    Compound Bieshe Kangā€™ai inhibits proliferation and induces apoptosis in HCT116 human colorectal cancer cells

    Purpose: To study the effect of Compound Bieshe Kangā€™ai (CBK) on proliferation and apoptosis in colorectal cancer cells.Methods: HCT116 colorectal cancer cells and FHs 74 Int intestinal cells were treated with CBK, followed by determination of cell proliferation with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Caspase-9 and caspase-3 activities as well as protein expressions of Bcl-2 and BAX, and mRNA levels of caspase-9, caspase-3, Bcl-2 and BAX in HCT116 cells were evaluated, followed by examination of the morphological alterations of HCT116 cells with Hoechst 33342 staining.Results: CBK suppressed proliferation of HCT116 cells in a concentration- and time-dependent pattern, without cytotoxicity to FHs 74 Int cells. CBK also elevated caspase-9 and caspase-3 activities, mitigated protein translation of Bcl-2 and augmented that of BAX. It also enhanced mRNA transcriptions of caspase-9, caspase-3 and BAX, but decreased that of Bcl-2 in HCT116 cells in aĀ  concentrationdependent manner, as well as induced cancer cell shrinkage, nuclear fragmentation and chromatin condensation.Conclusion: The findings highlight CBK as a promising therapeutic agent for colorectal cancers, by retarding proliferation and inducing apoptosis in cancer cells.Keywords: Apoptosis, BAX, Bcl-2, Cancer, Caspase, Compound Bieshe Kangā€™ai, Chromatin condensation, Nuclear fragmentatio

    Metabolomic Analysis of Biochemical Changes in the Plasma of High-Fat Diet and Streptozotocin-Induced Diabetic Rats after Treatment with Isoflavones Extract of Radix Puerariae

    The main purpose of this study was to investigate the protective effects of total isoflavones from Radix Puerariae (PTIF) in diabetic rats. Diabetes was induced by a high-fat diet and intraperitoneal injection of low-dose streptozotocin (STZ; 40ā€‰mg/kg). At 26 weeks onwards, PTIF 421ā€‰mg/kg was administrated to the rats once daily consecutively for 10 weeks. Metabolic profiling changes were analyzed by Ultraperformance Liquid Chromatography-Quadrupole-Exactive Orbitrap-Mass Spectrometry (UPLC-Q-Exactive Orbitrap-MS). The principal component discriminant analysis (PCA-DA), partial least-squares discriminant analysis (PLS-DA), and orthogonal partial least-squares discriminant analysis (OPLS-DA) were used for multivariate analysis. Moreover, free amino acids in serum were determined by high-performance liquid chromatography with fluorescence detector (HPLC-FLD). Additionally, oxidative stress and inflammatory cytokines were evaluated. Eleven potential metabolite biomarkers, which are mainly related to the coagulation, lipid metabolism, and amino acid metabolism, have been identified. PCA-DA scores plots indicated that biochemical changes in diabetic rats were gradually restored to normal after administration of PTIF. Furthermore, the levels of BCAAs, glutamate, arginine, and tyrosine were significantly increased in diabetic rats. Treatment with PTIF could regulate the disturbed amino acid metabolism. Consequently, PTIF has great therapeutic potential in the treatment of DM by improving metabolism disorders and inhibiting oxidative damage

    Protective Effects of Total Flavones of Elaeagnus rhamnoides

    The aim was to evaluate the protective effects of total flavones of Elaeagnus rhamnoides (L.) A. Nelson (TFE) against vascular endothelial injury in blood stasis model rats and explore the potential mechanisms preliminarily. The model of blood stasis rat model with vascular endothelial injury was induced by subcutaneous injection of adrenaline combined with ice-water bath. Whole blood viscosity (WBV), histological examination, and prothrombin time (PT), activated partial thromboplastin time (APTT), and fibrinogen (FIB) were measured. Meanwhile, the levels of Thromboxane B2 (TXB2), 6-keto-PGF1Ī±, von Willebrand factor (vWF), and thrombomodulin (TM) were detected. In addition, Quantitative Real-Time PCR (qPCR) was performed to identify PI3K, Erk2, Bcl-2, and caspase-3 gene expression. The results showed that TFE can relieve WBV, increase PT and APTT, and decrease FIB content obviously. Moreover, TFE might significantly downregulate the levels of TXB2, vWF, and TM in plasma and upregulate the level of 6-keto-PGF1Ī± in plasma. Expressions of PI3K and Bcl-2 were increased and the expression of caspase-3 was decreased by TFE pretreatment in the rat model. Consequently, the study suggested that TFE may have the potential against vascular endothelial injury in blood stasis model rats induced by a high dose of adrenaline with ice-water bath

    Substantial transition to clean household energy mix in rural China

    The household energy mix has significant impacts on human health and climate, as it contributes greatly to many health- and climate-relevant air pollutants. Compared to the well-established urban energy statistical system, the rural household energy statistical system is incomplete and is often associated with high biases. Via a nationwide investigation, this study revealed high contributions to energy supply from coal and biomass fuels in the rural household energy sector, while electricity comprised āˆ¼20%. Stacking (the use of multiple sources of energy) is significant, and the average number of energy types was 2.8 per household. Compared to 2012, the consumption of biomass and coals in 2017 decreased by 45% and 12%, respectively, while the gas consumption amount increased by 204%. Increased gas and decreased coal consumptions were mainly in cooking, while decreased biomass was in both cooking (41%) and heating (59%). The time-sharing fraction of electricity and gases (E&G) for daily cooking grew, reaching 69% in 2017, but for space heating, traditional solid fuels were still dominant, with the national average shared fraction of E&G being only 20%. The non-uniform spatial distribution and the non-linear increase in the fraction of E&G indicated challenges to achieving universal access to modern cooking energy by 2030, particularly in less-developed rural and mountainous areas. In some non-typical heating zones, the increased share of E&G for heating was significant and largely driven by income growth, but in typical heating zones, the time-sharing fraction was <5% and was not significantly increased, except in areas with policy intervention. The intervention policy not only led to dramatic increases in the clean energy fraction for heating but also accelerated the clean cooking transition. Higher income, higher education, younger age, less energy/stove stacking and smaller family size positively impacted the clean energy transition

    Endovascular treatment of A1 segment aneurysms of the anterior cerebral artery

    Aneurysms of the A1 segment of the anterior cerebral artery (ACA) are rare and challenging to treat. We evaluated our experience of endovascular treatment in ni11 patients with A1 segment aneurysms of ACA. Seven aneurysms were treated with coiling; three were treated with stent-assisted coiling and one was treated with balloon-assited coiling. Endovascular treatments were technically successful and without complication. Follow-up examinations showed complete cure in all ni11 patients

    Numerical simulation of slipstreams and wake flows of trains with different nose lengths passing through a tunnel

    This study examined the slipstreams induced by high-speed trains (HSTs) passing through a tunnel using the improved delayed detached eddy simulation (IDDES) method. First, the flow fields in the open air and in a tunnel were compared. Furthermore, the flow in a tunnel was analyzed in detail, considering the development of both instantaneous flow structures and slipstream profiles at various measurement points. Finally, by considering four different nose lengths (4 m, 7 m, 9 m, and 12 m), the differences in the slipstream profiles and the wake flow induced by HSTs passing through a tunnel were determined. The results show that the piston effect had a significant influence on the slipstream profiles, causing a larger positive peak when a train passed through a tunnel. The peaks of the slipstream profiles decrease as the distances from the center of the track (COT) and the top of the rail (TOR) increases. The results show that a long nose length can reduce the scale and strength of the instantaneous x-vorticity and y-vorticity in the wake propagation region, thereby lowering the maximum slipstream peaks. The 12-m nose length train induced 56.7% lower velocity than the 4-m nose length train at y = 2 m beside the COT and z = 0.2 m above the TOR. In particular, the standard deviations of the positive peaks of the seven cross-sections decrease by 38.4% with the increase in the nose length from 4 m to 12 m, which means that a longer nose length can reduce the turbulence level in the wake propagation region. Consequently, from the perspectives of the safety and comfort of trackside people, a long nose length train is strongly recommended

    Uniformity, Periodicity and Symmetry Characteristics of Forces Fluctuation in Helical-Edge Milling Cutter

    Under most processing conditions, the milling force is expected to be stable and not to fluctuate, in order to improve the processing quality. This study focuses on analyzing the force fluctuation characteristics under conditions of different processing and cutter parameters. An original model is proposed to predict the force fluctuation during the milling process of a helical-edge cutter. At the same time, three force fluctuation characteristics related to the axial cutting depth and cutter parameters are determined: uniformity, periodicity and symmetry. The corresponding mathematical derivation and proof method are given for the first time through a force transformation of projecting the superposition chip thickness on a virtual cutting edge. On this basis, a fast estimation method and an accurate simulation method for force fluctuation prediction are established to quantify the intensity of force fluctuations under different parameters. Both two prediction methods and the experimental cutting tests validate the proposed theory effectively. The result shows a high potential of the proposed theory for studying the force behavior under different milling parameters or cutter parameters and at least 75% of the test workload can be reduced
