Interannual variability of heat waves in South Korea and their connection with large-scale atmospheric circulation patterns
This study investigates the interannual variation of heat wave frequency (HWF) in South Korea during the past 42 years (1973-2014) and examines its connection with large-scale atmospheric circulation changes. Korean heat waves tend to develop most frequently in late summer during July and August. The leading Empirical Orthogonal Function accounting for 50% of the total variance shows a mono-signed pattern over South Korea, suggesting that the dominant mechanisms responsible for the heat wave are linked in a spatial scale much larger than the nation. It also exhibits a regional variation with more occurrences in the southeastern inland area. The regression of the leading principal component (PC) time series of HWF with large-scale atmospheric circulation identifies a north-south dipole pattern between the South China Sea and Northeast Asia. When this large-scale circulation mode facilitates deep convection in South China Sea, it tends to weaken moisture transport from the South China Sea to Northeast Asia. Enhanced deep convection in the South China Sea triggers a source of Rossby wave train along southerly wind that generates positive geopotential height anomalies around Korea. The anomalous high pressure pattern is accompanied by large-scale subsidence in Korea, thereby providing a favourable condition for extreme hot and dry days in Korea. This study highlights that there is a decadal change of the relationship between Korean heat waves and large-scale atmospheric circulation patterns. Modelling and Simulation of a River-Crossing Operation via Discrete Event Simulation with Engineering Details
From a military standpoint, a river is an area that should be avoided in a potential engagement because of lack of cover and the necessity of dividing the unit while crossing. Thus, a key point of a river-crossing operation is speed. Many efforts have been made to enable faster river crossing by improvement of tactics, techniques, and procedures (TTP). However, improvements in TTP are evaluated by modelling and simulation much less frequently than are the toe-to-toe engagements between two opposing forces, and to our knowledge, this is the first simulation model of brigade-level river crossing with engineering details. This study presents a simulation model of the river-crossing operation, applies real world parameters, and evaluates which tactics are preferable in a particular operational environments. This analysis has led to new operational methods of river crossing that have been suggested by experienced subject-matter experts. For instance, the current Republic of Korea Army Field Manual dictates to rotate river-crossing rafts in all situations, but our experiment suggests that no rotation is preferable when the width of river is less than 400 m based on the statistical analyses, which includes the regression-based meta-modelling and the ANOVA, of our simulation model that embodies the engineering details of river-crossing equipment.Defence Science Journal, Vol. 65, No. 2, March 2015, pp.135-143, DOI:http://dx.doi.org/10.14429/dsj.65.814
Observational Evidence of EHP Effects on the Melting of Snowpack over the Tibetan Plateau
Observational evidences are presented showing that the Indo-Gangetic Plain (IGP) regions, bounded by the high altitude Himalayan mountains, are subject to heavy loading of absorbing aerosols, i.e., black carbon and dust, which can lead to widespread enhancement warming over the Tibetan Plateau and accelerated snowmelt in the western Tibetan Plateau (WTP) and Himalayas. The two pre-monsoon seasons of 2004 and 2005 were strikingly contrasting in terms of the aerosol loading over IGP. The warming of the TP in 2004 relative to 2005 was widespread, covering most of the WTP and Himalayas. This warming is closely linked to patterns of the snow melt. Consistent with the Elevated Heat Pump hypothesis, we find that increased loading of absorbing aerosols over IGP in the pre-monsoon season is associated with increased heating of the upper troposphere by dynamical feedback induced by aerosol heating, and enhances the rate of snowmelt over Himalayas and the WTP in April-May. Composite analysis with more contrasting years also shows that the heating of the troposphere by elevated dust and black carbon aerosols in the boreal spring can lead to widespread enhanced land-atmosphere warming, and accelerated snow melt in the Himalayas and Tibetan Plateau
Fatigue Prediction of the Discharge Pipe in Reciprocating Compressor
In this paper, a fatigue prediction of the line discharge tube for reciprocating compressor being installed in a refrigerator was studied. The tube usually gets plenty of the repeated loads caused by the start and stop motion of a reciprocating compressor. There are two representative methods to predict the fatigue stress. At first the stress-life can be applied to the problem which takes a lot of repeated stress within the elastic strain range. Second is the strain-life method which can be used when it comes to the problem of a small repeated stress in the plastic strain range. This paper presents the stress-life method how the design parameters of a discharge pipe relate to the fatigue prediction and analyzes the co-relation between them
ArmA and RmtB Were the Predominant 16S RMTase Genes Responsible for Aminoglycoside-resistant Isolates in Korea
Pathogenic gram-negatives that produce 16S ribosomal RNA methyltransferases (16S RMTases) have already been distributed all over the world. To investigate the predominance of aminoglycoside resistance associated with 16S RMTases in Korea, we collected a total of 222 amikacin resistant Gram-negative clinical isolates from patient specimens between 1999 and 2015 from three hospital banks across Korea. ArmA and nntB were the predominant 16S RMTase genes responsible for aminoglycoside-resistant isolates circulating in Korean community settings although only one rmtA-producing isolate was detected in 2006.1
Synergetic strengthening of layered steel sheet investigated using an in situ neutron diffraction tensile test
Synergetic strengthening induced by plastic strain incompatibility at the interface, and the resulting extra geometrically necessary dislocations (GNDs) generated during plastic deformation, were investigated to understand the origin of extra strength in heterogeneous structured (HS) materials. The mechanism of extra GND generation in twinning-induced plasticity (TWIP)-interstitial free (IF) steel layered sheet was quantitatively analyzed by conducting in situ neutron scattering tensile test. Load partitioning due to the different mechanical properties between the TWIP-steel core and IF-steel sheath at the TWIP/IF interface was observed during the in situ tensile testing. Because of the plastic strain incompatibility from load partitioning, extra GNDs are generated and saturate during tensile deformation. The extra GNDs can be correlated with the back-stress evolution of the HS materials, which contributes to the strength of layered materials. Because of the back-stress evolution caused by load partitioning, the strength of TWIP-IF layered steel is higher than the strength estimated by the rule-of-mixtures. Identification of new CYP2C19 variants exhibiting decreased enzyme activity in the metabolism of S-mephenytoin and omeprazole. Drug Metab
ABSTRACT: Although many cases of interindividual variation in the metabolism of CYP2C19 drugs are explained by the CYP2C19*2, *3, and *17, a wide range of metabolic variation still occurs in people who do not carry these genetic variants. The objectives of this study were to identify new genetic variants and to characterize functional consequences of these variants in metabolism of CYP2C19 substrates. In total, 21 single-nucleotide polymorphisms including three new coding variants, V394M, E405K, and D256N, were identified by direct DNA sequencing in 50 randomly selected subjects and in individuals who exhibited an outlier phenotype response in the omeprazole study. Recombinant proteins produced from the coding variants V394M, E405K, and D256N were prepared by using an Escherichia coli expression system and purified. Metabolism of S-mephenytoin and omeprazole by V394M was comparable with that of the wild-type protein. E405K showed a moderate decrease in metabolism of the substrates. However, D256N exhibited a significantly decreased activity in S-mephenytoin metabolism, resulting in 50 and 76% decreases in V max and intrinsic clearance, respectively, compared with the wild type. This variant also exhibited a significant decrease in omeprazole metabolism in vivo. CYP2C19 D256N and E405K were assigned as CYP2C19*26 and *2D, respectively, by the Cytochrome P450 Nomenclature Committee. In summary, this report characterizes the allele frequency and haplotype distribution of CYP2C19 in a Korean population and provides functional analysis of new coding variants of the CYP2C19 gene. Our findings suggest that individuals carrying CYP2C19*26 would have lower activity for metabolizing CYP2C19 substrate drugs
Bioanalysis of alpelisib using liquid chromatography–tandem mass spectrometry and application to pharmacokinetic study
Abstract
Alpelisib is the first alpha-specific phosphatidylinositol-3-kinase (PI3K) inhibitor indicated for the treatment of hormone receptor-positive, human epidermal growth factor receptor 2-negative, PI3K catalytic subunit alpha-mutated, advanced, or metastatic breast cancer. Substantial attempts have been made to extend its clinical use to other types of cancer. Analytical methods proven to accurately quantify alpelisib would improve the reliability of the preclinical and clinical data of alpelisib. Therefore, we developed and validated a quantification method based on liquid chromatography–tandem mass spectrometry for alpelisib in mouse and human plasma samples. Alpelisib and an internal standard (IS; enzalutamide) were separated from endogenous substances using an XTerra MS C18 column with a linear gradient of 0.1% formic acid in water and 0.1% formic acid in acetonitrile. Multiple reaction monitoring transitions for alpelisib and the IS were m/z 442.1 > 328.0 and m/z 465.0 > 209.1, respectively. The calibration curve for alpelisib was confirmed to be linear in the range of 1–2000ng/mL in both mouse and human plasma. The intra- and inter-day accuracy and precision met the acceptance criteria, and no significant matrix effects were observed. Alpelisib was stable under various storage and handling conditions, and the carryover effect was overcome using the injection loop flushing method. We successfully used this assay to study the in vitro metabolic profiles and in vivo pharmacokinetics of alpelisib in mice. Here, to the best of our knowledge, we report for the first time a valid quantitative method for alpelisib in mouse and human plasma, which could aid in providing valuable pharmacokinetic information on alpelisib to increase its clinical availability
