Evaluating the effects of carbon nanoreactor diameter and internal structure on the pathways of the catalytic hydrosilylation reaction
Three different types of carbon nanoreactors, double-walled nanotubes (DWNT), multi-walled nanotubes (MWNT) and graphitised carbon nanofibers (GNF) have been appraised for the first time as containers for the reactions of phenylacetylene hydrosilylation catalysed by a confined molecular catalyst [Rh4(CO)12]. Interactions of [Rh4(CO)12] with carbon nanoreactors determining the ratio of ÎČ-addition products are unchanged for all nanoreactors and are virtually unaffected by the confinement of [Rh4(CO)12] inside carbon nanostructures. Conversely, the relative concentrations of reactants affecting the ratio of addition and dehydrogenative silylation products is very sensitive to nanoscale confinement, with all nanoreactors demonstrating significant effects on the distribution of reaction products as compared to control experiments with the catalyst in bulk solution or adsorbed on the outer surface of nanoreactors. Surprisingly, the widest nanoreactors (GNF) change the reaction pathway most significantly, which is attributed to the graphitic step-edges inside GNF providing effective anchoring points for the catalyst and creating local environments with greatly altered concentrations of reactants as compared to bulk solution. Possessing diameters significantly wider than molecules, GNF impose no restrictions on the transfer of reactants while providing the strongest confinement effects for the reaction. multiâpatient dose synthesis of [18F]Flumazenil via a copperâmediated 18Fâfluorination
Background
Flumazenil (FMZ) is a functionally silent imidazobenzodiazepine which binds to the benzodiazepine binding site of approximately 75% of the brain Îł-aminobutyric acid-A receptors (GABAARs). Positron Emission Tomography (PET) imaging of the GABAARs with [11C]FMZ has been used to evidence alterations in neuronal density, to assess target engagement of novel pharmacological agents, and to study disorders such as epilepsy and Huntingtonâs disease. Despite the potential of FMZ PET imaging the short half-life (t1/2) of carbon-11 (20 min) has limited the more widespread clinical use of [11C]FMZ. The fluorine-18 (18F) isotopologue with a longer t1/2 (110 min) is ideally suited to address this drawback. However, the majority of current radiochemical methods for the synthesis of [18F]FMZ are non-trivial and low yielding. We report a robust, automated protocol that is good manufacturing practice (GMP) compatible, and yields multi-patient doses of [18F]FMZ.
Results
The fully automated synthesis was developed on the Trasis AllinOne (AIO) platform using a single-use cassette. [18F]FMZ was synthesized in a one-step procedure from [18F]fluoride, via a copper-mediated 18F-fluorination of a boronate ester precursor. Purification was performed by semi-preparative radio-HPLC and the collected fraction formulated directly into the final product vial. The overall process from start of synthesis to delivery of product is approximately 55 min. Starting with an initial activity of 23.6â±â5.8 GBq (nâ=â3) activity yields of [18F]FMZ were 8.0â±â1 GBq (nâ=â3). The synthesis was successfully reproduced at two independent sites, where the product passed quality control release criteria in line with the European Pharmacopoeia standards and ICH Q3D(R1) guidelines to be suitable for human use.
Conclusion
Reported is a fully automated cassette-based synthesis of [18F]FMZ that is Good Manufacturing Practice (GMP) compatible and produces multi-patient doses of [18F]FMZ

Evolution simulée d'une nouvelle bibliothÚque de ligands chiraux
Ce projet de recherche, réalisé au sein du laboratoire du Professeur Olivier Riant, a pour thÚme la réduction énantiosélective de cétones prochirales.
La rĂ©duction de ces cĂ©tones donne accĂšs Ă des alcools secondaires Ă©nantiomĂ©riquement enrichis d'intĂ©rĂȘt synthĂ©tique. Dans le cadre de ce projet nous restons toutefois confinĂ©s Ă la rĂ©duction de l'acĂ©tophĂ©none.
Nous dĂ©butons par une brĂšve introduction sur l'origine de la chiralitĂ© ; elle est suivie par une revue des diverses structures de ligands utilisĂ©es en rĂ©ductions asymĂ©triques. Le fil conducteur reste la rĂ©duction de cĂ©tones prochirales, mĂȘme si quelques autres exemples de substrats sont proposĂ©s. Le projet de recherche compte alors trois grandes parties :
1 La synthÚse de nouveaux ligands chiraux, constitutifs d'une 4Úme génération de ligands (type A et type B). Il s'agit en fait de participer à la fin d'un processus d'optimisation dirigé. Nous montrerons qu'il est possible d'échanger les caractéristiques des ligands entre eux pour accélérer la sélection des meilleurs candidats, en termes de réduction énantiosélective notamment.
2 La deuxiÚme partie est consacrée à la synthÚse d'une nouvelle bibliothÚque de catalyseurs chiraux dont la structure est présentée ci-dessous.
Ils sont utilisés en réduction asymétrique de l'acétophénone par transfert d'hydrure. Pour ce faire, nous engageons divers amino-alcools en réaction d'amination réductrice avec plusieurs dérivés carbonylés. Nous avons couplé les ligands amino-alcools alkylés obtenus à quelques précurseurs métalliques in situ ; ainsi se constitue notre bibliothÚque de catalyseurs. Nous montrons qu'il est possible d'appliquer un processus d' " évolution simulée " à cette bibliothÚque, et que ce processus évolutif permet de retrouver les structures les plus intéressantes de catalyseurs sans pour autant devoir tester l'ensemble de la bibliothÚque.
3 Les ligands de type A et de type B font l'objet d'une étude plus approfondie. De nombreux tests mettent en lumiÚre un phénomÚne de racémisation, lors de la réduction asymétrique de l'acétophénone. Nous proposons donc une série d'expériences afin de dégager une hypothÚse mécanistique de la racémisation. Finalement, nous dévoilons la possibilité de tirer avantage de ce phénomÚne dans le cadre de dédoublements cinétiques dynamiques.(CHIM 3)--UCL, 200
Grafting of gold complexes or clusters onto nano-carbon by covalent or non-covalent pathways using a bifunctional ligand : application in heterogeneous and supported homogeneous catalysis
Nowadays, the recycling of costly homogeneous catalysts through their immobilization as well as the incorporation of multi-metallic nanoparticles onto nano-carbon materials via a controlled manner is of prime importance. This dissertation discloses a simple methodology to immobilize by a covalent or a non-covalent strategy both homogeneous gold catalysts and mono- or multi-metallic clusters (Ru, Ru-Pt, Ru-Au, Ru-Pt-Au) onto multi-walled nanotubes (MWNTs) and graphene (GO and RGO). It involves the use of bifunctional ligands able to first interact with gold complexes or clusters in order to obtain tailored adducts ready to be immobilized in a second step onto nanocarbon. Gold supported homogeneous catalysts were tested in a series of typical gold benchmark reactions such as cyclization of enynes and showed good activities and a conceivable recycling irrespective of the methodology of immobilization (i.e covalent or non-covalent route). Once grafted onto nanocarbon, the supported clusters were thermally activated at variable temperatures to obtain multi-metallic nanoparticles of variable sizes. The selective hydrogenation of cinnamaldehyde was selected as model reaction to compare the obtained catalysts in term of activity and selectivity. The methodology of incorporation, the support, the temperature of activation, the selected metals and the solvents were found to considerably influence the selectivity of the reaction and paved the way to obtain highly selective catalysts.(SC - Sciences) -- UCL, 201
La complémentarité des thérapies individuelles et familiales dans la problématique des troubles des conduites alimentaires
Ce mĂ©moire analytique propose dâĂ©tudier la question de la thĂ©rapie conjointe dans le contexte spĂ©cifique quâest celui des troubles des conduites alimentaires. La question de recherche qui oriente lâĂ©tude est la suivante : « Quels Ă©lĂ©ments dĂ©clencheurs peuvent amener Ă une thĂ©rapie familiale lors dâune thĂ©rapie individuelle et inversement, dans la problĂ©matique des troubles des conduites alimentaires, et plus prĂ©cisĂ©ment dans les cas de suivis de troubles dâanorexie ? ». Pour la bonne organisation de ce travail, la premiĂšre partie consiste en un volet thĂ©orique mettant en lumiĂšre lâĂ©tat actuel des recherches menĂ©es sur le sujet par lâexamen du panorama de la littĂ©rature scientifique contemporaine. Plusieurs hypothĂšses formulĂ©es permettront de mettre en place un questionnaire qui sera utilisĂ© lors du volet pratique de ce travail. La derniĂšre partie du travail consistera en une analyse qualitative et transversale des diffĂ©rents entretiens, qui seront ensuite confrontĂ©s Ă la littĂ©rature prĂ©sentĂ©e. La conclusion de ce mĂ©moire permet dâobserver certaines similitudes entre la pratique thĂ©rapeutique relatĂ©e durant les rencontres et la thĂ©orie prĂ©sentĂ©e dans le premier volet.
