    Opioid Use Disorder in the Active Service: Incidence Rates and Behavioral Health Considerations

    Military service members experience occupational specific injuries that often result in chronic pain, and comorbid behavioral health concerns that may be exacerbated by opioid use. Despite the vast amount of research examining substance use in the military, there is a dearth of literature examining the incidence of opioid use disorder (OUD). The present epidemiological study aims to examine the incident rate trends of OUD diagnoses among active duty service members between 2016 and 2021. The present study utilized data drawn from the Defense Medical Epidemiology Database (DMED) to conduct a retrospective cohort study and calculate the incidence rate of OUD (per 10,000) active duty United States service members. The average calculated incidence rate of OUD diagnoses from 2016 to 2021 was 6.71 (per 10,000) and declined by 34% each year on average. Opioid use disorder was most frequently diagnosed among service members who identified as male, married, white, between the ages of 30-39, junior enlisted, and serving in the Army. The incidence rate trends of OUD diagnoses among service members warrants further investigation into the mechanisms contributing to increased risk among identified groups and the development of mitigation strategies that can contribute to a reduction in opioid use and abuse

    Purine biosynthesis in archaea: variations on a theme

    <p>Abstract</p> <p>Background</p> <p>The ability to perform <it>de novo </it>biosynthesis of purines is present in organisms in all three domains of life, reflecting the essentiality of these molecules to life. Although the pathway is quite similar in eukaryotes and bacteria, the archaeal pathway is more variable. A careful manual curation of genes in this pathway demonstrates the value of manual curation in archaea, even in pathways that have been well-studied in other domains.</p> <p>Results</p> <p>We searched the Integrated Microbial Genome system (IMG) for the 17 distinct genes involved in the 11 steps of <it>de novo </it>purine biosynthesis in 65 sequenced archaea, finding 738 predicted proteins with sequence similarity to known purine biosynthesis enzymes. Each sequence was manually inspected for the presence of active site residues and other residues known or suspected to be required for function.</p> <p>Many apparently purine-biosynthesizing archaea lack evidence for a single enzyme, either glycinamide ribonucleotide formyltransferase or inosine monophosphate cyclohydrolase, suggesting that there are at least two more gene variants in the purine biosynthetic pathway to discover. Variations in domain arrangement of formylglycinamidine ribonucleotide synthetase and substantial problems in aminoimidazole carboxamide ribonucleotide formyltransferase and inosine monophosphate cyclohydrolase assignments were also identified.</p> <p>Manual curation revealed some overly specific annotations in the IMG gene product name, with predicted proteins without essential active site residues assigned product names implying enzymatic activity (21 proteins, 2.8% of proteins inspected) or Enzyme Commission (E. C.) numbers (57 proteins, 7.7%). There were also 57 proteins (7.7%) assigned overly generic names and 78 proteins (10.6%) without E.C. numbers as part of the assigned name when a specific enzyme name and E. C. number were well-justified.</p> <p>Conclusions</p> <p>The patchy distribution of purine biosynthetic genes in archaea is consistent with a pathway that has been shaped by horizontal gene transfer, duplication, and gene loss.     Radio observations of the planetary nebula around the OH/IR Star OH354.88-0.54 (V1018 Sco)

    We present radio observations of the unique, recently formed, planetary nebula (PN) associated with a very long-period OH/IR variable star V1018 Sco that is unequivocally still in its asymptoticgiant branch phase. Two regions within the optical nebula are clearly detected in nonthermal radio continuum emission, with radio spectral indices comparable to those seen in colliding-wind Wolf-Rayet binaries. We suggest that these represent shocked interactions between the hot, fast stellar wind and the cold nebular shell that represents the PN's slow wind moving away from the central star. This same interface produces both synchrotron radio continuum and the optical PN emission. The fast wind is neither spherical in geometry nor aligned withany obvious optical or radio axis.     Understanding the Effects of Climate Change via Disturbance on Pristine Arctic Lakes—Multitrophic Level Response and Recovery to a 12-Yr, Low-Level Fertilization Experiment

    Effects of climate change-driven disturbance on lake ecosystems can be subtle; indirect effects include increased nutrient loading that could impact ecosystem function. We designed a low-level fertilization experiment to mimic persistent, climate change-driven disturbances (deeper thaw, greater weathering, or thermokarst failure) delivering nutrients to arctic lakes. We measured responses of pelagic trophic levels over 12 yr in a fertilized deep lake with fish and a shallow fishless lake, compared to paired reference lakes, and monitored recovery for 6 yr. Relative to prefertilization in the deep lake, we observed a maximum pelagic response in chl a (+201%), dissolved oxygen (DO, −43%), and zooplankton biomass (+88%) during the fertilization period (2001–2012). Other responses to fertilization, such as water transparency and fish relative abundance, were delayed, but both ultimately declined. Phyto- and zooplankton biomass and community composition shifted with fertilization. The effects of fertilization were less pronounced in the paired shallow lakes, because of a natural thermokarst failure likely impacting the reference lake. In the deep lake there was (a) moderate resistance to change in ecosystem functions at all trophic levels, (b) eventual responses were often nonlinear, and (c) postfertilization recovery (return) times were most rapid at the base of the food web (2–4 yr) while higher trophic levels failed to recover after 6 yr. The timing and magnitude of responses to fertilization in these arctic lakes were similar to responses in other lakes, suggesting indirect effects of climate change that modify nutrient inputs may affect many lakes in the future
