    Icanbelimod (CBP-307), a next-generation Sphingosine-1-phosphate receptor modulator, in healthy men: pharmacokinetics, pharmacodynamics, safety, and tolerability in a randomized trial in Australia

    BackgroundIcanbelimod (formerly CBP-307) is a next-generation S1PR modulator, targeting S1PR1. In this first-in-human study, icanbelimod was investigated in healthy men in Australia.MethodsParticipants were randomized 3:1, double-blind, to icanbelimod or placebo in four single-dose cohorts (0.1 mg, 0.25 mg, 0.5 mg [n=8 per cohort], 2.5 mg [n=4]) or for 28-days once-daily treatment in two cohorts (0.15 mg, 0.25 mg [n=8 per cohort]). Participants in the 0.25-mg cohort received 0.1 mg on Day 1. Treatments were administered orally after fasting; following one-week washout, icanbelimod was administered after breakfast in the 0.5-mg cohort.ResultsIcanbelimod exposure increased rapidly and dose-dependently with single and multiple dosing (Tmax 4–7 hours). Lymphocyte counts decreased rapidly after single (-11%, 0.1 mg; -40%, 0.25 mg; -71%, 0.5 mg; -77%, 2.5 mg) and multiple doses (-49%, 0.15 mg; -75%, 0.25 mg), and recovered quickly, 7 days after dosing. After single-dose 0.5 mg, although a high-fat breakfast versus fasting did not affect maximal decrease, lymphocyte counts tended to be lower after breakfast across most timepoints up to 72 hours. Twenty-eight participants (63.6%) experienced mainly mild treatment-emergent adverse events (TEAEs). After single-dose icanbelimod, the most common TEAEs were headache (28.6%, n=6) and dizziness (19.0%, n=4). Three participants experienced transient bradycardia, with one serious, following single-dose 2.5 mg icanbelimod. After multiple-dose icanbelimod, the most common TEAEs were headache (50.0%, n=6) and lymphopenia (41.7%, n=5), and two participants withdrew due to non-serious TEAEs. Up-titration attenuated heart rate reductions.ConclusionIcanbelimod was well-tolerated up to 0.5 mg and effectively reduced lymphocyte counts.Clinical trial registrationClinicalTrials.gov, identifier NCT02280434.

    A mechanochemical synthesis of submicron-sized Li2S and a mesoporous Li2S/C hybrid for high performance lithium/sulfur battery cathodes

    Lithium sulfide, Li2S, is a promising cathode material for lithium–sulfur batteries (LSBs), with a high theoretical capacity of 1166 mA h g−1. However, it suffers from low cycling stability, low-rate capability and high initial activation potential. In addition, commercially available Li2S is of high cost and of large size, over ten microns, which further exacerbate its shortcomings as a sulfur cathode. Exploring new approaches to fabricate small-sized Li2S of low cost and to achieve Li2S cathodes of high electrochemical performance is highly desired. This work reports a novel mechanochemical method for synthesizing Li2S of high purity and submicron size by ball-milling LiH with sulfur in an Ar atmosphere at room temperature. By further milling the as-synthesized Li2S with polyacrylonitrile (PAN) followed by carbonization of PAN at 1000 °C, a Li2S/C hybrid with nano-sized Li2S embedded in a mesoporous carbon matrix is achieved. The hybrid with Li2S as high as 74 wt% shows a high initial capacity of 971 mA h g−1 at 0.1C and retains a capacity of 570 mA h g−1 after 200 cycles as a cathode material for LSBs. A capacity of 610 mA h g−1 is obtained at 1C. The synthesis method of Li2S is facile, environmentally benign, and of high output and low cost. The present work opens a new route for the scalable fabrication of submicron-sized Li2S and for the development of high performance Li2S-based cathodes

    Biogeography and Virulence of Staphylococcus aureus

    Staphylococcus aureus is commonly carried asymptomatically in the human anterior nares and occasionally enters the bloodstream to cause invasive disease. Much of the global diversity of S. aureus remains uncharacterised, and is not clear how disease propensity varies between strains, and between host populations.We compared 147 isolates recovered from five kindergartens in Chengdu, China, with 51 isolates contemporaneously recovered from cases of pediatric infection from the main hospital serving this community. The samples were characterised by MLST, the presence/absence of PVL, and antibiotic resistance profiling.Genotype frequencies within individual kindergartens differ, but the sample recovered from cases of disease shows a general enrichment of certain MLST genotypes and PVL positive isolates. Genotypes under-represented in the disease sample tend to correspond to a single sequence cluster, and this cluster is more common in China than in other parts of the world.Virulence propensity likely reflects a synergy between variation in the core genome (MLST) and accessory genome (PVL). By combining evidence form biogeography and virulence we demonstrate the existence of a "native" clade in West China which has lowered virulence, possibility due to acquired host immunity

    Soybean (Glycine max (L.) Merr.) is a typical short-day and thermophilic crop. This study aimed to reveal the required accumulative day length (ADL) and accumulative active temperature (AAT) for DSF (days of sowing to flowering) and DFM (days of flowering to maturity) in global soybeans. A sample consisted of 354 varieties from 27 countries in five geographic regions, which were tested in Nanjing, China in two spring-sowing and two summer-sowing seasons. The ADL and AAT were calculated from the climatological data provided by the Public Service of Nanjing Meteorological Bureau. The results showed that the average DSF and DFM of global soybeans were 41.0 d and 83.3 d, which required ADLDSF of 606.6 d·h and AATDSF of 1185.9 d·°C, ADLDFM of 1126.7 d·h and AATDFM of 2145.1 d·°C, respectively, all with a wide variation among/within geographic and MG(maturity-group)-set subpopulations. From the multiple regression of DSF and DFM on required ADL and AAT, the ADL, AAT and ADL×AAT contributed 38.5%, 44.79% and 17.10% to DSF variation and 86.98%, 11.42% and 0.54% to DFM variation, respectively, and their relative importance to DSF and DFM varied among the geographic and MG subpopulations. The geographic subpopulations matched only partially with the genomic marker clusters, indicating multiple genetic sources of each subpopulation and that genetic exchange happened among subpopulations
