    Challenges of modeling rainfall triggered landslides in a data-sparse region: A case study from the Western Ghats, India

    Accurate rainfall estimates are required to forecast the spatio-temporal distribution of rain-triggered landslides. In this study, a comparison between rain gauge and satellite rainfall data for assessing landslide distribution in a data-sparse region, the mountainous district of Idukki, along the Western Ghats of southwestern India, is carried out. Global Precipitation Mission Integrated Multi-satellitE Retrievals for GPM-Late (GPM IMERG-L) rainfall products were compared with rain gauge measurements, and it was found that the satellite rainfall observations were underpredicting the actual rainfall. A conditional merging algorithm was applied to develop a product that combines the accuracy of rain gauges and the spatial variability of satellite precipitation data. Correlation Coefficient (CC) and Root Mean Squared Error (RMSE) were used to check the performance of the conditional merging process. An example from a station with the least favorable statistics shows the CC increasing from 0.589 to 0.974 and the RMSE decreasing from 65.22 to 20.01. A case scenario was considered that evaluated the performance of a landslide prediction model by relying solely on a sparse rain gauge network. Rainfall thresholds computed from both the conditionally merged GPM IMERG-L and the rain gauge data were compared and the differences indicated that relying solely on a discrete, sparse rain gauge network would create false predictions. A total of 18.7% of landslide predictions only were identified as true positives, while 60.7% was the overall false-negative rate, and the remaining were false-positives. This pointed towards the need of having a continuous data that is both accurate in measurement and efficient in capturing spatial variability of rainfall

    Mycobiome of the Bat White Nose Syndrome (WNS) Affected Caves and Mines reveals High Diversity of Fungi and Local Adaptation by the Fungal Pathogen Pseudogymnoascus (Geomyces) destructans

    The investigations of the bat White Nose Syndrome (WNS) have yet to provide answers as to how the causative fungus Pseudogymnoascus (Geomyces) destructans (Pd) first appeared in the Northeast and how a single clone has spread rapidly in the US and Canada. We aimed to catalogue Pd and all other fungi (mycobiome) by the culture-dependent (CD) and culture-independent (CI) methods in four Mines and two Caves from the epicenter of WNS zoonotic. Six hundred sixty-five fungal isolates were obtained by CD method including the live recovery of Pd. Seven hundred three nucleotide sequences that met the definition of operational taxonomic units (OTUs) were recovered by CI methods. Most OTUs belonged to unidentified clones deposited in the databases as environmental nucleic acid sequences (ENAS). The core mycobiome of WNS affected sites comprised of 46 species of fungi from 31 genera recovered in culture, and 17 fungal genera and 31 ENAS identified from clone libraries. Fungi such as Arthroderma spp., Geomyces spp., Kernia spp., Mortierella spp., Penicillium spp., and Verticillium spp. were predominant in culture while Ganoderma spp., Geomyces spp., Mortierella spp., Penicillium spp. and Trichosporon spp. were abundant is clone libraries. Alpha diversity analyses from CI data revealed that fungal community structure was highly diverse. However, the true species diversity remains undetermined due to under sampling. The frequent recovery of Pd indicated that the pathogen has adapted to WNS-afflicted habitats. Further, this study supports the hypothesis that Pd is an introduced species. These findings underscore the need for integrated WNS control measures that target both bats and the fungal pathogen.Comment: 59 pages, 7figure

    Satellite-based assessment of the August 2018 flood in parts of Kerala, India

    From 1 June to 29 August 2018, Kerala, a state in southwestern India, recorded 36% excess rainfall than normal levels, leading to widespread floods and landslides events and resulting in 445 deaths. In this study, satellite-based data were used to map the flood inundation in the districts of Thrissur, Ernakulam, Alappuzha, Idukki and Kottayam. Specifically, flood delineation was enabled with Sentinel-1A radar data of 21 August 2018 and was compared with an average pre-flood, water-cover map based on Modified Normalized Difference Water Index (MNDWI) that was developed using a January and February 2018 Sentinel-2A dataset. A 90% increase in water cover was observed during the August 2018 flood event. Low lying areas in the coastal plains of Kuttanad and the Kole lands of Thrissur, had marked a rise of up to 5 and 10 m of water, respectively, during this deluge. These estimates are conservative as that the flood waters had started receding prior to the August 21 Sentinel-1A imagery

    Revealing natural relationships among arbuscular mycorrhizal fungi: culture line BEG47 represents Diversispora epigaea, not Glomus versiforme

    Background: Understanding the mechanisms underlying biological phenomena, such as evolutionarily conservative trait inheritance, is predicated on knowledge of the natural relationships among organisms. However, despite their enormous ecological significance, many of the ubiquitous soil inhabiting and plant symbiotic arbuscular mycorrhizal fungi (AMF, phylum Glomeromycota) are incorrectly classified. Methodology/Principal Findings: Here, we focused on a frequently used model AMF registered as culture BEG47. This fungus is a descendent of the ex-type culture-lineage of Glomus epigaeum, which in 1983 was synonymised with Glomus versiforme. It has since then been used as ‘G. versiforme BEG47’. We show by morphological comparisons, based on type material, collected 1860–61, of G. versiforme and on type material and living ex-type cultures of G. epigaeum, that these two AMF species cannot be conspecific, and by molecular phylogenetics that BEG47 is a member of the genus Diversispora. Conclusions: This study highlights that experimental works published during the last >25 years on an AMF named ‘G. versiforme’ or ‘BEG47’ refer to D. epigaea, a species that is actually evolutionarily separated by hundreds of millions of years from all members of the genera in the Glomerales and thus from most other commonly used AMF ‘laboratory strains’. Detailed redescriptions substantiate the renaming of G. epigaeum (BEG47) as D. epigaea, positioning it systematically in the order Diversisporales, thus enabling an evolutionary understanding of genetical, physiological, and ecological traits, relative to those of other AMF. Diversispora epigaea is widely cultured as a laboratory strain of AMF, whereas G. versiforme appears not to have been cultured nor found in the field since its original description

    In vivo proteomics identifies the competence regulon and AliB oligopeptide transporter as pathogenic factors in pneumococcal meningitis

    Streptococcus pneumoniae (pneumococci) is a leading cause of severe bacterial meningitis in many countries worldwide. To characterize the repertoire of fitness and virulence factors predominantly expressed during meningitis we performed niche-specific analysis of the in vivo proteome in a mouse meningitis model, in which bacteria are directly inoculated into the cerebrospinal fluid (CSF) cisterna magna. We generated a comprehensive mass spectrometry (MS) spectra library enabling bacterial proteome analysis even in the presence of eukaryotic proteins. We recovered 200,000 pneumococci from CSF obtained from meningitis mice and by MS we identified 685 pneumococci proteins in samples from in vitro filter controls and 249 in CSF isolates. Strikingly, the regulatory two-component system ComDE and substrate-binding protein AliB of the oligopeptide transporter system were exclusively detected in pneumococci recovered from the CSF. In the mouse meningitis model, AliB-, ComDE-, or AliB-ComDE-deficiency resulted in attenuated meningeal inflammation and disease severity when compared to wild-type pneumococci indicating the crucial role of ComDE and AliB in pneumococcal meningitis. In conclusion, we show here mechanisms of pneumococcal adaptation to a defined host compartment by a proteome-based approach. Further, this study provides the basis of a promising strategy for the identification of protein antigens critical for invasive disease caused by pneumococci and other meningeal pathogens

    Alpha-states in de Sitter space

    Field theory in de Sitter space admits a one-parameter family of vacua determined by a superselection parameter alpha. Of these vacua, the Euclidean vacuum uniquely extrapolates to the vacuum of flat Minkowski space. States which resemble the alpha-vacua can be constructed as excitations above the Euclidean vacuum. Such states have modes alpha(k) which decay faster that k^{(1-d)/2}. Fields in such states exhibit non-local correlations when examined from the perspective of fields in the Euclidean vacuum. The dynamics of such entangled states are fully consistent. If an alpha-state with properties that interpolate between an alpha-vacuum and the Euclidean vacuum were the initial condition for inflation, a signature for this may be found in a momentum dependent correction to the inflationary power spectrum. The functional formalism, which provides the tool for examining physics in an alpha-state, extends to fields of other spin. In particular, the extension to spin-2 may proffer a new class of infrared modifications to gravitational interactions.     Deconstruction and Holography

    It was recently pointed out that the physics of a single discrete gravitational extra dimension exhibits a peculiar UV/IR connection relating the UV scale to the radius of the effective extra dimension. Here we note that this non-locality is a manifestation of holography, encoding the correct scaling of the number of fundamental degrees of freedom of the UV theory. This in turn relates the Wilsonian RG flow in the UV theory to the effective gravitational dynamics in the extra dimension. The relevant holographic c-function is determined by the expression for the holographic bound. Holography in this context is a result of the requirements of unitarity and diffeomorphism invariance.     Human noroviruses are the primary causative agents of acute gastroenteritis and a pressing public health burden worldwide. There are currently no vaccines or small molecule therapeutics available for the treatment or prophylaxis of norovirus infections. Norovirus 3CL protease plays a vital role in viral replication by generating structural and nonstructural proteins via the cleavage of the viral polyprotein. Thus, molecules that inhibit the viral protease may have potential therapeutic value.
