    Analytic regularity for a singularly perturbed system of reaction-diffusion equations with multiple scales: proofs

    We consider a coupled system of two singularly perturbed reaction-diffusion equations, with two small parameters 0<ϵ≤μ≤10< \epsilon \le \mu \le 1, each multiplying the highest derivative in the equations. The presence of these parameters causes the solution(s) to have \emph{boundary layers} which overlap and interact, based on the relative size of ϵ\epsilon and % \mu. We construct full asymptotic expansions together with error bounds that cover the complete range 0<ϵ≤μ≤10 < \epsilon \leq \mu \leq 1. For the present case of analytic input data, we derive derivative growth estimates for the terms of the asymptotic expansion that are explicit in the perturbation parameters and the expansion order

    An evaluation of low volume high-intensity intermittent training (HIIT) for health risk reduction in overweight and obese men

    Both sprint interval training (SIT) and high-intensity intermittent training (HIIT) have been described as time-efficient strategies for inducing favourable metabolic and cardiorespiratory adaptations in healthy and diseased participants. BACKGROUND: To date, little attention has been given to profiling the potential health benefits of HIIT or modified HIIT training within overweight and obese cohorts with particular focus on inflammation. Within this pilot trial, we tested the hypothesis that 6 sessions of HIIT performed over 2 weeks with 1-2 days’rest would improve aerobic capacity, glucose metabolism and inflammatory profile in an overweight and obese male cohort. Additionally, we profiled the potential health benefits of 4 HIIT sessions performed over the same period. METHODS: 18 overweight or obese males (BMI = 31.2 ± 3.6; V̇O2 = 30.3 ± 4.4 ml.kg.min-1) were studied before and 72 h after HIIT. Training sessions consisted of 10 x 1 min intervals at 90% HRpeak separated by 1 min recovery periods. Exercise was performed either 6 (group 1, n = 8) or 4 (group 2, n = 10)times over a 2 week period. RESULTS: After training no changes were detected from baseline for body composition, aerobic capacity, glucose metabolism or inflammatory profile(p > 0.05) in either group. CONCLUSION: Both 6 and 4 sessions of HIIT performed over a 2-week period are ineffective in improving selected health markers within an overweight and obese cohort

    Analysis of the Singular Function Boundary Integral Method for a Biharmonic Problem with One Boundary Singularity

    In this article, we analyze the singular function boundary integral method (SFBIM) for a two-dimensional biharmonic problem with one boundary singularity, as a model for the Newtonian stick-slip flow problem. In the SFBIM, the leading terms of the local asymptotic solution expansion near the singular point are used to approximate the solution, and the Dirichlet boundary conditions are weakly enforced by means of Lagrange multiplier functions. By means of Green&apos;s theorem, the resulting discretized equations are posed and solved on the boundary of the domain, away from the point where the singularity arises. We analyze the convergence of the method and prove that the coefficients in the local asymptotic expansion, also referred to as stress intensity factors, are approximated at an exponential rate as the number of the employed expansion terms is increased. Our theoretical results are illustrated through a numerical experiment

    Appetite regulatory hormone responses on the day following a prolonged bout of moderate-intensity exercise

    Exercise increases energy expenditure however acutely this does not cause compensatory changes in appetite or food intake. This unresponsiveness contrasts the rapid counter-regulatory changes seen after food restriction. The present investigation examined whether corrective changes in appetite-regulatory parameters occur after a time delay, namely, on the day after a single bout of exercise. Nine healthy males completed two, two-day trials (exercise & control) in a random order. On the exercise trial participants completed 90 min of moderate-intensity treadmill running on day one (10:30–12:00 h). On day two appetite-regulatory hormones and subjective appetite perceptions were assessed frequently in response to two test meals provided at 08:00 and 12:00 h. Identical procedures occurred in the control trial except no exercise was performed on day one. Circulating levels of leptin were reduced on the day after exercise (AUC 5841 ± 3335 vs. 7266 ± 3949 ng− 1·mL− 1 · 7 h, P = 0.012). Conversely, no compensatory changes were seen for circulating acylated ghrelin, total PYY, insulin or appetite perceptions. Unexpectedly, levels of acylated ghrelin were reduced on the exercise trial following the second test meal on day two (AUC 279 ± 136 vs. 326 ± 136 pg− 1·mL− 1 · 3 h, P = 0.021). These findings indicate that short-term energy deficits induced by exercise initially prompt a compensatory response by chronic but not acute hormonal regulators of appetite and energy balance. Within this 24 h time-frame however there is no conscious recognition of the perturbation to energy balance

    Multicomponent analysis of the tumour microenvironment reveals low CD8 T cell number, low stromal caveolin-1 and high tenascin-C and their combination as significant prognostic markers in non-small cell lung cancer

    The complex interplay of the tumour microenvironment (TME) and its role in disease progression and response to therapy is poorly understood. The majority of studies to date focus on individual components or molecules within the TME and so lack the power correlative analysis. Here we have performed a multi-parameter analysis of the TME in 62 resectable non-small cell lung cancer (NSCLC) specimens detailing number and location of immune infiltrate, assessing markers of cancer-associated fibroblasts, caveolin-1 and tenascin-C, and correlating with clinicopathological details, as well as markers of disease progression such as epithelial-to-mesenchymal transition (EMT). The influence of individual parameters on overall survival was determined in univariate and multivariate analysis and the combination of risk factors and interplay between components analysed. Low numbers of CD8 T cells, low stromal levels of caveolin-1 or high levels of tenascin-C were significant prognostic markers of decreased overall survival in both univariate and multivariate analysis. Patients with two or more risk factors had dramatically reduced overall survival and those with all three a median survival of just 7.5 months. In addition, low levels of tumour E-cadherin correlated with reduced immune infiltrate into the tumour nests, possibly linking EMT to the avoidance of CD8 T cell control. The multicomponent approach has allowed identification of the dominant influences on overall survival, and exploration of the interplay between different components of the TME in NSCLC

    Low HDL Cholesterol, Smoking and IL-13 R130Q Polymorphism are Associated with Myocardial Infarction in Greek Cypriot Males. A Pilot Study

    This study was carried out in Greek Cypriot males to identify risk factors that predispose to myocardial infarction (MI). Genetic and lipid risk factors were investigated for the first time in a Greek Cypriot male case-control study.Contrary to other studies, mean low density lipoprotein cholesterol did not differ between cases and controls. High density lipoprotein cholesterol on the other hand, although within normal range in cases and controls, was significantly higher in the control population. In agreement with many other studies, smoking was significantly more prevalent in cases compared with controls. In pooled cases and controls, smokers had a significantly lower HDL-C level compared with non-smokers. The frequency of the IL-13 R130Q homozygotes for the mutation (QQ), as well as the mutant allele were significantly higher in cases compared with controls. The IL-13 R130Q variant, or another locus, linked to it, may increase the risk of MI

    Local inhibition of microRNA-24 improves reparative angiogenesis and left ventricle remodeling and function in mice with myocardial infarction

    Myocardial infarction (MI) is the leading cause of death worldwide. MicroRNAs regulate the expression of their target genes, thus mediating a plethora of pathophysiological functions. Recently, miRNA-24 emerged as an important but controversial miRNA involved in post-MI responses. Here, we aimed at clarifying the effect of adenovirus-mediate intra-myocardial delivery of a decoy for miRNA-24 in a mouse MI model and to investigate the impact of miRNA-24 inhibition on angiogenesis and cardiovascular apoptosis. After MI induction, miRNA-24 expression was lower in the peri-infarct tissue and its resident cardiomyocytes and fibroblasts; while it increased in endothelial cells (ECs). Local adenovirus-mediated miRNA-24 decoy delivery increased angiogenesis and blood perfusion in the peri-infarct myocardium, reduced infarct size, induced fibroblast apopotosis and overall improved cardiac function. Notwithstanding these beneficial effects, miRNA-24 decoy increased cardiomyocytes apoptosis. In vitro, miRNA-24 inhibition enhanced ECs survival, proliferation and networking in capillary-like tubes and induced cardiomyocyte and fibroblast apoptosis. Finally, we identified eNOS as a novel direct target of miR-24 in human cultured ECs and in vivo. Our findings suggest that miRNA-24 inhibition exerts distinct biological effects on ECs, cardiomyocytes and fibroblasts. The overall result of post-infarction local miRNA-24 inhibition appears to be therapeutic

    We consider the numerical approximation of boundary layer phenomena occuring in many singularly perturbed problems in mechanics, such as plate and shell problems. We present guidelines for the effective resolution of such layers in the context of exisiting, commercial p and hp finite element (FE) version codes. We show that if high order, &quot;spectral&quot; elements are available, then just two elements are sufficient to approximate these layers at a near-exponential rate, independently of the problem parameters thickness or Reynolds number. We present hp mesh design principles for situations where both corner singularities and boundary layers are present
