12p13.31, 17p13.3 and 19q13.2. Two of these loci map to genes (TCF7L2 and TGFB1) with established roles in colorectal tumorigenesis. Four other loci are located in or near genes involved in transcription regulation (ZMIZ1), genome maintenance (FEN1), fatty acid metabolism (FADS1 and FADS2), cancer cell motility and metastasis (CD9) and cell growth and differentiation (NXN). We also found suggestive evidence for three additional loci associated with CRC risk near genome-wide significance at 8q24.11, 10q21.1 and 10q24.2. Furthermore, we replicated 22 previously reported CRC loci. Our study provides insights into the genetic basis of CRC and suggests new biological pathways
