    Cultural Consensus on Salmon Fisheries and Ecology in the Copper River, Alaska

    This study assessed levels of agreement in knowledge and opinions about salmon fisheries and ecology of the Copper River, Alaska, in three user groups: the Ahtna, an Alaska Native people indigenous to the upper river; commercial fishers who fish at the mouth of the river; and fishery managers and biologists with jurisdiction over the entire watershed. We anticipated that cultural background, academic training, long-term experience on the water, and spatial focus would be reflected in each group’s knowledge and opinions. Cultural consensus analysis showed agreement within each group, similar opinions between commercial fishers and managers and biologists, and distinct Ahtna opinions. Managers and biologists were the most cohesive group; they related to the entire watershed and relied on quantitative information as the basis for fisheries management. Ahtna focused on the upper river and incorporated observed long-term sociocultural, economic, and environmental changes into their opinions about the fisheries. Commercial fishers focused on the lower river and had strong familiarity with scientific principles of fisheries management. The similar views of commercial fishers and managers and biologists may result from the fact that commercial fishers’ economic success also depends on their understanding of fisheries management. To respond to socioeconomic and ecological sustainability issues, fisheries management would benefit from recognizing these perspectives and promoting participation of all stakeholder groups and effective communication among them.Cette Ă©tude a permis d’évaluer les degrĂ©s d’accord en matiĂšre de connaissances et d’opinions sur l’écologie et la pĂȘche au saumon de la riviĂšre Copper, en Alaska, chez trois groupes d’utilisateurs : les Ahtna, peuple autochtone de l’Alaska natif du haut de la riviĂšre; les pĂȘcheurs commerciaux qui pĂȘchent Ă  l’embouchure de la riviĂšre; et les gestionnaires et biologistes des pĂȘches qui ont compĂ©tence sur l’ensemble du bassin versant. Nous nous attendions Ă  ce que les connaissances et les opinions de chaque groupe soient rattachĂ©es aux antĂ©cĂ©dents culturels, Ă  l’expĂ©rience acadĂ©mique, Ă  l’expĂ©rience Ă  long terme de la pĂȘche et l’orientation spatiale. L’analyse du consensus culturel a permis de dĂ©montrer un accord au sein de chaque groupe, des opinions similaires entre les pĂȘcheurs commerciaux et les biologistes-gestionnaires, et des opinions distinctes chez les Ahtna.     Antibiotic-refractory Lyme arthritis is associated with HLA-DR molecules that bind a Borrelia burgdorferi peptide

    An association has previously been shown between antibiotic-refractory Lyme arthritis, the human histocompatibility leukocyte antigen (HLA)–DR4 molecule, and T cell recognition of an epitope of Borrelia burgdorferi outer-surface protein A (OspA163–175). We studied the frequencies of HLA-DRB1-DQA1-DQB1 haplotypes in 121 patients with antibiotic-refractory or antibiotic-responsive Lyme arthritis and correlated these frequencies with in vitro binding of the OspA163–175 peptide to 14 DRB molecules. Among the 121 patients, the frequencies of HLA-DRB1-DQA1-DQB1 haplotypes were similar to those in control subjects. However, when stratified by antibiotic response, the frequencies of DRB1 alleles in the 71 patients with antibiotic-refractory arthritis differed significantly from those in the 50 antibiotic-responsive patients (log likelihood test, P = 0.006; exact test, P = 0.008; effect size, Wn = 0.38). 7 of the 14 DRB molecules (DRB1*0401, 0101, 0404, 0405, DRB5*0101, DRB1*0402, and 0102) showed strong to weak binding of OspA163–175, whereas the other seven showed negligible or no binding of the peptide. Altogether, 79% of the antibiotic-refractory patients had at least one of the seven known OspA peptide–binding DR molecules compared with 46% of the antibiotic-responsive patients (odds ratio = 4.4; P < 0.001). We conclude that binding of a single spirochetal peptide to certain DRB molecules is a marker for antibiotic-refractory Lyme arthritis and might play a role in the pathogenesis of the disease

    Acute Inflammatory Profiles Differ with Sex and Age After Spinal Cord Injury

    Background Sex and age are emerging as influential variables that affect spinal cord injury (SCI) recovery. Despite a changing demographic towards older age at the time of SCI, the effects of sex or age on inflammation remain to be elucidated. This study determined the sex- and age-dependency of the innate immune response acutely after SCI. Methods Male and female mice of ages 4- and 14-month-old received T9 contusion SCI and the proportion of microglia, monocyte-derived macrophages (MDM), and neutrophils surrounding the lesion were determined at 3- and 7-day post-injury (DPI) using flow cytometry. Cell counts of microglia and MDMs were obtained using immunohistochemistry to verify flow cytometry results at 3-DPI. Microglia and MDMs were separately isolated using fluorescence-activated cell sorting (FACS) at 3-day post-injury (DPI) to assess RNA expression of 27 genes associated with activation, redox, and debris metabolism/clearance. Results Flow cytometry revealed that being female and older at the time of injury significantly increased MDMs relative to other phagocytes, specifically increasing the ratio of MDMs to microglia at 3-DPI. Cell counts using immunohistochemistry revealed that male mice have more total microglia within SCI lesions that can account for a lower MDM/microglia ratio. With NanoString analyses of 27 genes, only 1 was differentially expressed between sexes in MDMs; specifically, complement protein C1qa was increased in males. No genes were affected by age in MDMs. Only 2 genes were differentially regulated in microglia between sexes after controlling for false discovery rate, specifically CYBB (NOX2) as a reactive oxygen species (ROS)-associated marker as well as MRC1 (CD206), a gene associated with reparative phenotypes. Both genes were increased in female microglia. No microglial genes were differentially regulated between ages. Differences between microglia and MDMs were found in 26 of 27 genes analyzed, all expressed higher in MDMs with three exceptions. Specifically, C1qa, cPLA2, and CD86 were expressed higher in microglia. Conclusions These findings indicate that inflammatory responses to SCI are sex-dependent at both the level of cellular recruitment and gene expression

    A Conditional Yeast E1 Mutant Blocks the Ubiquitin–Proteasome Pathway and Reveals a Role for Ubiquitin Conjugates in Targeting Rad23 to the Proteasome

    E1 ubiquitin activating enzyme catalyzes the initial step in all ubiquitin-dependent processes. We report the isolation of uba1-204, a temperature-sensitive allele of the essential Saccharomyces cerevisiae E1 gene, UBA1. Uba1-204 cells exhibit dramatic inhibition of the ubiquitin–proteasome system, resulting in rapid depletion of cellular ubiquitin conjugates and stabilization of multiple substrates. We have employed the tight phenotype of this mutant to investigate the role ubiquitin conjugates play in the dynamic interaction of the UbL/UBA adaptor proteins Rad23 and Dsk2 with the proteasome. Although proteasomes purified from mutant cells are intact and proteolytically active, they are depleted of ubiquitin conjugates, Rad23, and Dsk2. Binding of Rad23 to these proteasomes in vitro is enhanced by addition of either free or substrate-linked ubiquitin chains. Moreover, association of Rad23 with proteasomes in mutant and wild-type cells is improved upon stabilizing ubiquitin conjugates with proteasome inhibitor. We propose that recognition of polyubiquitin chains by Rad23 promotes its shuttling to the proteasome in vivo

    Ghosts in the Garden: locative gameplay and historical interpretation from below

The heritage industry now makes extensive use of digital audioguides and similar interpretation tools to reach new audiences but many remain rooted in authoritative and didactic conservatism. This paper critically evaluates the state of play in the field, from downloadable audio tours and apps, through more complex engagements with theatrically enhanced and affective simulation, to attempts at fuller dialogic visitor participation and the use of gps or RFID-triggered game mechanics. While ‘armchair’ and home screen-based game and interpretation models are addressed, particular attention is paid to the use of mobile and locative design, where embodiment in place is privileged over less associative or remote experience. The paper takes a research project led by the author as a case study. Ghosts in the Garden was conceived in collaboration with a museum and an experience design SME to test the potential of immersive, affective real world games on public understandings of history. It sought to engage visitors with researched history from below by using a pervasive media soundscape, the ‘ghosts’ of past visitors and a ‘choose-your-own-adventure’ game mechanic in which outcomes are variable, visitor agency is retained and a more radical model of historical knowledge suggested
