    Understanding the Differences Between Neuronal Calcium Sensor Proteins: A Comparison of Neurocalcin Delta and Hippocalcin

    Many neuronal functions, including learning and memory are driven by changes in intracellular Ca2+concentrations. The Neuronal Calcium Sensor (NCS) family of proteins is responsible for mediating the response to calcium. They are typically comprised of 4 EF hands; of which EF 2, 3, and 4 bind calcium. Hypothesis: NCS proteins carry out unique, non-overlapping functions, and that specific characteristics of the family can be mapped to precise regions of the proteins. Results: The effect on the following properties were investigated primarily on two highly similar NCS proteins, Neurocalcin Delta (NCALD) and Hippocalcin (HPCA): (1) Response to calcium was determined through two independent methods: change in tryptophan fluorescence and migration on a native polyacrylamide gel (CIMSA); (2) size exclusion chromatography, (3) Calcium binding, (4) Guanylate Cyclase stimulations and (5) sequence analysis. These results lead to the conclusion that specific aa locations are hyper-variable and pivotal in determining how the proteins respond

    Ticks in the box: Argas persicus occurrence in nest boxes of secondary cavity-nesting bird species in Italy

    Migratory organisms can be vectors of parasitic host organisms that can then be transported along seasonal migratory journeys and spread across continents. Ornithophilic nidicolous ticks (e.g. soft ticks) include species responsible for the transmission of pathogens and bacteria, thus representing a health problem not only for wild species that are directly parasitized, but also for those that share the same environments or reproductive sites with them. In this regard, artificial nests for birds may turn out to be site-sources of parasites. Here, we document the occurrence of different life stages of Argas persicus ticks in nest boxes of wild birds in a natural area (not associated with poultry activities) of central coastal Italy (Maremma Regional Park, Tuscany). Between 2018 and 2022, 168 ticks were collected from nest boxes occupied by different secondary cavity-nesting birds, such as European rollers Coracias garrulus and scops owl Otus scops. Ticks were analysed morphologically, and selected specimens were also identified by mitochondrial ribosomal 16S (16S) subunit gene to ascertain their taxonomic status. All ticks were identified as Argas persicus. This finding not only suggests that this tick species has formed a viable population in this Italian region, but also further confirms the previously doubtful natural origin of the species at country level and sheds new light on its underestimated and little investigated distribution. Possible pathways of introduction and its potential impacts on local avian community are discussed

    Interactive design of dental implant placements through CAD-CAM technologies: from 3D imaging to additive manufacturing

    In the field of oral rehabilitation, the combined use of 3D imaging technologies and computer-guided approaches allows the development of reliable tools to be used in preoperative assessment of implant placement. In particular, the accurate transfer of the virtual planning into the operative field through surgical guides represents the main challenge of modern dental implantology. Guided implant positioning allows surgical and prosthetic approaches with minimal trauma by reducing treatment time and decreasing patient’s discomfort. This paper aims at defining a CAD/CAM framework for the accurate planning of flapless dental implant surgery. The system embraces three major applications: (1) freeform modelling, including 3D tissue reconstruction and 2D/3D anatomy visualization, (2) computer-aided surgical planning and customised template modelling, (3) additive manufacturing of guided surgery template. The tissue modelling approach is based on the integration of two maxillofacial imaging techniques: tomographic scanning and surface optical scanning. A 3D virtual maxillofacial model is created by matching radiographic data, captured by a CBCT scanner, and surface anatomical data, acquired by a structured light scanner. The pre-surgical planning process is carried out and controlled within the CAD application by referring to the integrated anatomical model. A surgical guide is then created by solid modelling and manufactured by additive techniques. Two different clinical cases have been approached by inserting 11 different implants. CAD-based planned fixture placements have been transferred into the clinical field by customised surgical guides, made of a biocompatible resin and equipped with drilling sleeves

    Prevalence of Vaccine Type Infections in Vaccinated and Non-Vaccinated Young Women: HPV-IMPACT, a Self-Sampling Study.

    Background: The human papillomavirus (HPV) vaccination program for young girls aged 11⁻26 years was introduced in Switzerland in 2008. The objective of this study was to evaluate the prevalence of high- and low-risk HPV in a population of undergraduate students using self-sampling for monitoring the HPV vaccination program's effect. Undergraduate women aged between 18⁻31 years, attending the Medical School and University of Applied Sciences in Geneva, were invited to participate in the study. Included women were asked to perform vaginal self-sampling for HPV testing using a dry cotton swab. A total of 409 students participated in the study-aged 18⁻31 years-of which 69% of the participants were vaccinated with Gardasil HPV vaccine and 31% did not received the vaccine. About HPV prevalence, 7.2% of unvaccinated women were HPV 16 or 18 positive, while 1.1% of vaccinated women were infected by HPV 16 or 18 (p < 0.01). Prevalence of HPV 6 and 11 was 8.3% in non-vaccinated women versus 2.1% in vaccinated women (p < 0.02). We observed no cross-protection for the other HPV genotypes of a low- and high-risk strain. Prevalence of HPV 6/11/16/18 was lower in vaccinated women versus unvaccinated women. Continued assessment of HPV vaccine effectiveness in real population is needed

    Preliminary Report on the Efficacy of Music Therapy to Optimize the Compliance of Rett Syndrome Patients Attending Oral Hygiene Procedures

    Background: Individuals with intellectual developmental disability (IDD) often face barriers in accessing dental care. The main aim of the present study was to test the feasibility and the outcome of oral hygiene (OH) procedures in Rett syndrome (RS) patients, and a secondary aim was to test the utility of music therapy during dental hygiene procedures. Methods: A case group (Group 1) of female patients with RS (stage 3 of disease, age > 5 years and <18 years) and a control group of age-matched healthy females (Group 0) were formed, and subgroups received the intervention of oral hygiene combined with a music therapy (Subgroup 1M and Subgroup 0M) or standard oral hygiene (subgroup 1W and subgroup 0W). The Simplified Oral Hygiene Index (OHI-S) was used to assess the effectiveness of oral hygiene measures. Results: The OHI-S at T0 was 1.41 in Group 1 and 2.58 in Group 0 (p value < 0.001). The mean duration of the oral hygiene treatment was 7018′′ (SD 3272′′) in Group 1 and 873′′ (SD 161′′) in Group 0 (p value < 0.001). The mean duration of OH was significantly higher in Subgroup 1M and Subgroup 0M. Discussion: Based on the results of our case-control study, OH procedures can be successfully performed in RS patients with good compliance and efficacy. In addition, our study provides preliminary evidence to suggest that music therapy may be a useful adjunct, as it appears to improve patient cooperation

    First record of allonursing in golden jackal (Canis aureus, L. 1758): a case of double breeding and communal denning within the same social unit

    In the last decade, there has been a remarkable increase in the number of scientific publications on the ecology of the golden jackal Canis aureus, whereas information on its reproductive biology is still scanty, and the basic reproductive parameters of the species are not yet fully understood. In 2021, we began a research project in North-Eastern Italy, where the golden jackal is strictly protected under the National Law, to investigate the species behavioural ecology and reproductive biology at the den site, as a key element for defining conservation measures. Within this project, we documented allosuckling behaviour, possibly related to the breeding of two females within the same social unit, with den sharing and communal nursing of the cubs. Our observations open new scenarios and raise questions on the reproductive biology and social organisation of the golden jackal, stimulating further topics for research

Hydrogen has proved to be the most promising molecule, as a fuel, due to its low environmental impact. Even if various methods already exist for producing hydrogen, most of them are not sustainable. Thus, research focuses on the biological sector, studying microalgae, and other microorganisms’ ability to produce this precious molecule in a natural way. In this review, we provide a description of the biochemical and molecular processes for the production of biohydrogen and give a general overview of one of the most interesting technologies in which hydrogen finds application for electricity production: fuel cells

    Morphological and molecular characterization of trichuris sp. (nematoda: Trichuridae) in crested porcupines (hystrix cristata; rodentia: Hystricidae) from Italy

    Adult specimens of Trichuris sp. collected from crested porcupines (Hystrix cristata) from Italy were characterized using an integrative taxonomic approach involving morphological and molecular tools. The morphological features of this Trichuris sp. were compared to data already available for Trichuris spp. from Hystrix sp., revealing diagnostic traits, such as spicule length in males or vulva shape in females, which distinguish this Trichuris sp. from the other species. Evidence from sequences analysis of the partial mitochondrial COX1 region indicated that the taxon under study is a distinct lineage. Biometrical and genetic data suggested this Trichuris sp. to be a valid and separated taxon. However, since molecular data from other Trichuris spp. infecting Hystrix, such as T. infundibulus, T. hystricis, T. javanica, T. landak and T. lenkorani, are missing in public repositories, the number and identity of distinct lineages able to infect porcupines remain only partially defined

    Long-Term Safety of Anti-TNF Adalimumab in HBc Antibody-Positive Psoriatic Arthritis Patients: A Retrospective Case Series of 8 Patients

    Immunosuppressive drugs commonly used in the treatment of psoriatic arthritis make patients more susceptible to viral, bacterial, and fungal infections because of their mechanism of action. They not only increase the risk of new infections but also act altering the natural course of preexisting infections. While numerous data regarding the reactivation of tuberculosis infection are available in the literature, poor information about the risk of reactivation or exacerbation of hepatitis viruses B and C infections during treatment with biologics has been reported. Furthermore, reported series with biological therapy included short periods of followup, and therefore, they are not adequate to verify the risk of reactivation in the long-term treatment. Our study evaluated patients with a history of hepatitis B and psoriatic arthritis treated with adalimumab and monitored up to six years. During the observation period, treatment was effective and well tolerated in all patients, and liver function tests and viral load levels remained unchanged
