    Novel Microfiber Sensor and Its Biosensing Application for Detection of hCG Based on a Singlemode-Tapered Hollow Core-Singlemode Fiber Structure

    A novel microfiber sensor is proposed and demonstrated based on a singlemode-tapered hollow core -singlemode (STHS) fiber structure. Experimentally a STHS with taper waist diameter of 26.5 Ī¼m has been fabricated and RI sensitivity of 816, 1601.86, and 4775.5 nm/RIU has been achieved with RI ranges from 1.3335 to 1.3395 , from 1.369 to 1.378, and from 1.409 to 1.4175 respectively, which agrees very well with simulated RI sensitivity of 885, 1517, and 4540 nm/RIU at RI ranges from 1.3335 to 1.337, from 1.37 to 1.374, and from 1.41 to 1.414 . The taper waist diameter has impact on both temperature and strain sensitivity of the sensor structure: (1) the smaller the waist diameter, the higher the temperature sensitivity, and experimentally 26.82 pm/Ā°C has been achieved with a taper waist diameter of 21.4 Ī¼m; (2) as waist diameter decrease, strain sensitivity increase and 7.62 pm/Ī¼Īµ has been achieved with a taper diameter of 20.3 Ī¼m. The developed sensor was then functionalized for human chorionic gonadotropin (hCG) detection as an example for biosensing application. Experimentally for hCG concentration of 5 mIU/ml, the sensor has 0.5 nm wavelength shift, equivalent to limit of detection (LOD) of 0.6 mIU/ml by defining 3 times of the wavelength variation (0.06 nm) as measurement limit. The biosensor demonstrated relatively good reproducibility and specificity, which has potential for real medical diagnostics and other applications

    The purpose of this study was to evaluate muscle activity of the prime movers and core stabilizers on stable and unstable surfaces during push-up exercise. Subject: Fourteen healthy male participants (age, 21.6 Ā±2.3 years; height, 174.7 Ā±8.1cm; weight, 68.2 Ā±16.4kg) without low back pain and shoulder injury in the past year were recruited. The participants completed push-up exercise in three conditions: on the ground, air disc and sling. EMG activities of external abdominal oblique, pectoralis major, and anterior deltoid muscles and elbow joint kinematics were recorded.     Dynamic Distribution Pruning for Efficient Network Architecture Search

    Network architectures obtained by Neural Architecture Search (NAS) have shown state-of-the-art performance in various computer vision tasks. Despite the exciting progress, the computational complexity of the forward-backward propagation and the search process makes it difficult to apply NAS in practice. In particular, most previous methods require thousands of GPU days for the search process to converge. In this paper, we propose a dynamic distribution pruning method towards extremely efficient NAS, which samples architectures from a joint categorical distribution. The search space is dynamically pruned every a few epochs to update this distribution, and the optimal neural architecture is obtained when there is only one structure remained. We conduct experiments on two widely-used datasets in NAS. On CIFAR-10, the optimal structure obtained by our method achieves the state-of-the-art 1.91.9\% test error, while the search process is more than 1,0001,000 times faster (only 1.51.5 GPU hours on a Tesla V100) than the state-of-the-art NAS algorithms. On ImageNet, our model achieves 75.2\% top-1 accuracy under the MobileNet settings, with a time cost of only 22 GPU days that is 100%100\% acceleration over the fastest NAS algorithm.     Association of the shuffling of Streptococcus pyogenes clones and the fluctuation of scarlet fever cases between 2000 and 2006 in central Taiwan

    <p>Abstract</p> <p>Background</p> <p>The number of scarlet fever occurrences reported between 2000 and 2006 fluctuated considerably in central Taiwan and throughout the nation. Isolates of <it>Streptococcus pyogenes </it>were collected from scarlet fever patients in central Taiwan and were characterized by <it>emm </it>sequencing and a standardized pulsed-field gel electrophoresis (PFGE) method. National weekly report data were collected for investigating epidemiological trends.</p> <p>Results</p> <p>A total of 23 <it>emm </it>types were identified in 1,218 <it>S. pyogenes </it>isolates. The five most prevalent <it>emm </it>types were <it>emm</it>12 (50.4%), <it>emm</it>4 (23.2%), <it>emm</it>1 (16.4%), <it>emm</it>6 (3.8%) and <it>emm</it>22 (3.0%). PFGE analysis with <it>Sma</it>I suggested that, with a few exceptions, strains with a common <it>emm </it>type belonged to the same clone. There were two large <it>emm</it>12 clones, one with DNA resistant to cleavage by <it>Sma</it>I. Each prevalent <it>emm </it>clone had major PFGE strain(s) and many minor strains. Most of the minor strains emerged in the population and disappeared soon after. Even some major strains remained prevalent for only 2ā€“3 years before declining. The large fluctuation of scarlet fever cases between 2000 and 2006 was associated with the shuffling of six prevalent <it>emm </it>clones. In 2003, the dramatic drop in scarlet fever cases in central Taiwan and throughout the whole country was associated with the occurrence of a severe acute respiratory syndrome (SARS) outbreak that occurred between late-February and mid-June in Taiwan.</p> <p>Conclusion</p> <p>The occurrences of scarlet fever in central Taiwan in 2000ā€“2006 were primarily caused by five <it>emm </it>types, which accounted for 96.8% of the isolates collected. Most of the <it>S. pyogenes </it>strains (as defined by PFGE genotypes) emerged and lasted for only a few years. The fluctuation in the number of scarlet fever cases during the seven years can be primarily attributed to the shuffling of six prevalent <it>emm </it>clones and to the SARS outbreak in 2003.</p

    Proteomic dissection of LPS-inducible, PHF8-dependent secretome reveals novel roles of PHF8 in TLR4-induced acute inflammation and T cell proliferation

    Endotoxin (LPS)-induced changes in histone lysine methylation contribute to the gene-specific transcription for control of inflammation. Still unidentified are the chromatin regulators that drive the transition from a transcriptional-repressive to a transcriptional-active chromatin state of pro-inflammatory genes. Here, using combined approaches to analyze LPS-induced changes in both gene-specific transcription and protein secretion to the extracellular compartment, we characterize novel functions of the lysine demethylase PHF8 as a pro-inflammatory, gene-specific chromatin regulator. First, in the LPS-induced, acute-inflamed macrophages, PHF8 knockdown led to both a reduction of pro-inflammatory factors and an increase in a transcriptional-repressive code (H3K9me2) written by the methyltransferase G9a. Through unbiased quantitative secretome screening we discovered that LPS induces the secretion of a cluster of PHF8-dependent, ā€˜tolerizableā€™ proteins that are related to diverse extracellular pathways/processes including those for the activation of adaptive immunity. Specifically, we determined that PHF8 promotes T-cell activation and proliferation, thus providing the first link between the epigenetic regulation of inflammation and adaptive immunity. Further, we found that, in the acute-inflamed macrophages, the acute-active PHF8 opposes the H3K9me1/2-writing activity of G9a to activate specific protein secretions that are suppressed by G9a in the endotoxin-tolerant cells, revealing the inflammatory-phenotypic chromatin drivers that regulate the gene-specific chromatin plasticity

    Exome sequencing study of 20 patients with high myopia

    Background High myopia is a common ocular disease worldwide. To expand our current understanding of the genetic basis of high myopia, we carried out a whole exome sequencing (WES) study to identify potential causal gene mutations. Methods A total of 20 individuals with high myopia were exome sequenced. A novel filtering strategy combining phenotypes and functional impact of variants was applied to identify candidate genes by multi-step bioinformatics analyses. Network and enrichment analysis were employed to examine the biological pathways involved in the candidate genes. Results In 16 out of 20 patients, we identified 20 potential pathogenic gene variants for high myopia. A total of 18 variants were located in myopia-associated chromosomal regions. In addition to the novel mutations found in five known myopia genes (ADAMTS18, CSMD1, P3H2, RPGR, and SLC39A5), we also identified pathogenic variants in seven ocular disease genes (ABCA4, CEP290, HSPG2, PCDH15, SAG, SEMA4A, and USH2A) as novel candidate genes. The biological processes associated with vision were significantly enriched in our candidate genes, including visual perception, photoreceptor cell maintenance, retinoid metabolic process, and cellular response to zinc ion starvation. Discussion Systematic mutation analysis of candidate genes was performed using WES data, functional interaction (FI) network, Gene Ontology and pathway enrichment. FI network analysis revealed important network modules and regulator linker genes (EP300, CTNNB1) potentially related to high myopia development. Our study expanded the list of candidate genes associated with high myopia, which increased the genetic screening performance and provided implications for future studies on the molecular genetics of myopia

    AGROBEST: an efficient Agrobacterium-mediated transient expression method for versatile gene function analyses in Arabidopsis seedlings

    Background: Transient gene expression via Agrobacterium-mediated DNA transfer offers a simple and fast method to analyze transgene functions. Although Arabidopsis is the most-studied model plant with powerful genetic and genomic resources, achieving highly efficient and consistent transient expression for gene function analysis in Arabidopsis remains challenging. Results: We developed a highly efficient and robust Agrobacterium-mediated transient expression system, named AGROBEST (Agrobacterium-mediated enhanced seedling transformation), which achieves versatile analysis of diverse gene functions in intact Arabidopsis seedlings. Using Ī²-glucuronidase (GUS) as a reporter for Agrobacterium-mediated transformation assay, we show that the use of a specific disarmed Agrobacterium strain with vir gene pre-induction resulted in homogenous GUS staining in cotyledons of young Arabidopsis seedlings. Optimization with AB salts in plant culture medium buffered with acidic pH 5.5 during Agrobacterium infection greatly enhanced the transient expression levels, which were significantly higher than with two existing methods. Importantly, the optimized method conferred 100% infected seedlings with highly increased transient expression in shoots and also transformation events in roots of ~70% infected seedlings in both the immune receptor mutant efr-1 and wild-type Col-0 seedlings. Finally, we demonstrated the versatile applicability of the method for examining transcription factor action and circadian reporter-gene regulation as well as protein subcellular localization and proteinā€“protein interactions in physiological contexts. Conclusions: AGROBEST is a simple, fast, reliable, and robust transient expression system enabling high transient expression and transformation efficiency in Arabidopsis seedlings. Demonstration of the proof-of-concept experiments elevates the transient expression technology to the level of functional studies in Arabidopsis seedlings in addition to previous applications in fluorescent protein localization and proteinā€“protein interaction studies.     Curbing Bribe-Giving in Malaysia: The Role of Attitudes and Parents

While there are many efforts to curb bribery, most focused at macro level, such as law, while little has been examined at the micro level, e.g., individual behavior and intention. Those who did investigate at the micro level tend to focus on the recipients rather than the ones giving the bribe. We explored eight factors that influence Malaysian young adultsā€™ bribe giving intention based on the Reasoned Action Approach (RAA). A total of 345 respondents (M age = 20.68, SD = 2.01, 189 are females) completed questionnaires about all RAA variables. Partial Least Squares Structural Equation Modelling (PLS-SEM) was carried out using smartPLS3.0 to analyze the result. The result revealed that out of the eight variables, four variablesā€”Instrumental attitudes, Experiential Attitudes, Parentsā€™ descriptive norms and Capacityā€”explain 74% of the variance in bribe giving intention. An important take-away is that young adultā€™s perception of whether their parents gave or did not give bribes in a given situation is important in influencing their bribe giving intention. Bribe giving prevention messages must be targeted explicitly toward parents, where they play a crucial role in curbing this dishonesty.Peer reviewe

    Chondroprotective effects and mechanisms of resveratrol in advanced glycation end products-stimulated chondrocytes

    [[abstract]]INTRODUCTION: Accumulation of advanced glycation end products (AGEs) in joints contributes to the pathogenesis of cartilage damage in osteoarthritis (OA). We aim to explore the potential chondroprotective effects of resveratrol on AGEs-stimulated porcine chondrocytes and cartilage explants. METHODS: Chondrocytes were isolated from pig joints. Activation of the IkappaB kinase (IKK)-IkappaBalpha-nuclear factor-kappaB (NF-kappaB) and c-Jun N-terminal kinase (JNK)/extracellular signal-regulated kinase (ERK)-activator protein-1 (AP-1) pathways was assessed by electrophoretic mobility shift assay (EMSA), Western blot and transfection assay. The levels of inducible nitric oxide synthase (iNOS)-NO and cyclooxygenase-2 (COX-2)-prostaglandin E2 (PGE2) were measured by Western blot, Griess reaction or ELISA. The expression and enzyme activity of matrix metalloproteinase-13 (MMP-13) were determined by real time RT/PCR and gelatin zymography, respectively. RESULTS: We show that AGEs-induced expression of iNOS and COX-2 and production of NO and PGE2 were suppressed by resveratrol. Such effects of resveratrol were likely mediated through inhibiting IKK-IkappaBalpha-NF-kappaB and JNK/ERK-AP-1 signaling pathways induced by AGEs. By targeting these critical signaling pathways, resveratrol decreased AGEs-stimulated expression and activity of MMP-13 and prevented AGEs-mediated destruction of collagen II. Histochemistry analysis further confirms that resveratrol could prevent AGEs-induced degradation of proteoglycan and aggrecan in cartilage explants. CONCLUSIONS: The present study reveals not only the effects and mechanisms regarding how resveratrol may protect cartilage from AGEs-mediated damage but also the potential therapeutic benefit of resveratrol in the treatment of OA

    Thymoquinone attenuates liver fibrosis via PI3K and TLR4 signaling pathways in activated hepatic stellate cells

    Thymoquinone (TQ) is the major active compound derived from the medicinal Nigella sativa. In the present study, we investigated the anti-fibrotic mechanism of TQ in lipopolysaccharide (LPS)-activated rat hepatic stellate cells line, T-HSC/Cl-6. T-HSC/Cl-6 cells were treated with TQ (3.125, 6.25 and 12.5 Ī¼M) prior to LPS (1 Ī¼g/ml). Our data demonstrated that TQ effectively decreased activated T-HSC/Cl-6 cell viability. TQ significantly attenuated the expression of CD14 and Toll-like receptor 4 (TLR4). TQ also significantly inhibited phosphatidylinositol 3-kinase (PI3K) and serine/threonine kinase-protein kinase B (Akt) phosphorylation. The expression of Ī±-SMA and collagen-I were significantly decreased by TQ. Furthermore, TQ decreased X linked inhibitor of apoptosis (XIAP) and cellular FLIP (c-FLIP L ) expression, which are related with the regulation of apoptosis. Furthermore, TQ significantly increased the survival against LPS challenge in D-galactosamine (D-GlaN)-sensitized mice, and decreased the levels of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), which were in line with in vitro results. Our data demonstrated that TQ attenuates liver fibrosis partially via blocking TLR4 expression and PI3K phosphorylation on the activated HSCs. Therefore, TQ may be a potential candidate for the therapy of hepatic fibrosis
