    IMIDACLOPRID TOXICITY AND ITS ATTENUATION BY AQUEOUS EXTRACT OF MORINGA OLEIFERA LEAF IN ZEBRA FISH, DANIO RERIO

    Objective: Imidacloprid (IMC) is a systemic chloro-nicotinyl insecticide that is applied in crop fields to control a wide variety of harmful insects and pests. The objective of the present study was to evaluate the mitigating effects of Moringa leaf extract (MLE) against Imidacloprid (IMC) induced mortality after acute toxicity bioassay in zebrafish, Danio rerio. Methods: The fishes were exposed to various concentrations of IMC to estimate the LC50 values at different exposure periods using the probit analysis. To assess the attenuating effect on mortality fishes were exposed to different concentrations of MLE along with 24 and 96 h LC50 of IMC. Mortality rate of fishes was recorded during all experimental periods. Results: The LC50 values were found to be 0.423, 0.352, 0.297 and 0.270 ml/l after 24, 48, 72 and 96 h respectively. The upper confidence limits were 0.491, 0.408, 0.334 and 0.302 ml/l and lower confidence limits were 0.355, 0.296, 0.260 and 0.238 ml/l for 24, 48, 72 and 96 h respectively. The fishes remain alive at 12 ml/l and 10 ml/l of MLE against 24 and 96 h LC50 respectively. Some behavioral responses such as violent, sluggish and erratic movement, respiratory distress, mucous secretion all over the body, discoloration were also observed in IMC treated fishes in comparison to MLE treated fishes. Conclusion: These findings demonstrated that MLE has mitigating effects against the toxic influence of IMC on mortality in zebra fishes which is attributed to its antioxidant effects

    Effect of cavity varnish and intermediate restorative material on coronal microleakage in endodontically treated tooth

    Aim: Evaluation of microleakage in endodontically treated teeth that have been coronally sealed with eugenol and noneugenol containing zinc oxide cement, with and without the use of cavity varnish. Materials and Methods: The crowns of single rooted teeth were removed at the cementoenamel junction (CEJ) and the root canals were prepared and obturated with gutta-percha points and zinc oxide eugenol (ZOE) sealer using lateral condensation technique. The samples were randomly divided into four groups-group 1, coronal seal was obtained with ZOE cement; in group 2, three coats of cavity varnish followed with coronal seal of ZOE cement; in group 3, Litark a (noneugenol cement) was placed in the prepared cavity as in group 1; and in group 4, cavity varnish and Litark were placed instead of ZOE as in group 2. The specimens were immersed in 2% methylene blue dye and examined under traveling microscope for evidence of dye penetration of the material. Statistical Analysis: In this study, Kruskal-Wallis test, Mann-Whitney U, and Wilcoxon W tests were used. Results: Kruskal-Wallis test with P=0.000 indicated that a significant difference exists between the four groups. Mann-Whitney U test and Wilcoxon W test showed a significant difference at 0.05 levels as follows: group 1 with group 3 and 4, group 2 with group 3 and 4, and group 3 with group 4. There was no significant difference at 0.05 levels between group 1 and group 2. Conclusion: Group 4 showed significant less amount of dye penetration as compared with other groups

    Efficacy and safety of 90,000 IU versus 300,000 IU single dose oral Vitamin D in nutritional rickets: A randomized controlled trial

    Aim: To compare efficacy and safety of 90,000 IU versus 300,000 IU oral single dose vitamin D for treatment of nutritional rickets. Study Design: Randomized controlled trial. Setting: Tertiary care hospital. Participants: One hundred ten children (6 months to 5 years, median age 10.5 months) with rickets. Exclusion criteria were disease affecting absorption, intake of calcium/vitamin D preparation in last 6 months, abnormal renal function, and rickets other than nutritional. Intervention: Vitamin D3 as a single oral dose 90,000 IU (group A, n = 55) or 300,000 IU (group B, n = 55). Methodology: Severity of rickets was scored on knee and wrist X-ray as per Thacher's radiographic score. Baseline serum levels of calcium, SAP, 25(OH)D, iPTH were measured. Follow up was done at 1 week, 4 weeks, and 12 weeks. Outcome Variable: Primary – Radiographic score at 3 months. Secondary – Serum levels of 25(OH)D, SAP, and iPTH at 3 months, clinical and biochemical adverse effects. Results: Eighty-six subjects (43 in each group) completed the study. The radiographic score reduced from 6.90 to 0.16 in group A and from 6.93 to 0.23 in group B. The levels of 25(OH)D, ALP, and PTH were similar between the groups at baseline and follow up. Hypercalciuria and hypercalcemia were seen more often in group B as was hypervitaminosis D. There were no clinical adverse events. Conclusions: Single oral dose vitamin D3 90,000 IU is safe and effective in achieving healing of rickets

    Background: An in vivo comparative evaluation to determine the accuracy of working length between radiographic and electronic apex locators. Aim: The study was aimed at evaluating the accuracy of electronic apex locator, to determine the working length of root canal, and to compare it with the radiographic method of working length determination. Materials and Methods: A total of 20 teeth selected for the study had to go for extraction because of periodontal or orthodontic reasons. Access cavity was prepared and the clinical estimated working length (CEWL) was determined with 10-25 no. K-file. A radiograph was then taken for determining the radiographic estimated working length (REWL). For electronic measurement of root canal, a 10 no. K-file was advanced toward the apex until it reached a 0.5 mm short of apex as shown by the apex locator. After fixing the file with a light cured composite, the tooth was extracted, the tooth surface was then longitudinally grounded using straight fissure diamond bur until the root canal and the tip of the file were visible. The distance of file from the minor constriction was measured with help of stereomicroscope. Statistical analysis : The chi-square test was used for statistical analysis for this study. Results: The chi-square test where χ2 = 21.034 with P = 0.000 indicated that a significant difference exists among the groups. The electronic method showed highest number of cases with the working length at the minor constrictor. Conclusion: The electronic method for determining the working length of root canal was found to be more accurate than the radiographic method

    CAP2 enhances germination of transgenic tobacco seeds at high temperature and promotes heat stress tolerance in yeast

    We reported earlier that ectopic expression of CAP2, a single AP2 domain containing transcription activator from chickpea (Cicer arietinum) in tobacco improves growth and development, and tolerance to dehydration and salt stress, of the transgenic plants. Here, we report that, in addition, the CAP2-transgenic tobacco seeds also exhibit higher germination efficiency at high temperature and show higher expression levels of genes for tobacco heat shock proteins and a heat shock factor. CAP2 was able to activate the 5′-upstream activating sequence of tobacco heat shock factor. Surprisingly, expression of CAP2 cDNA in Saccharomyces cerevisiae also enhanced heat tolerance, with increased expression of the gene for yeast heat shock factor 1 (Hsf1) and its target, the gene for yeast heat shock protein 104 (Hsp104). Sequence analysis of the Hsf1 promoter revealed the presence of a dehydration-responsive element/C-repeat-like element (DRE/CRE). Recombinant CAP2 protein bound to the DRE/CRE in the Hsf1 promoter in a gel shift assay and transactivated the Hsf1 promoter–His reporter construct. The full-length CAP2 protein was required to provide thermotolerance in yeast. If these findings are taken together, our results suggest that CAP2 is involved in the heat stress response and provides an example of functioning of a plant transcription factor in yeast, highlighting the strong evolutionary conservation of the stress response mechanism

    Multicomponent Template Effectsî—¸Preparation of Highly Porous Polyaniline Nanorods Using Crude Lemon Juice and Its Application for Selective Detection of Catechol

    This paper deals with the preparation and characterization of three different polyaniline samples, namely, PANI-HCl (synthesis in inorganic acid), PANI-Citric acid (synthesis in organic acid), and PANI-Lemon (synthesis in crude lemon juice–biological-derived acidic solution). PANI-Lemon is a low-density polymer with well-defined nanorod shape morphology (∼100 nm in diameter and ∼300–600 nm in length). Lemon juice manipulates the morphology of PANI because it has a lower p<i>K</i><sub>a</sub> and the matrix molecules produce template-like effects. The distinct structure of PANI-Lemon was confirmed from microscopic and spectroscopic analysis. In the same protonating condition, the alignment of chains is the dominating factor which governs the conductivity of polyaniline. PANI-Lemon was successfully applied for the electrocatalytic detection of catechol. The selective interaction of PANI-Lemon with catechol leads to the generation of a new redox center. PANI-Lemon-modified carbon paste electrode exhibited high sensitivity (49.68 μA μM<sup>–1</sup> cm<sup>–2</sup>), specificity, a wide linear range (from 5 μM to 100 mM), a low detection limit (2.1 μM), and less response time (2 s). On the basis of our results, we propose an eco-friendly concept "multicomponent template effects" to bring nanoscale morphological manipulation using a suitable natural extract. This concept could be equally implemented to synthetic polymers and other nanoparticles
