    Anti-tumor effect of polysaccharides from rhizome of Curculigo orchioides Gaertn on cervical cancer

    Purpose: To investigate the anti-tumor effects of polysaccharides from Curculigo orchioides (PDC) on cervical cancer and the possible mechanisms involved.Methods: A Box–Behnken design (BBD) was employed to optimize extraction conditions for PDC. The anti-tumor effect of PDC on cervical cancer was investigated in vivo in mice injected with Hela cells. The parameters measured were tumor volume and weight. In vitro anti-tumor effects of PDC were assessed by measuring expressions of caspase-3, caspase-9 and P53 proteins in Hela cells via ELISA assay. Thymus and spleen indices were calculated for assessment of PDC effect on immune function.Results: The optimum extraction conditions predicted by the response surface methodology (RSM) were: extraction time = 1.58 h, ratio-of-water-to-sample = 30.05 mL/g and extraction number = 1.95. PDC showed significant anti-tumor effect on cervical cancer in mice. It significantly increased thymus and spleen indices in mice; and significantly up-regulated expressions of caspase-3, caspase-9 and P53 proteins in HeLa cells.Conclusion: PDC has significant anti-tumor effect on cervical cancer in vivo and in vitro, most probably through mechanisms involving enhancement on immune function and induction of apoptosis.Keyword: Curculigo orchioides, Polysaccharides, Cervical cancer, HeLa cells, Apoptosi

    Relevance of EGFR gene mutation with pathological features and prognosis in patients with non–small–cell lung carcinoma

    AbstractObjectiveTo study the relevance of EGFR gene mutation with pathological features and prognosis in patients with non–small–cell lung carcinoma.MethodsA total of 297 patients from July 2009 to May 2013 were chosen as objects. EGFR gene mutation were detected with fluorescence quantitative PCR. Relevance of EGFR gene mutation with clinical and pathological features was analyzed, and the prognosis of EGFR– mutant–patients and that of EGFR– wide type–patients was compared.ResultsIn 297 patients, 136 (45.79%) showed EGFR gene mutation. EGFR gene mutation had no significant relevance with age, gender, smoking history, family history of cancer and clinical stage (P>0.05); there was significant relevance between EGFR gene mutation and blood type, pathologic types, differentiation and diameter of cancer (P<0.05). The difference between prognosis of EGFR– mutant–patients and that of EGFR– wide type–patients was statistical significance (P<0.05).ConclusionsEGFR gene mutation has significant relevance with pathological features, the prognosis of EGFR–mutant–patients is better than that of EGFR– wide type–patients

    Common promoter variants of the NDUFV2 gene do not confer susceptibility to schizophrenia in Han Chinese

    <p>Abstract</p> <p>Background</p> <p>The NADH-ubiquinone oxidoreductase flavoprotein gene (<it>NDUFV2</it>), which encodes a 24 kD mitochondrial complex I subunit, has been reported to be positively associated with schizophrenia and bipolar disorder in different populations.</p> <p>Methods</p> <p>We genotyped the promoter variants of this gene (rs6506640 and rs1156044) by direct sequencing in 529 unrelated Han Chinese schizophrenia patients and 505 matched controls. Fisher's Exact test was performed to assess whether these two reported single nucleotide polymorphisms (SNPs) confer susceptibility to schizophrenia in Chinese.</p> <p>Results</p> <p>Allele, genotype and haplotype comparison between the case and control groups showed no statistical significance, suggesting no association between the <it>NDUFV2 </it>gene promoter variants and schizophrenia in Han Chinese.</p> <p>Conclusion</p> <p>The role of NDUFV2 played in schizophrenia needs to be further studied. Different racial background and/or population substructure might account for the inconsistent results between studies.</p
