    Growing islands of interest: nurturing the development of young children’s working theories

    This presentation draws on the work from a 2-year collaborative practitioner research project, Moments of wonder, every day events: how are young children theorising and making sense of their world. The project aimed to contribute perspectives to the discussion around the ways young children express and develop working theories, how practitioners understand these and how best to respond to this learning in five Playcentres (parent-led early childhood education settings) in Canterbury, New Zealand. Children’s working theories, as described in Te Whāriki (the New Zealand early childhood education curriculum), are derived from Claxton’s view that knowledge consists of a large number of purpose-built situation specific packages called ‘mini theories’, and that ‘learning involved a gradual process of editing these mini theories so that they come to contain better knowledge and skill and be better located with respect to the area of experience for which they are suitable’. When children are engaged with others in complex thinking they are forming and strengthening their working theories. In exploring working theories we recognise that children have many interests. Some of these are fleeting, while others are more connected or revisited more frequently by children. Over the course of our research, we have come to think of these interests as ‘islands’ and in doing so have adopted this as a metaphor for working theories. We were keen to see how we can grow some of these islands of interest: making them more complex, more connected, and more compelling to children. The research team explored the different ways opportunities can be created for children to express and develop working theories and the outcomes for children’s learning as a result. The presentation will focus on some of the strategies implemented and the ways these have contributed to children’s ‘working theories’ learning as the practitioner researchers attempted to build communities of thinkers and ‘wonderers’

    Macronuclear persistence of sequences normally eliminated during development in Tetrahymena thermophila

    During conjugation in the ciliated protozoan, Tetrahymena thermophila , a somatic MAC-ronucleus develops from the germinal MICronucleus. Ten to 20 percent of the MIC genome is eliminated during this process. Three repetitive families have been identified which have different levels of repetition in the MIC and are eliminated to different degrees in the MAC. Some members of two of these families persist in the MAC. In this study, we have looked at these persistent sequences in the MAC of cell lines from a variety of sources including several inbed strains, two sets of caryonides, caryonidal subclones, and vegetatively aged cell clones. The results suggest that the sequences that remain in the MAC have a genetic predisposition to persist. However, epigenetic variations occur as the MAC develops so that only some of the persistent sequences are actually observed in a particular MAC. Polymorphisms may be generated if alternative processing of a single MIC segment occurs. These polymorphisms can later be resolved by phenotypic assortment during vegetative growth.     Systematic review of patients’ views on the quality of primary health care in sub-Saharan Africa

    This is the first systematic review of patient views on the quality of primary health care services in sub-Saharan Africa using studies identified from MEDLINE, CINAHL Plus, EMBASE and PsycINFO. In total, 20 studies (3 qualitative, 3 mixed method and 14 quantitative) were included. Meta-analysis was done using quantitative findings from facility- and community-based studies of patient evaluation of primary health care. There was low use of validated measures, and the most common scales assessed were humanness (70%) and access (70%). While 66% (standard deviation = 21%) of respondents gave favourable feedback, there were discrepancies between surveys in community and facility contexts. Findings suggest that patient views could vary with subject recruitment site. We recommend improvement in the methods used to examine patient views on quality of primary health care

    Highly Purified Micro- and Macronuclei from Tetrahymena thermophila Isolated by Percoll Gradients 1

    A new procedure is described that utilizes Percoll gradients for purifying micronuclei (MIC) and macronuclei (MAC) from Tetrahymena thermophila . Separation of MIC from MAC during certrifugation in Percoll gradients occurs as a result of their difference in size rather than density. Three kinds of tests were used to evaluate the purity of the nuclei: visualization of the nuclei by light microscopy; examination of the nuclei by electron microscopy; and Southern blots of MIC and MAC DNA probed with the 5s rRNA genes or a fragment from the MAC extrachromosomal rDNA molecule. When examined under the light microscope, the isolated MIC and MAC have much lower nuclear cross contamination levels than previous methods have reported. MIC's contaminated with less than 1 MAC in 1000 MIC and MAC's contaminated with less than 1 MIC in 500 MAC can be routinely prepared. Quantitative analyses of electron micrographs gave higher estimates of cross contamination in our purified nuclei, which may, in part, be explained by the difficulty in identifying small MIC or MAC fragments. Southern blots of MIC and MAC DNA probed with 5s rDNA confirmed the level of MAC contamination in the MIC estimated by light microscopy during purification of the nuclei. The level of nucleolar contamination in the MIC was estimated at 10% by Southern blots of MIC and MAC DNA, derived from a heterokaryon with distinctive MIC and MAC Bam HI sites, using an rDNA probe.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/75629/1/j.1550-7408.1983.tb01027.x.pd
