    This work records the results of a study of the spinal cords, spinal sensory ganglia, sympathetic ganglia, Gasserian ganglia and neurones in the adrenal glands of sheep affected with scrapie disease.It was suggested that many minor changes in sympathetic ganglia may be of a non -specific nature and it was concluded that there was no significant morphological difference, including vacuolation of neurones, between the gelglia of clinically healthy sheep and those with scrapie disease

    The Relationship Between Deuterium Excess and Uranium Groundwater Concentrations in Whitehall, Montana

    Uranium can become elevated in the environment and pose a human health risk to water resources. Therefore, it is important to understand the origin, transport and concentration of uranium in the environment. With this exploratory study we propose that deuterium (ÎŽ2H) excess may be a viable way to study uranium groundwater concentrations that originate from the surface. The Boulder Batholith of southwestern Montana has been associated with varying levels of uranium. The Whitehall, MT area includes the Boulder Batholith as a geologic unit. We collected 23 water samples and compiled data from a study done by the United States Geological Survey (USGS) within the area of Whitehall, MT. With a linear regression we show that there is a significant negative relationship between deuterium excess and uranium water concentrations. As expected we showed that there is a significant positive, linear relationship between uranium water concentrations and nitrate water concentrations. As expected we also showed that there is a significant positive, linear relationship between calcium carbonate and uranium groundwater concentrations for the compiled USGS data. The results of a Wilcoxon rank sum test showed that median uranium concentration for wells close to ponds is 14.5 ÎŒg/L higher for our data added to the USGS data. These results may be consistent with a model that involves insoluble uranium being transported in suspension and settling out in ponds where an influx of nitrates and/or carbonates react to produce higher concentrations of water-soluble forms of uranium. We suggest that the relationship found between uranium concentration and deuterium excess in water samples may be related to the higher rates of evaporation in ponds. Thus, in this specific hydrologic environment of valley ponds close to the Boulder Batholith source, deuterium excess may serve to help predict levels of uranium concentrations

    Targeting Versican as a Potential Immunotherapeutic Strategy in the Treatment of Cancer.

    A growing body of literature links events associated with the progression and severity of immunity and inflammatory disease with the composition of the tissue extracellular matrix as defined by the matrisome. One protein in the matrisome that is common to many inflammatory diseases is the large proteoglycan versican, whose varied function is achieved through multiple isoforms and post-translational modifications of glycosaminoglycan structures. In cancer, increased levels of versican are associated with immune cell phenotype, disease prognosis and failure to respond to treatment. Whether these associations between versican expression and tumour immunity are the result of a direct role in the pathogenesis of tumours is not clear. In this review, we have focused on the role of versican in the immune response as it relates to tumour progression, with the aim of determining whether our current understanding of the immunobiology of versican warrants further study as a cancer immunotherapy target

    G1 Domain of Versican Regulates Hyaluronan Organization and the Phenotype of Cultured Human Dermal Fibroblasts

    Variants of versican have wide-ranging effects on cell and tissue phenotype, impacting proliferation, adhesion, pericellular matrix composition, and elastogenesis. The G1 domain of versican, which contains two Link modules that bind to hyaluronan (HA), may be central to these effects. Recombinant human G1 (rhG1) with an N-terminal 8 amino acid histidine (His) tag, produced in Nicotiana benthamiana, was applied to cultures of dermal fibroblasts, and effects on proliferation and pericellular HA organization determined. rhG1 located to individual strands of cell surface HA which aggregated into structures resembling HA cables. On both individual and aggregated strands, the spacing of attached rhG1 was similar (~120 nm), suggesting interaction between rhG1 molecules. Endogenous V0/V1, present on HA between attached rhG1, did not prevent cable formation, while treatment with V0/V1 alone, which also bound to HA, did not induce cables. A single treatment with rhG1 suppressed cell proliferation for an extended period. Treating cells for 4 weeks with rhG1 resulted in condensed layers of elongated, differentiated α actin-positive fibroblasts, with rhG1 localized to cell surfaces, and a compact extracellular matrix including both collagen and elastin. These results demonstrate that the G1 domain of versican can regulate the organization of pericellular HA and affect phenotype

    Chromatin Profiles of Chromosomally Integrated Human Herpesvirus-6A

    Human herpesvirus-6A (HHV-6A) and 6B (HHV-6B) are two closely related betaherpesviruses that are associated with various diseases including seizures and encephalitis. The HHV-6A/B genomes have been shown to be present in an integrated state in the telomeres of latently infected cells. In addition, integration of HHV-6A/B in germ cells has resulted in individuals harboring this inherited chromosomally integrated HHV-6A/B (iciHHV-6) in every cell of their body. Until now, the viral transcriptome and the epigenetic modifications that contribute to the silencing of the integrated virus genome remain elusive. In the current study, we used a patient-derived iciHHV-6A cell line to assess the global viral gene expression profile by RNA-seq, and the chromatin profiles by MNase-seq and ChIP-seq analyses. In addition, we investigated an in vitro generated cell line (293-HHV-6A) that expresses GFP upon the addition of agents commonly used to induce herpesvirus reactivation such as TPA. No viral gene expression including miRNAs was detected from the HHV-6A genomes, indicating that the integrated virus is transcriptionally silent. Intriguingly, upon stimulation of the 293-HHV-6A cell line with TPA, only foreign promoters in the virus genome were activated, while all HHV-6A promoters remained completely silenced. The transcriptional silencing of latent HHV-6A was further supported by MNase-seq results, which demonstrate that the latent viral genome resides in a highly condensed nucleosome-associated state. We further explored the enrichment profiles of histone modifications via ChIP-seq analysis. Our results indicated that the HHV-6 genome is modestly enriched with the repressive histone marks H3K9me3/H3K27me3 and does not possess the active histone modifications H3K27ac/H3K4me3. Overall, these results indicate that HHV-6 genomes reside in a condensed chromatin state, providing insight into the epigenetic mechanisms associated with the silencing of the integrated HHV-6A genome

    Information architecture for effective Workload Control: an insight from a successful implementation

    The implementation of Workload Control (WLC), a Production Planning and Control concept uniquely designed for Make-To-Order companies, has been a constant challenge. Scholars argued that WLC is largely developed through simulations of well-defined environments while much more complex circumstances (e.g. information availability) have emerged in field research. A recent trend of WLC research is to improve the practical applicability of the concept, where empirical evidence is essential. However, success in WLC implementation remains impeded. The availability of data has been a significant area that frustrates the implementation process. While there is a tendency to simplify data requirements in recent WLC theory development, it is important to understand and maintain the information that is essential for the concept to be effective. For the first time in the field, this paper details the information architecture for WLC. Key informational entities of relevance to the input/output control functions in WLC as well as performance measurement are discussed based on evidence from a successful implementation. The paper not only sheds light for practitioners on how to construct an information system that facilitates successful WLC implementation but also has implications for future development of WLC mechanisms coping with information uncertainties in practice
