    RS4651 suppresses lung fibroblast activation via the TGF-β1/SMAD signalling pathway.

    ABSTRACT Background Idiopathic pulmonary fibrosis (IPF) is a progressive disease resulting in respiratory failure with no efficient treatment options. We investigated the protective effect of RS4651 on pulmonary fibrosis in mice and the mechanism. Methods Intratracheal injection of bleomycin (BLM) was used to induce pulmonary fibrosis in mice. RS4561 was administered intraperitoneally at different doses. Histopathological changes were observed. The level of alpha-smooth muscle actin (α-SMA) were also tested. In vitro, the proliferation and migratory effects of RS4651 treatment on MRC-5 cells pre-treated with transforming growth factor (TGF-β1) were examined. RNA-sequencing was used to detect differentially expressed target genes. Then, the expression of α-SMA, pSMAD2 and SMAD7 were analysed during RS4651 treatment of MRC-5 cells with or without silencing by SMAD7 siRNA. Results Histopathological staining results showed decreased collagen deposition in RS4651 administered mice. Additionally, a lower level of α-SMA was also observed compared to the BLM group. The results of in vitro studies confirmed that RS4651 can inhibit the proliferation and migration, as well as α-SMA and pSMAD2 expression in MRC-5 cells treated with TGF-β1. RNA-sequencing data identified the target gene SMAD7. We found that RS4651 could upregulate SMAD7 expression and inhibit the proliferation and migration of MRC-5 cells via SMAD7, and RS4651 inhibition of α-SMA and pSMAD2 expression was blocked in SMAD7-siRNA MRC-5 cells. In vivo studies further confirmed that RS4651 could upregulate SMAD7 expression in BLM-induced lung fibrosis in mice. Conclusions Our data suggest that RS4651 alleviates BLM-induced pulmonary fibrosis in mice by inhibiting the TGF-β1/SMAD signalling pathway

    Clean BN encapsulated 2D FETs with lithography compatible contacts

    Device passivation through ultraclean hexagonal BN encapsulation is proven one of the most effective ways for constructing high-quality devices with atomically thin semiconductors that preserves the ultraclean interface quality and intrinsic charge transport behavior. However, it remains challenging to integrate lithography compatible contact electrodes with flexible distributions and patterns. Here, we report the feasibility in straightforwardly integrating lithography defined contacts into BN encapsulated 2D FETs, giving rise to overall device quality comparable to the state-of-the-art results from the painstaking pure dry transfer processing. Electronic characterization on FETs consisting of WSe2_2 and MoS2_2 channels reveals an extremely low scanning hysteresis of ca. 2 mV on average, a low density of interfacial charged impurity of ca. 1011 10^{11}\,cm−2^{-2}, and generally high charge mobilities over 1000 1000\,cm2⋅^{2}\cdotV−1⋅^{-1}\cdots−1^{-1} at low temperatures. The overall high device qualities verify the viability in directly integrating lithography defined contacts into BN encapsulated devices to exploit their intrinsic charge transport properties for advanced electronics.Comment: 17 pages, 4 figure

    Investigation on Internal Short Circuit Identification of Lithium-Ion Battery Based on Mean-Difference Model and Recursive Least Square Algorithm

    Electric vehicles powered by lithium-ion batteries take advantages for urban transportation. However, the safety of lithium-ion battery needs to be improved. Self-induced internal short circuit of lithium-ion batteries is a serious problem which may cause battery thermal runaway. Accurate and fast identification of internal short circuit is critical, while difficult for lithium-ion battery management system. In this study, the influences of the parameters of significance test on the performance of an algorithm for internal short circuit identification are evaluated experimentally. The designed identification is based on the mean-difference model and the recursive least square algorithm. First, the identification method is presented. Then, two characteristic parameters are determined. Subsequently, the parameters of the significance calculation are optimized based on the measured data. Finally, the effectiveness of the method for the early stage internal short circuit detection is studied by an equivalent experiment. The results indicate that the detection time can be shortened significantly via a proper configuration of the parameters for the significance test

    Transcriptome profiling of the initial segment and proximal caput of mouse epididymis

    BackgroundThe proximal region of the mouse epididymis plays a pivotal role in sperm transport, sperm maturation, and male fertility. Several studies have focused on segment-dependent gene expression of the mouse epididymis through high-throughput sequencing without the precision of the microdissection.Methods and resultsHerein, we isolated the initial segment (IS) and proximal caput (P-caput) by physical microdissection using an Lcn9-cre; Rosa26tdTomato mouse model. We defined the transcriptome changes of caput epididymis by RNA sequencing (RNA-seq), which identified 1,961 genes that were abundantly expressed in the IS and 1,739 genes that were prominently expressed in the P-caput. In addition, we found that many differentially expressed genes (DEGs) were predominantly or uniquely expressed in the epididymis and region-specific genes were highly associated with transport, secretion, sperm motility, fertilization, and male fertility.ConclusionThus, this study provides an RNA-seq resource to identify region-specific genes in the caput epididymis. The epididymal-selective/specific genes are potential targets for male contraception and may provide new insights into understanding segment-specific epididymal microenvironment-mediated sperm transport, maturation, and male fertility

    Assessment of Methods for Forecasting Shale Gas Supply in China Based on Economic Considerations

    Shale gas, with its lower carbon content and pollution potential, is the most promising natural gas resource in China. When modeling the shale gas supply in a specific gas field, it is of paramount importance to determine the gas supply under economic considerations. Two common calculation methods are used in China for this purpose: Method 1 (M1) is the breakeven analysis, where the gas supply is based on the relationship between costs and revenues, while Method 2 (M2) is the Geologic Resource Supply-Demand Model, where the supply relies on demand and price scenarios. No comparisons has been made between these two methods. In this study, the Fuling shale gas field in the Sichuan Basin was chosen as a study case to forecast the shale gas supply using these two different methods. A sensitivity analysis was performed to discuss the influencing factors of each method and error measures were used to compare the different shale gas supply values calculated by each method. The results shows that M1 is more sensitive to initial production, while M2 is more sensitive to gas price. In addition, M2 is more feasible for its simplicity and accuracy at high price scenarios and M1 is considered to be reliable for low price scenarios with profit. This study can provide a quick and comprehensive assessment method for the shale gas supply in China

    Research on the Robustness of the Chinese Input–Output Network Based on Relative Entropy Theory

    The input–output (IO) network is the quantitative description of an IO-based economy in which nodes represent industries and edges connecting nodes represent the economic connection between industries. Robustness refers to the ability of tolerating perturbations that might affect the system’s functional body. There is both practical and theoretical significance to explore the robustness of the IO network for economic development. In this paper, we probe the robustness of the Chinese IO network based on the relative entropy of the probability distribution of network parameters (node degree, strongest path betweenness, downstream closeness and upstream closeness) under random node or edge failure and intentional node or edge attack. It is found that the Chinese IO network shows relatively weak robustness when it is under intentional attack, but relatively strong robustness when it is under random failure. Our experiment also verifies the applicability and effectiveness of the relative entropy model in measuring the robustness of the IO network

    Research on Nickel Material Trade Redistribution Strategy Based on the Maximum Entropy Principle

    In the double carbon background, riding the wind of new energy vehicles and the battery high nickelization, nickel resources rise along with the trend. In recent years, due to the influence of geopolitical conflicts and emergencies, as well as the speculation and control of international capital with its advantages and rules, the world may face price and security supply risks to a certain extent. Therefore, to obtain the most objective trade redistribution strategy, this paper first constructs the nickel material trade network, identifies the core trading countries and the main trade relations of nickel material trade, and finds that the flow of nickel material mainly occurred between a few countries. On this basis, a trade redistribution model is constructed based on the maximum entropy principle. Taking Indonesia, the largest exporter, and the largest trade relationship (Indonesia exports to China) as examples, the nickel material redistribution between countries when different supply risks occur are simulated. The results can provide an important reference for national resource recovery after the risk of the nickel trade

    Expanded S-Curve Model of Relationship between Domestic Water Usage and Economic Development: A Case Study of Typical Countries

    Domestic water plays a growing role with the unprecedented economic development and rising urbanization. The lack of long-term evaluation of domestic water usage trends limits our understanding of the relationship between domestic water usage and economics. Here, we present a pragmatic approach to assess the long-term relationship between domestic water usage and economics through historical data of the last 100 years from 10 typical countries to establish an evaluation method for different economics. The relationship between domestic water usage and GDP per capita was described as an expanded S-curve model and the mathematical modeling was derived to simulate this relationship for four typical countries as case studies. The simulation results show that the expanded S-curve of different countries can be calibrated with three key points: takeoff point, turning point, and zero-growth point, and four transitional sections: slow growth, accelerated growth, decelerated growth, and zero/negative growth, corresponding to the same economic development level. In addition, other factors influencing domestic water usage are also discussed in this research, including urbanization, industrial structure, and technical progress. We hope to provide a case study of an expanded S-curve as a foundation for forecasting domestic water usage in different countries or in the same economy at different developmental stages

    Factors Affecting Wind Power Efficiency: Evidence from Provincial-Level Data in China

    As a significant energy consumer, China is under tremendous pressure from the international community to address climate change issues by reducing carbon emissions; thus, the use of clean energy is imperative. Wind power is an essential source of renewable energy, and improving the efficiency of wind power generation will contribute substantially to China’s ability to achieve its energy-saving and emission reduction goals. This paper measured the wind power efficiency of 30 provinces in China from 2012 to 2017 using the data envelopment analysis (DEA) method. Moran’s I index and the spatial Durbin model were applied to analyse the spatial distribution of the wind power efficiency and the spatial effects of influencing factors. The results show obvious differences in the spatial distribution of wind power efficiency in China; specifically, the wind power efficiency in the eastern and western regions is higher than that in the central areas. Moreover, wind power efficiency has a significant positive spatial correlation between regions: the eastern and western regions show certain high-high clustering characteristics, and the central area shows certain low-low clustering characteristics. Among the influencing factors, the fixed asset investment and carbon emission intensity of the wind power property have a negative impact on the efficiency of regional wind power production, while the urbanization process and carbon emission intensity have significant spatial spillover effects. The optimization of the economic structure, technological innovation and the construction of energy infrastructure are expected to improve the regional wind power efficiency. The results present a new approach for accurately identifying the spatial characteristics of wind power efficiency and the spatial effects of the influencing factors, thus providing a reference for policymakers
