    Impact of short-term dietary modification on postprandial oxidative stress

    <p>Abstract</p> <p>Background</p> <p>We have recently reported that short-term (21-day) dietary modification in accordance with a stringent vegan diet (i.e., a Daniel Fast) lowers blood lipids as well as biomarkers of oxidative stress. However, this work only involved measurements obtained in a fasted state. In the present study, we determined the postprandial response to a high-fat milkshake with regards to blood triglycerides (TAG), biomarkers of oxidative stress, and hemodynamic variables before and following a 21-day Daniel Fast.</p> <p>Methods</p> <p>Twenty-two subjects (10 men and 12 women; aged 35 ± 3 years) completed a 21-day Daniel Fast. To induce oxidative stress, a milkshake (fat = 0.8 g·kg<sup>-1</sup>; carbohydrate = 1.0 g·kg<sup>-1</sup>; protein = 0.25 g·kg<sup>-1</sup>) was consumed by subjects on day one and day 22 in a rested and 12-hour fasted state. Before and at 2 and 4 h after consumption of the milkshake, heart rate (HR) and blood pressure were measured. Blood samples were also collected at these times and analyzed for TAG, malondialdehyde (MDA), hydrogen peroxide (H<sub>2</sub>O<sub>2</sub>), advanced oxidation protein products (AOPP), nitrate/nitrite (NOx), and Trolox Equivalent Antioxidant Capacity (TEAC).</p> <p>Results</p> <p>A time effect was noted for HR (<it>p </it>= 0.006), with values higher at 2 hr post intake of the milkshake as compared to pre intake (<it>p </it>< 0.05). Diastolic blood pressure was lower post fast as compared to pre fast (<it>p </it>= 0.02), and a trend for lower systolic blood pressure was noted (<it>p </it>= 0.07). Time effects were noted for TAG (<it>p </it>= 0.001), MDA (<it>p </it>< 0.0001), H<sub>2</sub>O<sub>2 </sub>(<it>p </it>< 0.0001), AOPP (<it>p </it>< 0.0001), and TEAC (<it>p </it>< 0.0001); all concentrations were higher at 2 h and 4 h post intake compared to pre intake, except for TEAC, which was lower at these times (<it>p </it>< 0.05). A condition effect was noted for NOx (<it>p </it>= 0.02), which was higher post fast as compared to pre fast. No pre/post fast × time interactions were noted (<it>p </it>> 0.05), with the area under the curve from pre to post fast reduced only slightly for TAG (11%), MDA (11%), H<sub>2</sub>O<sub>2 </sub>(8%), and AOPP (12%), with a 37% increase noted for NOx.</p> <p>Conclusion</p> <p>Partaking in a 21-day Daniel Fast does not result in a statistically significant reduction in postprandial oxidative stress. It is possible that a longer time course of adherence to the Daniel Fast eating plan may be needed to observe significant findings.</p

    Life cycle human health impacts of 875 pesticides

    PurposeResidues in field crops grown and harvested for human consumption are the main contributor to overall human exposure toward agricultural pesticides for the general population. However, exposure from crop residues is currently not considered in life cycle assessment practice. We therefore present a consistent framework for characterizing human toxicological impacts associated with pesticides applied to agricultural crops in the frame of life cycle impact assessment based on state-of-the-art data and methods.MethodsWe combine a dynamic multicrop plant uptake model designed for evaluating human exposure to residues for a wide range of pesticide-crop combinations with latest findings of pesticide dissipation kinetics in crops and post-harvest food processing. Outcome is a set of intake fractions and characterization factors for 875 organic pesticides and six major food crops along with specific confidence intervals for each factor.Results and discussionIntake fractions aggregating exposure via crop residues and exposure via fractions lost to air and soil for pesticides applied to agricultural crops vary between 10−8 and 10−1 kg intake per kilogram applied as a function of pesticide and crop. Intake fractions are typically highest for lettuce and tomato and lowest for potato due to differences in application times before crop harvest and soil as additional barrier for uptake into potato tubers. Uncertainty in intake fractions is mainly associated with dissipation dynamics in crops, where results demonstrate that using pesticide- and crop-specific data is crucial. Combined with the uncertainty in effect modeling, characterization factors per pesticide and crop show squared geometric mean standard deviations ranging from 38 to 15,560 over a variability range across pesticide-crop combinations of 10 orders of magnitude.ConclusionsOur framework is operational for use in current life cycle impact assessment models, is made available for USEtox, and closes an important gap in the assessment of human exposure to pesticides. For ready use in life cycle assessment studies, we present pesticide-crop combination-specific characterization factors normalized to pesticide mass applied and provide default data for application times and loss due to post-harvest food processing. When using our data, we emphasize the need to consult current pesticide regulation, since each pesticide is registered for use on certain crops only, which varies between countries.<br/

    Adolescent Girls and Their Mothers Talk About Experiences of Binge and Loss of Control Eating

    Evidence suggests that adolescents’ experience of binge eating (BE) might differ in important ways from that of adults. Moreover, although BE appears more common in African American women than other disordered eating behaviors, little is known about the influence of cultural factors on this behavior in adolescents. The current investigation used qualitative methodology to examine the perceptions of White and African American adolescent girls and their mothers regarding experiences of binge and loss of control eating. Five focus groups were completed with 19 adolescent girls (aged 13–17, 58 % African American, 41 % White) who endorsed loss of control eating behaviors. Their mothers (N = 19) also completed separate, concurrent focus groups addressing food and eating behaviors. Responses to focus group questions were analyzed using thematic qualitative analysis. Adolescents’ awareness of their eating behaviors varied greatly. Girls reported some awareness of how emotions influence their eating behaviors, and described using food to achieve autonomy. Mothers evidenced awareness of their daughters’ problematic eating behaviors, the effects of emotions on eating for both their daughters and themselves, and sociocultural factors influencing diet. Data from these focus groups can inform the development of innovative interventions for adolescent girls engaging in loss of control eating

    Anti-angiogenic effects of resveratrol mediated by decreased VEGF and increased TSP1 expression in melanoma-endothelial cell co-culture

    Resveratrol, a naturally occurring polyphenol, has been reported to be an anti-tumor and chemopreventive agent. Recent data show that it may also exert anti-angiogenic effects. We hypothesized that the anti-angiogenic activity of resveratrol may be caused by modulation of tumor cell release of thrombospondin-1 (TSP1) and vascular endothelial growth factor (VEGF) into the extracellular matrix, leading to vascular endothelial cell (VEC) apoptosis. We therefore evaluated the effects of resveratrol on melanoma cell lines co-cultured with vascular endothelial cells in monolayer and in three dimensional spheroids. We found that resveratrol stimulated isolated VEC proliferation, while it caused growth inhibition of VECs grown with melanoma cells in three-dimensional co-culture. This effect was associated with increased melanoma cell expression of tumor suppressor protein 53 and matrix protein TSP1, as well as decreased hypoxia-driven expression of hypoxia inducible factor-1α and inhibition of VEGF production
