SelbständigkeitserklärungTrotz einer umfangreichen pferdespezifischen Literatur bestehen immer noch
erhebliche Defizite im Wissen über das Verhalten von Pferden, ihr
Akklimatisationsvermögen und ihre Anpassungsfähigkeit an ihre Umwelt.
Unabhängig davon ist, ob es sich um Wildpferde oder domestizierte Hauspferde
handelt. Für die Gestaltung einer naturnahen Pferdehaltung ist das Verständnis
für das Wesen Pferd von grundlegender Bedeutung. Für den optimalen Umgang mit
Pferden bis hin zum Entwickeln seiner Leistung, ist es wichtig, mehr über das
artspezifische natürliche Verhalten, seine Befindlichkeiten und Wünsche zu
erfahren und dem Pferd so mehr Wohlbefinden und mehr Lebensqualität und
Lebensfreude zu schenken. Das Ziel dieser Arbeit ist, Einblicke in die
Lebensgewohnheiten, Tagesrhythmen und wetterbedingten saisonalen
Verhaltensmuster einer Pferdeherde zu geben, welche neue Anregungen und
größeres Wissen in die ganzjährige Weidehaltung bringen soll. Dazu wurde in
einer einjährigen Beobachtungszeit das Nahrungsaufnahmeverhalten, das
Ausruhverhalten, das Komfortverhalten, und das Bewegungsverhalten und die
Wohnraumgestaltung und -nutzung von einer unter naturnahen Bedingungen
lebenden Pferdeherde in der Schorfheide (Deutschland) im Rahmen einer
visuellen Beobachtung untersucht. Analysen über die Nutzung verschiedener
räumlicher Ressourcen stellen das spezifische Verhalten in Bezug auf
klimatische Bedingungen dar. Zusätzlich wurden telemetrische Daten über das
Speichersystem ETHOSYS gewonnen. Die Pferde leben so gut wie ohne menschliche
Betreuung auf 80 ha Weideland und dienen vordergründig der Landschaftspflege.
Daher sollen Aspekte im Hinblick auf die Bedürfnisse und das Wohlbefinden der
Tiere in ihrem natürlichen Lebensraum dargestellt und beurteilt werden. Die
Nahrungsaufnahme und das Ruheverhalten nehmen die meiste Zeit des 24-Stunden-
Tages ein, jedoch verfügen die Pferde über eine gewisse kontinuierliche
Dynamik, die in Jahres-, Monats- und Tagesverläufen dargestellt werden kann.
Deutlich kristallisiert sich ein 4-Stunden-Rhythmus heraus mit dem
Sonnenaufgang als ersten Bezugspunkt. Dieser Rhythmus wird alle vier
Jahreszeiten durchgehalten und beginnt in dieser Arbeit mit der morgendlich
ersten Fressperiode. Trotz äußeren klimatischen Einflüssen wie Kälte, Hitze,
Nässe und Wind weichen die Liebenthaler Pferde nur geringfügig von diesem
kontinuierlichen Tagesrhythmus ab. Leichte Verschiebungen ereignen sich bei
extremer Hitze, wenn die Pferde stärkere Fressaktivität in die Abendstunden
verlagern oder bei extremer Kälte, wenn die Pferde geschützte Bereiche
aufsuchen und dort ruhen, um Energie zu sparen. Die Daten der telemetrischen
Messungen über die Aktivität der Pferde bestätigen die visuelle Beobachtung.
Die Darstellung der Aktivitätsmessungen zeigt darüber hinaus eine
Jahresrhythmik mit Hauptfressperioden während des Sonnenaufgangs und vor dem
Sonnenuntergang. Die Analyse des Wohnraumes ergibt, dass sehr große Hitze die
Pferde weniger in ihrem Rhythmus beeinflusst als viel mehr eine hohe
Insektenbelastung. Nässe ohne Wind stört die Pferde nicht, wohingegen sie bei
Regen in Kombination mit Wind gerne geschützte Bereiche aufsuchen. Kälte,
besonders in der Nacht, veranlasst die Tiere, sich in windgeschützte Bereiche
zu begeben und erst im Morgengrauen auf die freie Fläche zurück zu kehren.
Wichtiger ist also ein Schutz vor starkem Wind für die Tiere, als ein Schutz
vor Nässe. Daher wäre das Beste, wenn beides den Pferden zur freien Verfügung
stehen würde.Although there is a great horse specific literature, there still is a big
deficiency about the behaviour of horses, their capability of acclimatization
and their adaptability to the environment. It is independent, whether they are
wild horses or domesticated horses. For the arrangement of a natural horse
keeping it is very important to know about the being of horses. In order to
create the best possible relationship to the horses up to the development of
their abilities, it is very important to know more about the art specific
behavior of them, their feelings and their wishes, so that they have a better
quality and comfort of life. The aim of this work was to show the inside of
the habits, day rhythms and the behaviours depending on weather seasons if a
herd of horses, which should give new suggestions and greater knowledge in the
whole seasons horse keeping on feedlots. Therefore 100 horses in a herd were
under observation for 1 year. Their habits in eating, resting, welfare and
moving were observed even though the arrangement and the using of their living
area in a natural environment of the Schorfheide in Germany. The analysis of
using the different capabilities of the area showed the specific behaviour
connected with the climatic conditions. Telemetric dates from the memory
system ETHOSYS were added. The horses lived on an 80 acres field without being
looked after and are used for landscape cultivation. Therefore the aspects of
their needs and their wellbeing in the horses´ natural environment should be
shown and judged. The eating and resting habits took up most of the 24 hours,
but there was a continuously dynamic movement in the horses, which could be
seen in the course of the year, month and day. Very clearly a 4 hour rhythm is
found with the sunrise as the starting point. This rhythm continuous through
all seasons and starts in this work with the first eating period in the
morning. Although outside climatic influenced such as cold, heat, wetness and
wind the Liebenthaler horses deviated only a bit from this continual day
rhythm. Little changes took place with extreme heat, when the horses had their
biggest eating activity in the evening hours or with extreme cold, when the
horses went to the protected areas to rest for saving energy. The result of
the telemetric measures about the activity of the horses confirm with the
observation. The graph of the activity measure showed that in a year rhythm
the high period of eating was in the sun rise and before sun down. The
analysis of the living areas showed that big heat affected the horses´ rhythm
less than the high insect pestering. Wetness without wind had not bothered the
horses, but rain combined with wind made them look for protected areas.
Especially cold at night time made the horses go to wind protected areas and
at sun rise they came back to the free areas. It is more important for the
horses to be protected against the wind, than to be protected against the
wetness. It would be better to protect them from both

"PolyMin": software for identification of the minimum number of polymorphisms required for haplotype and genotype differentiation
Background
Analysis of allelic variation for relevant genes and monitoring chromosome segment transmission during selection are important approaches in plant breeding and ecology. To minimize the number of required molecular markers for this purpose is crucial due to cost and time constraints. To date, software for identification of the minimum number of required markers has been optimized for human genetics and is only partly matching the needs of plant scientists and breeders. In addition, different software packages with insufficient interoperability need to be combined to extract this information from available allele sequence data, resulting in an error-prone multi-step process of data handling. Results
PolyMin, a computer program combining the detection of a minimum set of single nucleotide polymorphisms (SNPs) and/or insertions/deletions (INDELs) necessary for allele differentiation with the subsequent genotype differentiation in plant populations has been developed. Its efficiency in finding minimum sets of polymorphisms is comparable to other available program packages. Conclusion
A computer program detecting the minimum number of SNPs for haplotype discrimination and subsequent genotype differentiation has been developed, and its performance compared to other relevant software. The main advantages of PolyMin, especially for plant scientists, is the integration of procedures from sequence analysis to polymorphism selection within a single program, including both haplotype and genotype differentiation
Einfluss von Ethanol auf Methylhydroxypropylcellulose stabilisierte Öl-in-Wasser Emulsionen
Die vorliegende Arbeit beschreibt den Einfluss von Ethanol auf die physiko-chemischen Eigenschaften von Methylhydroxypropylcellulose (MHPC)-Lösungen und MHPC-stabilisierten Öl-in-Wasser Emulsionen, die als Ölphase mittelkettige Triglyceride enthalten. Da Qualität und Stabilität der Emulsionen maßgeblich durch die äußere Phase bestimmt werden, ist eine genaue Kenntnis der Eigenschaften der MHPC-Lösungen von Bedeutung. Die Polymerlöslichkeit wird durch Trübungsmessungen untersucht. Mit Hilfe der Viskosimetrie werden die hydrodynamischen Eigenschaften der MHPC charakterisiert. Die Grenzflächeneigenschaften der Polymere werden mittels Tensiometrie und Ellipsometrie untersucht. Die Charakterisierung und Stabilitätsbeurteilung der entsprechenden ethanolhaltigen Emulsionen erfolgt durch laserdiffraktometrische Bestimmung der Tropfengrößen sowie durch rotations- und oszillationsrheologische Messungen. Weiterhin wird die Freisetzung des Modellwirkstoffes Ibuprofen aus Emulsionen mit einer in situ Methode untersucht. Die Tropfengröße der Emulsionen wird im wesentlichen durch den Energieeintrag während der Herstellung und durch die Grenzflächenspannung zwischen wässriger Phase und Ölphase bestimmt. Mit steigendem Ethanolgehalt nimmt die Tropfengröße aufgrund sinkender Grenzflächenspannung ab. Rheologische Untersuchungen zeigen, dass die Emulsionen mit dem 'hard sphere model' als Dispersion harter Kugeln beschrieben werden können. Emulsionen mit einen Ethanolgehalt der wässrigen Phase bis zu 20 sind bei 20°C über einen Zeitraum von 12 Monaten lagerstabil. Die Freisetzung von Ibuprofen aus der Ölphase der Emulsionen erfolgt rasch und vollständig und wird durch Ethanol nur geringfügig beeinflusst.This work deals with the influence of ethanol on the physico-chemical properties of Hydroxypropylmethylcellulose (HPMC) solutions and oil-in-water emulsions, which contain HPMC as emulsifier and medium chain triglyceride as the dispersed phase. Since the quality and the stability of the emulsions are substantially influenced by the external phase, an exact knowledge of the properties of the HPMC solutions is required. The HPMC-solubility is investigated by the turbidimetry. Using the viscosimetry method the hydrodynamic parameters for HPMC are detected. The interfacial properties of the polymers are examined by the tensiometry and the ellipsometry. The characterization and stability testing of the corresponding emulsions were carried out by determine the oil droplet size using a laser diffraction method and by rheological methods as rotary and oscillatory experiments. Furthermore the release of the model drug Ibuprofen from the oil droplets is examined using an in situ method. The droplet size of the emulsion depend on the energy input during the preparation and on the interfacial tension between the aqueous phase and the oil phase. With increasing ethanol content (< 20 ) the droplet size decreases due to sinking interfacial tension. Rheological measurements show that the emulsions can be described with the 'hard sphere model' indicating that HPMC forms a stable interfacial film around the oil droplets. All emulsions with an ethanol content < 20 are storage stable during a period of 12 months at 20°C. The release of Ibuprofen from the oil droplets takes place rapidly and completely. Improved tolerance to drought stress after anthesis due to priming before anthesis in wheat (Triticum aestivum L.) var. Vinjett.
Drought stress occurring during the reproductive growth stage leads to considerable reductions in crop production and has become an important limiting factor for food security globally. In order to explore the possible role of drought priming (pre-exposure of the plants to mild drought stress) on the alleviation of a severe drought stress event later in development, wheat plants were subjected to single or double mild drought episodes (soil relative water content around 35–40%) before anthesis and/or to a severe drought stress event (soil relative water content around 20–25%) 15 d after anthesis. Here, single or double drought priming before anthesis resulted in higher grain yield than in non-primed plants under drought stress during grain filling. The photosynthesis rate and ascorbate peroxidase activity were higher while malondialdehyde content was lower in primed plants than in the non-primed plants under drought stress during grain filling. Proteins in flag leaves differently expressed by the priming and drought stress were mainly related to photosynthesis, stress defence, metabolism, molecular chaperone, and cell structure. Furthermore, the protein abundance of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) small subunit, Rubisco activase and ascorbate peroxidase were upregulated in primed plants compared with non-primed plants under drought stress during grain filling. In conclusion, the altered protein expression and upregulated activities of photosynthesis and ascorbate peroxidase in primed plants may indicate their potential roles in alleviating a later-occurring drought stress episode, thereby contributing to higher wheat grain yield under drought stress during grain filling
