    Anti-Helicobacter pylori activity of Plumbago zeylanica L

    It has been shown that the presence of infection by Helicobacter pylori is strongly associated with gastric cancer and peptic ulceration. In western medicine, a 3-fold therapeutic regimen, emphasizing the use of antibiotics,,is typically used to suppress H. pylori activity. However, antibiotic drug resistance frequently develops as a consequence of such treatment. In our previous study, 50 Taiwanese folk medicinal plants were screened for their anti-H. pylori activities. The results revealed that Plumbago zeylanica L. had the highest inhibitory effects against H. pylori. In this study, therefore, we have focused on establishing the anti-H. pylori activities of P. zeylanica L. Water and the organic solvents ethanol, ethyl acetate and acetone were used for P. zeylanica L. extraction, obtaining yields of 1.66-6.84% (w/w). Excluding the water extract, higher anti-H. pylori activity was demonstrated for all the extracts, both using the agar diffusion and dilution methods. The ethyl acetate extract exhibited the lowest minimum inhibitory concentrations against five H. pylori strains, of which ranged from 0.32 to 1.28 mg ml(-1), followed, in ascending order, by the acetone, ethanol and water analogs. Bactericidal activity was determined for P. zeylanica L. extracts, with the lowest minimum bactericidal concentrations (5.12-20.48 mg ml(-1)) demonstrated for the ethyl acetate, followed, in ascending order, by the acetone and ethanol analogs. Bactericidal activity appeared to be in a dose-dependent manner. Through a broad pH range (2-7), bactericidal activity was not affected when extract concentrations were greater than or equal to the minimum bactericidal concentration. High stability was demonstrated for the ethyl acetate P. zeylanica L. extract within pH range of 1-7, exhibiting all pH treatments bactericidal activity.     Time-distance analysis of the emerging active region NOAA 10790

    We investigate the emergence of Active Region NOAA 10790 by means of time – distance helioseismology. Shallow regions of increased sound speed at the location of increased magnetic activity are observed, with regions becoming deeper at the locations of sunspot pores. We also see a long-lasting region of decreased sound speed located underneath the region of the flux emergence, possibly relating to a temperature perturbation due to magnetic quenching of eddy diffusivity, or to a dense flux tube. We detect and track an object in the subsurface layers of the Sun characterised by increased sound speed which could be related to emerging magnetic-flux and thus obtain a provisional estimate of the speed of emergence of around 1 km s−1

    Predicting Consensus Structures for RNA Alignments Via Pseudo-Energy Minimization

    Thermodynamic processes with free energy parameters are often used in algorithms that solve the free energy minimization problem to predict secondary structures of single RNA sequences. While results from these algorithms are promising, an observation is that single sequence-based methods have moderate accuracy and more information is needed to improve on RNA secondary structure prediction, such as covariance scores obtained from multiple sequence alignments. We present in this paper a new approach to predicting the consensus secondary structure of a set of aligned RNA sequences via pseudo-energy minimization. Our tool, called RSpredict, takes into account sequence covariation and employs effective heuristics for accuracy improvement. RSpredict accepts, as input data, a multiple sequence alignment in FASTA or ClustalW format and outputs the consensus secondary structure of the input sequences in both the Vienna style Dot Bracket format and the Connectivity Table format. Our method was compared with some widely used tools including KNetFold, Pfold and RNAalifold. A comprehensive test on different datasets including Rfam sequence alignments and a multiple sequence alignment obtained from our study on the Drosophila X chromosome reveals that RSpredict is competitive with the existing tools on the tested datasets.     Melting behavior of ultrathin titanium nanowires

    The thermal stability and melting behavior of ultrathin titanium nanowires with multi-shell cylindrical structures are studied using molecular dynamic simulation. The melting temperatures of titanium nanowires show remarkable dependence on wire sizes and structures. For the nanowire thinner than 1.2 nm, there is no clear characteristic of first-order phase transition during the melting, implying a coexistence of solid and liquid phases due to finite size effect. An interesting structural transformation from helical multi-shell cylindrical to bulk-like rectangular is observed in the melting process of a thicker hexagonal nanowire with 1.7 nm diameter.Comment: 4 pages, 4 figure

    Organogenesis of Phaseolus angularis L.: high efficiency of adventitious shoot regeneration from etiolated seedlings in the presence of N6-benzylaminopurine and thidiazuron

    A step-wise procedure for the regeneration of fertile plants by organogenesis from cultures of the economically important Phaseolus angularis L., cultivars: KS-6, KS-7 and KS-8 using etiolated seedlings was established. Pre-culture of 5-day old seedling explants with MS (Murashige and Skoog (1962) Physiol Plant 15:473-493) + B-5-vitamins (Gamborg et al. (1968) Exp Cell Res 50:151-158) liquid medium containing either 5.0 mu M TDZ or 5.0 mu M BAP under dark condition was essential for organogenesis. Bud growth and shoot multiplication were stimulated by reducing the BAP concentrations from 5.0 to 2.5 mu M after 3 weeks. The maximum frequency of shoot induction was 65.2% (33.8 +/- 2.54 shoots/explant) in cultivar KS-8 followed by KS-7 34.6% (23.4 +/- 1.91 shoots/explant) and KS-6 30.6% (21.2 +/- 2.28 shoots/explant). The multiplied buds elongated after transferring to solid MSB5 medium supplemented with 4.0 mu M GA(3), 12.5 mu M AgNO3 and 0.4 mu M IBA. Up to 98% rooting efficiency of was obtained when the shoots were pulse-treated with liquid medium containing 4.5 mu M IBA for 10 min. The rooted plantlets were transferred to pots in the greenhouse, where they grew, mature, flowered and bared pod normally. The efficient shoot bud induction capability was found to be cultivar dependent. All the three cultivars tested formed multiple shoots. This efficient and rapid regeneration system may also be helpful for Agrobacterium- or particle gun-mediated transformation for this important legume crop

    Three-dimensional flow structure behind an Ahmed vehicle model

    Paper presented at the 9th International Conference on Heat Transfer, Fluid Mechanics and Thermodynamics, Malta, 16-18 July, 2012.The lasting high fuel cost has recently inspired resurgence in drag reduction research for vehicles, which calls for a thorough understanding of the vehicle wake. The simplified Ahmed vehicle model is characterized by controllable flow separation, thus especially suitable for this purpose. In spite of a considerable number of previous investigations, our knowledge of flow around this model remains incomplete. This work aims to revisit turbulent flow structure behind this model. Two rear slant angles, i.e., α = 25º and 35º, of the model were examined, representing two distinct flow regimes. The Reynolds number was 5.26×104 based on the model height (H) and incident flow velocity. Using particle image velocimetry (PIV), flow was measured with and without a gap (g/H = 0.174) between the vehicle underside and ground in three orthogonal planes, viz. the x-z, x-y and y-z planes, where x, y and z are the coordinates along longitudinal, transverse and spanwise directions, respectively. The flow at g/H = 0 serves as an important reference for the understanding of the highly complicated vehicle wake (g/H ≠ 0). While reconfirming the well documented major characteristics of the mean flow structure, both instantaneous and time-averaged PIV data unveil a number of important features of the flow structure, which have not been previously reported. As such, considerably modified flow structure models are proposed for both regimes. The time-averaged velocities, second moments of fluctuating velocities and vorticity components are presented and discussed, along with their dependence on g/H in the two distinct flow regimes.dc201

Using 5′ untranslated region reverse transcription–PCR, we detected HPeV in feces of monkeys with diarrhea and sequenced the complete genome of 1 isolate (SH6). Monkeys may serve as reservoirs for zoonotic HPeV transmissions and as models for studies of HPeV pathogenesis
