    Wolbachia-Mediated Antibacterial Protection and Immune Gene Regulation in Drosophila

    The outcome of microbial infection of insects is dependent not only on interactions between the host and pathogen, but also on the interactions between microbes that co-infect the host. Recently the maternally inherited endosymbiotic bacteria Wolbachia has been shown to protect insects from a range of microbial and eukaryotic pathogens. Mosquitoes experimentally infected with Wolbachia have upregulated immune responses and are protected from a number of pathogens including viruses, bacteria, Plasmodium and filarial nematodes. It has been hypothesised that immune upregulation underpins Wolbachia-mediated protection. Drosophila is a strong model for understanding host-Wolbachia-pathogen interactions. Wolbachia-mediated antiviral protection in Drosophila has been demonstrated for a number of different Wolbachia strains. In this study we investigate whether Wolbachia-infected flies are also protected against pathogenic bacteria. Drosophila simulans lines infected with five different Wolbachia strains were challenged with the pathogenic bacteria Pseudomonas aeruginosa PA01, Serratia marcescens and Erwinia carotovora and mortality compared to paired lines without Wolbachia. No difference in mortality was observed in the flies with or without Wolbachia. Similarly no antibacterial protection was observed for D. melanogaster infected with Wolbachia. Interestingly, D. melanogaster Oregon RC flies which are naturally infected with Wolbachia showed no upregulation of the antibacterial immune genes TepIV, Defensin, Diptericin B, PGRP-SD, Cecropin A1 and Attacin D compared to paired flies without Wolbachia. Taken together these results indicate that Wolbachia-mediated antibacterial protection is not ubiquitous in insects and furthermore that the mechanisms of antibacterial and antiviral protection are independent. We suggest that the immune priming and antibacterial protection observed in Wolbachia-infected mosquitoes may be a consequence of the recent artificial introduction of the symbiont into insects that normally do not carry Wolbachia and that antibacterial protection is unlikely to be found in insects carrying long-term Wolbachia infections

    A practical method for assessing lip compression strengthening in healthy adults

    © 2020 by the authors. There is no practical and accessible assessment method to evaluate lip muscle compression strength. The purpose of this study was to examine the relationship between the standard method (i.e., Iowa Oral Performance Instrument) and a practical method in healthy adults. In order to achieve our research purpose, ninety-eight healthy adults (18-40 years) completed lip compression strength measurements (standard method) and lip grasping performance tests using a standard recyclable plastic water bottle (practical method). In the overall sample, the mean and standard deviation for standard method and practical method was 26.7 (7.0) kPa and 255 (119) g, respectively. For the overall sample (n = 98), there was a positive relationship between the two strength tasks [r = 0.56 (0.41, 0.68)]. When separated by sex, positive correlations were observed for men and women with no differences between the observed correlations [difference of 0.06 (-0.2646, 0.3917)]. This result indicates that those individuals who are strong in the standard task will often be strong in the practical task. Future research is needed to determine how well changes in each test track with each other in response to a lip strength training program

    Assessments of facial muscle thickness by ultrasound in younger adults: Absolute and relative reliability

    © 2019 by the authors. The absolute reliability (i.e., standard error of measurement and minimal difference) of a measurement is important to consider when assessing training effects. However, the absolute reliability for ultrasound measured facial muscle thickness had not been investigated. In order to examine the absolute and relative reliability of measuring facial muscles, 98 healthy, young, and middle-aged adults (18-40 years) had ultrasound measurements taken twice, separated by an average of three days. Six facial muscles were selected to determine the reliability of facial muscle thickness. The relative reliability (ICC3,1) ranged from 0.425 for the orbicularis oris (inferior) to 0.943 for the frontalis muscle. The absolute reliability (minimal difference) ranged from 0.25 mm for the orbicularis oculi to 1.82 mm for the masseter. The percentage minimal difference was 22%, 25%, 26%, 29%, 21%, and 10% for the frontalis, orbicularis oculi, orbicularis oris (superior), orbicularis oris (inferior), depressor anguli oris, and masseter, respectively. Our results indicated that the relative reliability was similar to that observed previously. The absolute reliability indicated that the measurement error associated with measuring muscle thickness of the face may be greater than that of the trunk/limb muscles. This may be related to the difficulty of accurately determining the borders of each muscle

    NMR spectroscopy of native and in vitro tissues implicates polyADP ribose in biomineralization.

    Nuclear magnetic resonance (NMR) spectroscopy is useful to determine molecular structure in tissues grown in vitro only if their fidelity, relative to native tissue, can be established. Here, we use multidimensional NMR spectra of animal and in vitro model tissues as fingerprints of their respective molecular structures, allowing us to compare the intact tissues at atomic length scales. To obtain spectra from animal tissues, we developed a heavy mouse enriched by about 20% in the NMR-active isotopes carbon-13 and nitrogen-15. The resulting spectra allowed us to refine an in vitro model of developing bone and to probe its detailed structure. The identification of an unexpected molecule, poly(adenosine diphosphate ribose), that may be implicated in calcification of the bone matrix, illustrates the analytical power of this approach

    Recursive partitioning vs computerized adaptive testing to reduce the burden of health assessments in cleft lip and/or palate : comparative simulation study

    Background: Computerized adaptive testing (CAT) has been shown to deliver short, accurate, and personalized versions of the CLEFT-Q patient-reported outcome measure for children and young adults born with a cleft lip and/or palate. Decision trees may integrate clinician-reported data (eg, age, gender, cleft type, and planned treatments) to make these assessments even shorter and more accurate. Objective: We aimed to create decision tree models incorporating clinician-reported data into adaptive CLEFT-Q assessments and compare their accuracy to traditional CAT models. Methods: We used relevant clinician-reported data and patient-reported item responses from the CLEFT-Q field test to train and test decision tree models using recursive partitioning. We compared the prediction accuracy of decision trees to CAT assessments of similar length. Participant scores from the full-length questionnaire were used as ground truth. Accuracy was assessed through Pearson’s correlation coefficient of predicted and ground truth scores, mean absolute error, root mean squared error, and a two-tailed Wilcoxon signed-rank test comparing squared error. Results: Decision trees demonstrated poorer accuracy than CAT comparators and generally made data splits based on item responses rather than clinician-reported data. Conclusions: When predicting CLEFT-Q scores, individual item responses are generally more informative than clinician-reported data. Decision trees that make binary splits are at risk of underfitting polytomous patient-reported outcome measure data and demonstrated poorer performance than CATs in this study
