Multi-drug Resistant (MDR), Extended Spectrum Beta-lactamase (ESBL) and New Delhi Metallo Beta-lactamase (NDM) Producing Escherichia Coli in Rescued Sloth Bears (Melursus Ursinus), India
Abstract
The study reports the MDR, ESBL, and NDM producing Escherichia coli (CRE) isolated from the rescued sloth bear (Melursus ursinus), India. The faecal samples of adult rescued sloth bear (n=21) were collected from a rescue center located in India during 2015-2016 and processed for isolation and antibacterial susceptibility pattern of E. coli. 45E. coli isolates were recovered, and on phenotypic screening, 23 were MDR, 17 were ESBL producers, and five were carbapenem-resistant (CR). The MDR isolates carried beta-lactamase, chloramphenicol, aminoglycosides, tetracycline, fluroquinone, and sulphadimidine resistance genes. All the phenotypic ESBL producing isolates had blaCTX-M genes. On genotypic screening, three CRE (60.0 %, 3/5) were positive for blaNDM carbapenemase gene. Efflux pump-mediated carbapenem resistance was noticed in two CRE isolates (40.0 %, 2/5).The CRE also isolates co-harbored AMR genes like blaTEM-1, blaAmpC, qnrA, qnrB, qnrS, tetA, tetB and sulI. Virulence screening of the resistant isolates revealed the presence of Stx1, Stx2, eae, hlyA genes.Plasmid incompatibility (Inc) typesof three NDM positive isolates revealed that two isolates blaNDM-5 gene on Incl1 an one isolate on IncF plasmid. Apart from NDM genes, the plasmids also carried tetracycline, beta-lactamase and quinolone resistance genes. The plasmid multilocus sequence typing (pMLST) of the E. coli Incl1 plasmid showed the Sequence Type (ST) 297.This appears to be the first report of multi-drug resistant, extended spectrum beta-lactamase (ESBL) producing and blaNDM-5geneson Incl1 and IncF plasmids in rescued sloth bear.</jats:p
Non-structural Enterotoxin (NSP4) Gene based Molecular Characterization of Caprine and Ovine Rotavirus A, India
Rotavirus A (RVA) causes viral gastroenteritis in humans and animals, including calves, piglets, and foals. The current study reports the genetic characterization of the full-length enterotoxin gene, NSP4, from caprine and ovine species. Upon characterizing eight full-length NSP4 genes by sequencing, it was found that the four caprine and three ovine RVAs NSP4 genes are of E2 genotype and the sole ovine RVA isolate was found to be of E1 genotype. In the sequence and phyloanalysis of the NSP4 gene the seven E2 genotypes clustered with bovine, human, and caprine isolates from India and Bangladesh, respectively. The E1 genotype of ovine RVA was closer to human RVA isolate from India. The nucleotide per cent identity analysis revealed that all E2 genotype strains of caprine and ovine species ranged from 88.4% to 90.4% and it was found common to both the reference human RVA isolates DS-1 and AU-1. Whereas, the E1 genotype ovine strain clustered with human RVA isolates with 93.1% nucleotide per cent identity. The RVA strains circulating in caprine and ovine populations may share a common origin which is usually found in artiodactyl species because humans share a common dwelling with animals. Future studies are needed to confirm these findings of their relationship with humans and large animals

Genomic Analysis of an Indian G8P[1] Caprine Rotavirus-A Strain Revealing Artiodactyl and DS-1-Like Human Multispecies Reassortment
The surveillance studies for the presence of caprine rotavirus A (RVA) are limited in India, and the data for the whole-genome analysis of the caprine RVA is not available. This study describes the whole-genome-based analysis of a caprine rotavirus A strain, RVA/Goat-wt/IND/K-98/2015, from a goat kid in India. The genomic analysis revealed that the caprine RVA strain K-98, possess artiodactyl-like and DS-1 human-like genome constellation G8P[1]-I2-R2-C2-M2-A3-N2-T6-E2-H3. The three structural genes (VP2, VP4, and VP7) were close to caprine host having nucleotide-based identity range between 97.5 and 98.9%. Apart from them, other gene segments showed similarity with either bovine or human like genes, ultimately pointing toward a common evolutionary origin having an artiodactyl-type backbone of strain K-98. Phylogenetically, the various genes of the current study isolate also clustered inside clades comprising Human-Bovine-Caprine isolates from worldwide. The current findings add to the knowledge on caprine rotaviruses and might play a substantial role in designing future vaccines or different alternative strategies combating such infections having public health significance. To the best of our knowledge, this is the first report on the whole-genome characterization of a caprine RVA G8P[1] strain from India. Concerning the complex nature of the K-98 genome, whole-genome analyses of more numbers of RVA strains from different parts of the country are needed to comprehend the genomic nature and genetic diversity among caprine RVA.</jats:p
Estudo retrospectivo de 66 casos de urolitíase obstrutiva em ovinos
Objetivou-se realizar um estudo retrospectivo dos principais achados epidemiológicos, clínicos, patológicos e laboratoriais de ovinos acometidos por urolitíase obstrutiva, atendidos na Clínica de Bovinos, Campus Garanhuns/ UFRPE. Para isso, foram analisadas 66 fichas clínicas de ovinos, todos machos não castrados, com idade entre dois meses e cinco anos. A maioria dos casos (79,63%) ocorreu na época chuvosa. A maioria dos animais (87,88%) era criada intensivamente e todos eram alimentados com concentrados. Os sinais clínicos mais importantes foram as manifestações de dor, congestão de mucosas, hipertermia e aumento da tensão abdominal associados a obstrução do trato urinário. Metade dos animais apresentou obstrução total da uretra. Destes, 69,70% morreram, enquanto nos casos de obstrução parcial, 30,30% morreram. Nos exames laboratoriais verificou-se neutrofilia e desvio à esquerda regenerativo, hiperfibrinogenemia e azotemia. Na urinálise constatou-se hematúria em 89,29% dos casos e pH ácido em 46,43%. Na sedimentoscopia predominaram hemácias, leucócitos, células de descamação do epitélio uretral e cristais de urato amorfo. Os achados necroscópicos renais mais frequentes foram pielonefrite (61,54% dos casos) e hidronefrose (50%). Nos ureteres foram visualizados ureterite e hidroureter. Na bexiga foi mais evidente a cistite hemorrágica difusa (50%) e a presença de urólitos (57,69%). Na uretra observaram-se urólitos (61,54%) e uretrite hemorrágica difusa (57,69%). Quanto à composição dos urólitos predominaram os compostos por oxalato de cálcio hidratado. Conclui-se que no Agreste de Pernambuco a urolitiase é uma doença importante de ovinos, com alta letalidade, estando associada à alimentação rica em concentrados e sal mineral
