    Sample size determination for bibliographic retrieval studies

    <p>Abstract</p> <p>Background</p> <p>Research for developing search strategies to retrieve high-quality clinical journal articles from MEDLINE is expensive and time-consuming. The objective of this study was to determine the minimal number of high-quality articles in a journal subset that would need to be hand-searched to update or create new MEDLINE search strategies for treatment, diagnosis, and prognosis studies.</p> <p>Methods</p> <p>The desired width of the 95% confidence intervals (W) for the lowest sensitivity among existing search strategies was used to calculate the number of high-quality articles needed to reliably update search strategies. New search strategies were derived in journal subsets formed by 2 approaches: random sampling of journals and top journals (having the most high-quality articles). The new strategies were tested in both the original large journal database and in a low-yielding journal (having few high-quality articles) subset.</p> <p>Results</p> <p>For treatment studies, if W was 10% or less for the lowest sensitivity among our existing search strategies, a subset of 15 randomly selected journals or 2 top journals were adequate for updating search strategies, based on each approach having at least 99 high-quality articles. The new strategies derived in 15 randomly selected journals or 2 top journals performed well in the original large journal database. Nevertheless, the new search strategies developed using the random sampling approach performed better than those developed using the top journal approach in a low-yielding journal subset. For studies of diagnosis and prognosis, no journal subset had enough high-quality articles to achieve the expected W (10%).</p> <p>Conclusion</p> <p>The approach of randomly sampling a small subset of journals that includes sufficient high-quality articles is an efficient way to update or create search strategies for high-quality articles on therapy in MEDLINE. The concentrations of diagnosis and prognosis articles are too low for this approach.</p

    Search Filters for Finding Prognostic and Diagnostic Prediction Studies in Medline to Enhance Systematic Reviews

    Background: The interest in prognostic reviews is increasing, but to properly review existing evidence an accurate search filer for finding prediction research is needed. The aim of this paper was to validate and update two previously introduced search filters for finding prediction research in Medline: the Ingui filter and the Haynes Broad filter. Methodology/Principal Findings: Based on a hand search of 6 general journals in 2008 we constructed two sets of papers. Set 1 consisted of prediction research papers (n = 71), and set 2 consisted of the remaining papers (n = 1133). Both search filters were validated in two ways, using diagnostic accuracy measures as performance measures. First, we compared studies in set 1 (reference) with studies retrieved by the search strategies as applied in Medline. Second, we compared studies from 4 published systematic reviews (reference) with studies retrieved by the search filter as applied in Medline. Next -using word frequency methods - we constructed an additional search string for finding prediction research. Both search filters were good in identifying clinical prediction models: sensitivity ranged from 0.94 to 1.0 using our hand search as reference, and 0.78 to 0.89 using the systematic reviews as reference. This latter performance measure even increased to around 0.95 (range 0.90 to 0.97) when either search filter was combined with the additional string that we developed. Retrieval rate of explorative prediction research was poor, both using our hand search or our systematic review as reference, and even combined with our additional search string: sensitivity ranged from 0.44 to 0.85. Conclusions/Significance: Explorative prediction research is difficult to find in Medline, using any of the currently available search filters.     STARD 2015: An Updated List of Essential Items for Reporting Diagnostic Accuracy Studies.

    Incomplete reporting has been identified as a major source of avoidable waste in biomedical research. Essential information is often not provided in study reports, impeding the identification, critical appraisal, and replication of studies. To improve the quality of reporting of diagnostic accuracy studies, the Standards for Reporting of Diagnostic Accuracy Studies (STARD) statement was developed. Here we present STARD 2015, an updated list of 30 essential items that should be included in every report of a diagnostic accuracy study. This update incorporates recent evidence about sources of bias and variability in diagnostic accuracy and is intended to facilitate the use of STARD. As such, STARD 2015 may help to improve completeness and transparency in reporting of diagnostic accuracy studies

    Sex-Specific Modulation of Cell Proliferation by Socially Relevant Stimuli in the Adult Green Treefrog Brain (Hyla cinerea)

    Social experience plays an important role in regulating the neural, physiological and hormonal changes that accompany the expression of reproductive behavior in vertebrates. This suite of functions is sexually dimorphic, with different neural control areas preeminent in males and females. In anuran amphibians, social experience comes in the form of acoustic communication, which is central to their reproductive behavior. We sought to determine whether acoustic cues regulate cell proliferation in the brain of adult green treefrogs (Hyla cinerea). Our results show that both male and female treefrogs that heard a conspecific chorus during the breeding season exhibited increased brain cell proliferation compared to animals that heard random tones. Increased cell proliferation, as assessed by the number of 5-bromo-2′-deoxyuridine-immunoreactive (BrdU+) cells, were found near the ventricles of acoustically sensitive brain regions such as the preoptic area (POA) and the infundibular hypothalamus (IF). Sex differences emerged in the location of this socially modulated cell proliferation: increases occurred primarily in the male POA and the female IF. In addition, gonadal steroid hormones might have played a role in the social modulation of cell proliferation: by statistically control- ling for hormone level, we revealed that androgens might influence socially induced increases in BrdU+ cells in the male POA, but estrogen did not contribute to socially induced increases in the female IF. These results indicate that the reception of social cues increases cell proliferation in brain regions mediating sexual behavior and endocrine regulation, and moreover that social modulation of cell proliferation occurs in a sexually differentiated fashion

    Changes in Plasma Testosterone Levels and Brain AVT Cell Number during the Breeding Season in the Green Treefrog

    We exposed groups of adult male green treefrogs, Hyla cinerea, to acoustic stimuli (natural chorus or random tones) for seven consecutive nights at three time points during their natural breeding season (May, July, and September) and assessed seasonal changes in plasma androgen levels and number of arginine vasotocin (AVT) immunoreactive cells in the brain over this time period. We also tested whether social cues altered either androgens or AVT-ir cell number or size at each time point. Finally, we analyzed how these factors related to calling behavior. Data were collected over two breeding seasons. Call rate (calls/h) was assessed during the stimulus time (i.e. ‘evoked calling’) and during the remainder of the day (‘spontaneous calling’). Plasma hormone levels were measured at the end of the acoustic treatment when brains were collected for immunocytochemistry. Circulating androgen levels declined over the breeding season. Males exposed to chorus sounds, however, had higher androgen levels than males exposed to tones. AVT-ir cell number increased across the breeding season in the nucleus accumbens but not the amygdala, anterior preoptic area, or magnocellular preoptic area, and soma size decreased in the nucleus accumbens as cell number increased. Social stimulation had no significant influence on either AVT-ir cell measure. Evoked call rate was higher in males exposed to natural chorus sounds compared to those exposed to random tones, but did not change during the season. In contrast, spontaneous call rate was higher at the beginning of the breeding season compared to the end, and unlike evoked calling was correlated with circulating androgen levels across all treatments and time points. AVT-ir soma size was positively correlated with both evoked and spontaneous calling. These results suggest that social exposure can prolong the elevation of gonadal hormones in the bloodstream, thus mitigating or slowing the seasonal decline of such hormones. In contrast, social exposure does not affect the seasonal pattern of AVT-ir cell number or soma size. The reciprocal relationship between social cues and hormones and the subsequent effect on behavior may provide hidden benefits to animals engaging in social interactions. However, unlike steroid hormone levels, the seasonal change in AVT-ir cell number and size is not counteracted by social stimulation
