    Submerzni uzgoj i karakterizacija polisaharida iz gljive Grifola frondosa – primjena u kozmetičkoj industriji

    Grifola frondosa (maitake) is traditionally called \u27the king of mushrooms\u27 and \u27the hen of the woods\u27. Both the fruiting bodies and the mycelium of maitake have been reported to have antitumor and antiviral activities. Recently, submerged culture processes have been developed, with the intention of providing opportunities for increased economic exploitation of maitake. Commonly the aim of these processes is to produce extracellular polysaccharides (EPS), mostly glucans, and to explore their applications, particularly in the cosmetic industry. A wide variety of EPS with different molecular chain length and chemical compositions are produced under different culture conditions. In this article, various biological and physicochemical properties of the EPS of G. frondosa (GF-EPS) are described, with a view to applications in the area of functional cosmeceuticals. The GF-EPS, together with GF mycelial extract (GF-MPS), showed antioxidative activity, stimulation of collagen biosynthetic activity, cell proliferation activity, and inhibitory activity of melanogenesis, without significant cytotoxicity. These diverse functionalities suggest that both GF-EPS and GF-MPS can be promising cosmetic ingredients.Gljivu Grifola frondosa (maitake) u Koreji tradicionalno zovu kraljicom gljiva i šumskim bogatstvom. Dokazano je da plodište i micelij gljiva maitake imaju antitumorski i antivirusni učinak. Ekonomsko iskorištenje te gljive omogućeno je submerznim uzgojem. Uobičajeni je cilj uzgoja proizvodnja ekstracelularnih polisaharida (EPS), uglavnom glukana i istraživanje mogućnosti njihove primjene u kozmetičkoj industriji. Pri raznim uvjetima proizvodnje dobivaju se ekstracelularni polisaharidi različitih duljina lanaca i kemijskoga sastava. U ovom su radu opisana razna biološka i fizikalno-kemijska svojstva ekstracelularnih polisaharida gljive Grifola frondosa (GF-EPS), te primjena u proizvodnji kozmetičkih preparata s povoljnim utjecajem na ljudsko zdravlje. Ekstracelularni polisaharidi i ekstrakt micelija gljive Grifola frondosa (GFMPS) imaju antioksidativni učinak, stimuliraju biosintezu kolagena, bujanje stanica, sprečavaju nastanak melanoma i nemaju jače citotoksično djelovanje, zbog čega su prikladni za primjenu u kozmetičkoj industriji

    Genome sequence of the chromate-resistant bacterium Leucobacter salsicius type strain M1-8T

    Leucobacter salsicius M1-8(T) is a member of the Microbacteriaceae family within the class Actinomycetales. This strain is a Gram-positive, rod-shaped bacterium and was previously isolated from a Korean fermented food. Most members of the genus Leucobacter are chromate-resistant and this feature could be exploited in biotechnological applications. However, the genus Leucobacter is poorly characterized at the genome level, despite its potential importance. Thus, the present study determined the features of Leucobacter salsicius M1-8(T), as well as its genome sequence and annotation. The genome comprised 3,185,418 bp with a G+C content of 64.5%, which included 2,865 protein-coding genes and 68 RNA genes. This strain possessed two predicted genes associated with chromate resistance, which might facilitate its growth in heavy metal-rich environments.

    Long-term outcome of vertebral artery origin stenosis in patients with acute ischemic stroke

    BACKGROUND: Vertebral artery origin (VAO) stenosis is occasionally observed in patients who have acute ischemic stroke. We investigated the long-term outcomes and clinical significance of VAO stenosis in patients with acute ischemic stroke. METHODS: We performed a prospective observational study using a single stroke center registry to investigate the risk of recurrent stroke and vascular outcomes in patients with acute ischemic stroke and VAO stenosis. To relate the clinical significance of VAO stenosis to the vascular territory of the index stroke, patients were classified into an asymptomatic VAO stenosis group and a symptomatic VAO stenosis group. RESULTS: Of the 774 patients who had acute ischemic stroke, 149 (19.3%) of them had more than 50% stenosis of the VAO. During 309 patient-years of follow-up (mean, 2.3 years), there were 7 ischemic strokes, 6 hemorrhagic strokes, and 2 unknown strokes. The annual event rates were 0.97% for posterior circulation ischemic stroke, 4.86% for all stroke, and 6.80% for the composite cardiovascular outcome. The annual event rate for ischemic stroke in the posterior circulation was significantly higher in patients who had symptomatic VAO stenosis than in patients who had asymptomatic stenosis (1.88% vs. 0%, p = 0.046). In a multivariate analysis, the hazard ratio, per one point increase of the Essen Stroke Risk Score (ESRS) for the composite cardiovascular outcome, was 1.46 (95% CI, 1.02-2.08, p = 0.036). CONCLUSIONS: Long-term outcomes of more than 50% stenosis of the VAO in patients with acute ischemic stroke were generally favorable. Additionally, ESRS was a predictor for the composite cardiovascular outcome. Asymptomatic VAO stenosis may not be a specific risk factor for recurrent ischemic stroke in the posterior circulation. However, VAO stenosis may require more clinical attention as a potential source of recurrent stroke when VAO stenosis is observed in patients who have concurrent ischemic stroke in the posterior circulation

    Comb-rooted multi-channel synthesis of ultra-narrow optical frequencies of few Hz linewidth

    We report a multi-channel optical frequency synthesizer developed to generate extremely stable continuous wave lasers directly out of the optical comb of an Er-doped fiber oscillator. Being stabilized to a high-finesse cavity with a fractional frequency stability of 3.8×10153.8\times10^{-15} at 0.1 s, the comb-rooted synthesizer produces multiple optical frequencies of ultra-narrow linewidth of 1.0 Hz at 1 s concurrently with an output power of tens of mW per each channel. Diode-based stimulated emission by injection locking is a key mechanism that allows comb frequency modes to sprout up with sufficient power amplification but no loss of original comb frequency stability. Channel frequencies are individually selectable with a 0.1 GHz increment over the entire comb bandwidth spanning 4.25 THz around a 1550 nm center wavelength. A series of out-of-loop test results is discussed to demonstrate that the synthesizer is able to provide stable optical frequencies with the potential for advancing diverse ultra-precision applications such as optical clocks comparison, atomic line spectroscopy, photonic microwaves generation, and coherent optical telecommunications.Comment: 19 pages, 4 figure

    The Korean Bird Information System (KBIS) through open and public participation

    <p>Abstract</p> <p>Background</p> <p>The importance of biodiversity conservation has been increasing steadily due to its benefits to human beings. Recently, producing and managing biodiversity databases have become much easier because of the information technology (IT) advancement. This made the general public's participation in biodiversity conservation much more practical than ever. For example, an openfree web service can be devised for a wider spectrum of people to collaborate with each other for sharing biodiversity information. Bird migration is one such area of the collaboration. Korean migratory birds are usually traceable in the important routes of the East Asian-Australia Flyway (EAAF), and they play a key role as an environmental change indicator of the Earth. Therefore, the preservation of migratory birds requires an information system which involves a broader range of voluntary and interactive knowledge network to process bird information production, circulation, and dissemination.</p> <p>Results</p> <p>The Korean Bird Information System (KBIS) aims to construct a cooperative partnership domestically and internationally through the acquisition, management, and sharing of Korean bird information involving both expert and non-expert groups. KBIS has six goals: data standard, system linkage, data diversity, utilization, bird knowledge network, and statistics. The key features of KBIS are to provide a simple search, gallery (photographs), and community to lead the participation of numerous non-experts, especially amateur bird watchers. The function of real-time observation data submission through the internet has been accomplished. It also provides bird banding database, statistics, and taxon network for experts. Especially, the statistics part provides the user with easy understanding of ecological trends of species based on the time and region.</p> <p>Conclusion</p> <p>KBIS is a tool for the conservation and management of bird diversity and ecosystem that encourages users to participate by providing the openfree data access and real-time data input web-interface. It will enhance bird knowledge networking activities locally, nationally, and internationally. In addition, it provides opportunities to enhance the public awareness for the preservation of bird diversity and species information in relevant localities through the database construction and networking activities.     Enhancement of radiation response in human cervical cancer cells in vitro and in vivo by arsenic trioxide (As2O3)

    AbstractArsenic trioxide (As2O3) inhibits cell growth and induces apoptosis in certain types of cancer cells including acute promyelocytic leukemia, prostate and ovarian carcinomas, but its effect on response of tumor cells to ionizing radiation has never been explored before. Here we demonstrate that As2O3 can sensitize human cervical cancer cells to ionizing radiation both in vitro and in vivo. As2O3 in combination with ionizing radiation have a synergistic effect in decreasing clonogenic survival and in the regression of established human cervical tumor xenografts. Pretreatment of the cells with As2O3 also synergistically enhanced radiation-induced apoptosis. Apoptosis of the cells by combined treatment of As2O3 and radiation was associated with reactive oxygen species generation and loss of mitochondrial membrane potential, resulting in the activation of caspase-9 and caspase-3. The combined treatment also resulted in an increased G2/M cell cycle distribution at the concentration of As2O3 which did not alter cell cycle when applied alone. These results indicate that As2O3 can synergistically enhance radiosensitivity of human cervix carcinoma cells in vitro and in vivo, suggesting a potential clinical applicability of combination treatment of As2O3 and ionizing radiation in cancer therapies

    Functional characterization of the pathogenesis-related protein family 10 gene, PgPR10-4, from Panax ginseng in response to environmental stresses

    Pathogenesis-related proteins (PRs) are known to function in higher plants as a protein-based defensive system against abiotic and biotic stress, particularly pathogen infections. A full-length cDNA sequence of PR BetV1 was isolated and characterized from a 14-year-old ginseng expressed sequence tags library and we named this as PgPR10-4, because of similar identities with previous isolated PgPR10s sequences. The PgPR10-4 gene encodes a 477 bp open reading frame and its deduced protein contains 158 amino acids with a 53 % identity with that of the Actinidia chinensis BetV1 allergen. The expression of PgPR10-4 gene was abundant in leaves and its transcripts showed differentially up-regulated patterns against several ginseng pathogens and abiotic stimuli such as high light and salinity. In addition, PgPR10-4 expression was strongly responsive towards the stress signaling molecules H2O2 and jasmonic acid (JA), while weekly responsive to salicylic acid and abscisic acid. A functional role of PgPR10-4 in environmental stress tolerance was further validated through its overexpression in Arabidopsis. An analysis of T2 transgenic Arabidopsis plants overexpressing the PgPR10-4 gene showed an enhanced tolerance to bacterial and fungal infection, but not to salt stress. When we tagged with cyan fluorescent protein fusion protein, the PgPR10-4-was found to localize to the cytoplasm. The enhanced antifungal activity observed from the Arabidopsis transgenic lines suggests the possible involvement of PgPR10-4 in a defense-related mechanism via the JA signaling pathway
