    Prevalence and genotyping of human isolates of Giardia duodenalis from Albania

    Microscopical and PCR-based techniques were performed in order to investigate the prevalence of infection and the genotypes of Giardia duodenalis from 125 stool samples collected from children living in the urban and the rural areas of Tirana (Albania) and hospitalized with acute gastroenteritis. 7 out of 125 samples resulted positive for Giardia at the microscopic examination (5.6%). In 50 selected samples including the 7 samples positive for Giardia by microscopy, 3 and 15 additional positive samples were detected by immunofluorescence and PCR, respectively. Seasonality appeared as an important parameter to be evaluated in order to better understand the prevalence of infection. Sequence analysis revealed both human Assemblage A and B.     S-acylation stabilizes ligand-induced receptor kinase complex formation during plant pattern-triggered immune signalling

    SummaryPlant receptor kinases are key transducers of extracellular stimuli, such as the presence of beneficial or pathogenic microbes or secreted signalling molecules. Receptor kinases are regulated by numerous post-translational modifications. Here, using the immune receptor kinases FLS2 and EFR, we show that S-acylation at a cysteine conserved in all plant receptor kinases is crucial for function. S-acylation involves the addition of long-chain fatty acids to cysteine residues within proteins, altering their biophysical properties and behaviour within the membrane environment. We observe S-acylation of FLS2 at C-terminal kinase domain cysteine residues within minutes following perception of its ligand flg22, in a BAK1 co-receptor dependent manner. We demonstrate that S-acylation is essential for FLS2-mediated immune signalling and resistance to bacterial infection. Similarly, mutating the corresponding conserved cysteine residue in EFR supressed elf18 triggered signalling. Analysis of unstimulated and activated FLS2-containing complexes using microscopy, detergents and native membrane DIBMA nanodiscs indicates that S-acylation stabilises and promotes retention of activated receptor kinase complexes at the plasma membrane to increase signalling efficiency

    Cruise ships like buildings:Wayfinding solutions to improve emergency evacuation

    This research aims at demonstrating that wayfinding solutions can improve the effectiveness of the evacuation processes in complex architectural spaces such as those of cruise ships. We investigated the behaviours of passengers in maritime disaster to figure out whether people act similarly during buildings egress and ships evacuation. Data were collected through questionnaires administered in 2015 to passengers boarding various cruise ships at the port of Ancona (Italy), and through the analysis of real footage of the evacuation of the Costa Concordia. The open source software Fire Dynamics Simulator with Evacuation (FDS+EVAC), used in building egress analyses, was adapted to include these behavioural and event information such as familiarity with ship layout, ship rotation and lifeboats boarding. Simulation results on the case study confirmed similarities between ships and buildings evacuations, underlining the effectiveness of wayfinding solutions to improve passengers’ evacuation flows and routes selection. This study also demonstrated that computer simulation could benefit the ship design process, the preparation of safety guidelines, and the crewmembers during naval emergency management training
