    Modeling T Tauri Winds from He I 10830 Profiles

    The high opacity of He I 10830 makes it an exceptionally sensitive probe of the inner wind geometry of accreting T Tauri stars. In this line blueshifted absorption below the continuum results from simple scattering of stellar photons, a situation which is readily modeled without definite knowledge of the physical conditions and recourse to multi-level radiative transfer. We present theoretical line profiles for scattering in two possible wind geometries, a disk wind and a wind emerging radially from the star, and compare them to observed He I 10830 profiles from a survey of classical T Tauri stars. The comparison indicates that subcontinuum blueshifted absorption is characteristic of disk winds in ~30% of the stars and of stellar winds in ~40%. We further conclude that for many stars the emission profile of helium likely arises in stellar winds, increasing the fraction of accreting stars inferred to have accretion-powered stellar winds to ~60%. Stars with the highest disk accretion rates are more likely to have stellar wind than disk wind signatures and less likely to have redshifted absorption from magnetospheric funnel flows. This suggests the possibility that when accretion rates are high, disks can extend closer to the star, magnetospheric accretion zones can be reduced in size and conditions arise that favor radially outflowing stellar winds.Comment: 41 pages, 11 figures.     The infection efficacy of Metarhizium strains (Hypocreales: Clavicipitaceae) against the Queensland fruit fly Bactrocera tryoni (Diptera: Tephritidae)

    The Queensland fruit fly (Qfly), Bactrocera tryoni Froggatt, is a devastating pest of Australia's commercial fruit systems. Fruit fly mitigation is heavily centered around the use chemical insecticides, with limited investigation into microbial control alternatives. The wet tropics of northern Queensland is a highly biodiverse ecosystem containing many species of insect pathogenic fungi, but it is unclear whether any of these entomopathogens could contribute to Qfly management programs. In laboratory trials, we investigated the potential for Qfly microbial control by 3 locally sourced strains of entomopathogenic fungi comprising 2 species, Metarhizium guizhouense (Chen and Guo) and Metarhizium lepidiotae (Driver and Milner). Additionally, we evaluated 2 different inoculation methods to derive the most effective way to expose the flies to conidia-either through dry conidia or in a conidial suspension. All 3 strains were successful in causing Qfly mortality. Metarhizium lepidiotae resulted in the highest mean mortality over the trials, while M. guizhouense resulted in the highest mortality in a single replicate. Laboratory experiments revealed exposure through dry conidia to be the most effective method to inoculate the flies. These results suggest that fungal entomopathogens could be a viable pathway to Qfly suppression

    DNMT inhibitors reverse a specific signature of aberrant promoter DNA methylation and associated gene silencing in AML

    <b>Background</b>. Myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) are neoplastic disorders of hematopoietic stem cells. DNA methyltransferase inhibitors (DNMTi), 5-azacytidine (AzaC) and 5-aza-2’-deoxycytidine (Decitabine), benefit some MDS/AML patients. However, the role of DNMTi-induced DNA hypomethylation in regulation of gene expression in AML is unclear.<p></p> <b>Results. </b> We compared the effects of AzaC on DNA methylation and gene expression using whole-genome single-nucleotide bisulfite-sequencing (WGBS) and RNA-sequencing in OCI-AML3 (AML3) cells. For data analysis, we used an approach recently developed for discovery of differential patterns of DNA methylation associated with changes in gene expression, that is tailored to single-nucleotide bisulfite-sequencing data (Washington University Interpolated Methylation Signatures (WIMSi)). By this approach, a subset of genes upregulated by AzaC was found to be characterized by AzaC-induced signature methylation loss flanking the transcription start site. These genes are enriched for genes increased in methylation and decreased in expression in AML3 cells compared to normal hematopoietic stem and progenitor cells. Moreover, these genes are preferentially upregulated by Decitabine in human primary AML blasts, and control cell proliferation, death and development. <p></p> <b>Conclusions.</b> Our WGBS and WIMSi data analysis approach has identified a set of genes whose is methylation and silencing in AML is reversed by DNMTi. These genes are good candidates for direct regulation by DNMTi, and their reactivation by DNMTi may contribute to therapeutic activity. This study also demonstrates the ability of WIMSi to reveal relationships between DNA methylation and gene expression, based on single-nucleotide bisulfite-sequencing and RNA-seq data.<p></p&gt

    Diagnosis of lipomatous hypertrophy of the atrial septum by two-dimensional echocardiography

    Originally described in 1964, lipomatous hypertrophy of the atrial septum currently remains a diagnosis established primarily at autopsy. Clinical interest in this disorder has centered on the reported association with supraventricular arrhythmias and sudden death. Because two-dimensional echocardiography allows detailed assessment of atrial septal configuration, we reviewed two-dimensional echocardiographic reports obtained over a 1 year period and identified 17 patients who had features consistent with lipomatous hypertrophy of the atrial septum. Nine were men and the average age was 70 years. Autopsy confirmation of the echographic findings was possible in one patient. In nine patients, ideal body weight was exceeded by 10% or more. The atrial septum viewed from the subcostal transducer position showed a distinctive echo-dense globular thickening sparing the valve of the fossa ovalis. The resultant tomographic image of the atrial septum had a characteristic dumbbell appearance. The mean thickness of the atrial septum was 21 mm (range 15 to 29). Seven patients had supraventricular arrhythmias, and eight had P wave abnormalities.The two-dimensional echocardiographic features described are distinctive and suggest that this technique is the procedure of choice not only for establishing the diagnosis of lipomatous hypertrophy of the atrial septum but also for providing a means for prospective follow-up of patients with this little known entity
