    A method for using system identification techniques to improve airframe finite element models was developed and demonstrated. The method uses linear sensitivity matrices to relate changes in selected physical parameters to changes in total system matrices. The values for these physical parameters were determined using constrained optimization with singular value decomposition. The method was confirmed using both simple and complex finite element models for which pseudo-experimental data was synthesized directly from the finite element model. The method was then applied to a real airframe model which incorporated all the complexities and details of a large finite element model and for which extensive test data was available. The method was shown to work, and the differences between the identified model and the measured results were considered satisfactory

    A motivational music and video intervention improves high-intensity exercise performance.

    UNLABELLED Music and video are utilised by recreational gym users to enhance their exercise experience. Music and video have not been investigated for their combined ergogenic effect during high intensity exercise. To induce fatigue, this study was performed in warm (~26°C), moist conditions (~50%RH). Six, non-acclimated, male participants took part in the study. Each participant completed three 30-minute exercise bouts on a motorised treadmill under three counterbalanced conditions on separate days: control (CON), motivational music plus video intervention (M), non-motivational intervention (NM). They completed a warm-up (5 km·h(-1) [5 minutes], 9km·h(-1) [10 minutes]) followed by a maximal effort run (15 minutes). Participants did not receive any feedback of time elapsed, distance run or speed. MEASURES Distance covered (metres), heart rate, blood lactate accumulation (B(lac)) and ratings of perceived exertion (RPE). Participants in the M condition ran significantly further than in the NM (M: 3524 [388]metres; NM: 3110 [561]metres; CON: 3273 [458]metres) and CON conditions, accumulated more B(lac), but did not increase their peak RPE rating (p < 0.05). The M intervention improved tolerance of high intensity exercise in warm conditions. It was proposed that a change in attentional processing from internal (physical sensations) to external perspective (music and video) may have facilitated this improvement. These findings have strong implications for improving health, fitness and engagement in gym-based exercise programs.     Direct determination of urinary creatinine by reactive-thermal desorption-extractive electrospray-ion mobility-tandem mass spectrometry.

    A direct, ambient ionization method has been developed for the determination of creatinine in urine that combines derivatization and thermal desorption with extractive electrospray ionization and ion mobility-mass spectrometry. The volatility of creatinine was enhanced by a rapid on-probe aqueous acylation reaction, using a custom-made thermal desorption probe, allowing thermal desorption and ionization of the monoacylated derivative. The monoacyl creatinine [M + H] ion (m/z 156) was subjected to mass-to-charge selection and collision induced dissociation to remove the acyl group, generating the protonated creatinine [M + H] product ion at m/z 114 before an ion mobility separation was applied to reduce chemical noise. Stable isotope dilution using creatinine-d as internal standard was used for quantitative measurements. The direct on-probe derivatization allows high sample throughput with a typical cycle time of 1 min per sample. The method shows good linearity (R = 0.986) and repeatability (%RSD 8-10%) in the range of 0.25-2.0 mg/mL. The creatinine concentrations in diluted urine samples from a healthy individual were determined to contain a mean concentration of 1.44 mg/mL creatinine with a precision (%RSD) of 9.9%. The reactive ambient ionization approach demonstrated here has potential for the determination of involatile analytes in urine and other biofluids. © 2013 American Chemical Society

    Determining the Relationship Between Seizure-Free Days and Other Predictors of Quality of Life in Patients with Dravet Syndrome and Their Carers from FFA Registration Studies

    INTRODUCTION Dravet syndrome (DS) is a rare, lifelong epileptic encephalopathy characterised by frequent and severe seizures associated with premature mortality. Typically diagnosed in infancy, patients also experience progressive behavioural, motor-function and cognitive decline. Twenty percent of patients do not reach adulthood. Quality of life (QoL) is impaired for both patients and their carers. Reducing convulsive seizure frequency, increasing convulsive seizure-free days (SFDs) and improving patient/carer QoL are primary treatment goals in DS. This study explored the relationship between SFDs and patients' and carers' QoL to inform a cost-utility analysis of fenfluramine (FFA). METHODS In FFA registration studies, patients (or their carer proxies) completed the Paediatric QoL inventory (PedsQL). These data were mapped to EuroQol-5 Dimensions Youth version (EQ-5D-Y) to provide patient utilities. Carer utilities were collected using EQ-5D-5L and mapped to EQ-5D-3L to align patient and carer QoL on the same scale. Linear mixed-effects and panel regression models were tested and Hausman tests identified the most appropriate approach for each group. On this basis, a linear mixed-effects regression model was used to examine the relationships between patient EQ-5D-Y and clinically relevant variables (age, frequency of SFDs per 28 days, motor impairments and treatment dose). A linear panel regression model examined the relationship between SFDs and carer QoL. RESULTS After adjustment for age and underlying comorbidities, the patient regression model showed that SFDs per 28 days was a significant predictor of QoL. Each additional patient-SFD increased utility by 0.005 (p < 0.001). The carer linear panel model also showed that increasing SFDs per 28 days was a significant predictor of improved QoL. Each additional SFD increased carer utility by 0.014 (p < 0.001). CONCLUSION This regression framework highlights that SFDs are significantly correlated with both patients' and carers' QoL. Treatment with effective antiseizure medications that increase SFDs directly improves QoL for patients and their carers

    Determination of free desmosine and isodesmosine as urinary biomarkers of lung disorder by ultra performance liquid chromatography-ion mobility-mass spectrometry

    The elastin degradation products, desmosine (DES) and isodesmosine (IDES) are highly stable, cross-linking amino-acids that are unique to mature elastin. The excretion of DES/IDES in urine, in the free form and with associated peptide fragments, provides an indicator of lung damage in chronic obstructive pulmonary disease (COPD). A quantitative ion mobility-mass spectrometry (IM-MS) method has been developed for the analysis of free DES/IDES in urine with deuterated IDES as an internal standard. Resolution of DES/IDES isomers was achieved in less than five minutes using ultra performance liquid chromatography (UPLC) combined with ion pairing. The optimized UPLC-IM-MS method provided a linear dynamic range of 10-300 ng/mL and a limit of quantitation of 0.028 ng/mL for IDES and 0.03 ng/mL for DES (0.55 ng and 0.61 ng on column respectively). The method reproducibility (%RSD) was < 4% for DES and IDES. The UPLC-IM-MS method was applied to the analysis of urine samples obtained from healthy volunteers and COPD patients. The DES/IDES concentrations in healthy and COPD urine showed an increase in DES (79%) and IDES (74%) in the COPD samples, relative to healthy controls. The incorporation of an IM separation prior to m/z measurement by MS was shown to reduce non-target ion responses from the bio-fluid matrix

    This article engages with the relationship between social theory, architectural theory and material culture. The article is a reply to an article in a previous volume of the journal in question (Smith, M. (2001) ‘Repetition and difference: Lefebvre, Le Corbusier and modernity’s (im)moral landscape’, Ethics, Place and Environment, 4(1), 31-34) and, consequently, is also a direct engagement with another academic's scholarship. It represents a critique of their work as well as a recasting of their ideas, arguing that the matter in question went beyond interpretative issues to a direct critique of another author's scholarship on both Le Corbusier and Lefebvre. A reply to my article from the author of the original article was carried in a later issue of the journal (Smith, M. (2002) ‘Ethical Difference(s): a Response to Maycroft on Le Corbusier and Lefebvre’, Ethics, Place and Environment, 5(3), 260-269)
