Tolleranza a stress salini in germoplasma sardo di orzo (<i>Hordeum vulgare</i> L.)
Sardinia is one of the Italian regions were the salty soil is most frequently found. Soil borne salinity often occurs together
with air bome salinity due to the effects of the salty wind. In Sinis, a peninsula located in Centrai Western Sardinia, both types of
salinity stress are present. In this area barley is widely cultivated and many farmers generally prefer to grow local populations
which are well adapted to local environmental conditions. The aim of the research was to characterize the responses to both types
of salinity stress (via root and via leaf) of two genotypes («Sinis 0/27» and «Sinis 0/28») extracted from a local Sinis population of
six-row barley, and compare them with two varieties, «California Mariout 67» and «Sunbar 400», well known to be salt tolerant
to soil borne salinity. The results showed that in barley differences in the effects of salt absorbed via the root vs foliar uptake exist,
togheter with genetic diversity for air borne salinity tolerance which is usually found only in coastal and cliff species. In particular
the Sinis genotypes appeared to be tolerant to salty spray due to their specific adaptation. According to these results, the use of
adapted germplasm seems to be worthwhile in breeding programs which are aimed at the synthesis of varieties suitable for areas
were salinity stresses frequently occur.
La Sardegna è una fra le regioni italiane in cui la presenza di terreni salsi è più frequente. A questa forma di stress salino si
accompagna spesso l'azione dei venti che trasportano acqua marina in sospensione inducendo uno stress salino per via
fogliare. Nel Sinis, una penisola della Sardegna centro-occidentale, entrambe le forme di stress salino sono presenti. Qui l'orzo
(Hordeum vulgare L.) è molto coltivato e generalmente sono utilizzate delle popolazioni locali adattate alle condizioni
pedoclimatiche della zona. Scopo del presente lavoro è lo studio della risposta a entrambe le forme di stress salino, per via
radicale e per via fogliare, in due genotipi («Sinis 0/27» e «Sinis 0/28») estratti da una popolazione di orzo polistico collezionata
nella penisola del Sinis a confronto con due varietà note per la loro tolleranza alla salinità del terreno, «California Mariout 67»
e «Sunbar 400». I risultati ottenuti indicano la possibilità, per quanto riguarda l'orzo, di distinguere fra una tolleranza allo
stress salino dovuto all'assunzione di sale per via radicale e una tolleranza allo stress salino causato dall'assorbimento di sale
per via fogliare che fino a oggi è stata riscontrata solamente in specie di non elevato interesse agronomico che tendono
a colonizzare gli ambienti costieri. I due genotipi del Sinis valutati, in particolare «Sinis 0/27», sono risultati dotati di una
buona capacità di tollerare lo stress salino applicato per via fogliare grazie al loro specifico adattamento. Ciò indica
l'importanza dell'utilizzazione di germoplasma adattato nel miglioramento genetico per ambienti caratterizzati dalla presenza
di stress salini
Osservazioni su una popolazione locale sarda di orzo polistico (<i>Hordeum vulgare</i> L.)
The aim of the research has been the evaluation of a local population of barley (Hordeum vulgare L.) grown in
Sardinia. In 1989, 41 lines of the local population and 5 controi varieties («Arma», «Barberousse», «Gerbel», «Jaidor»
and «Pirate») were evaluated for agronomie and morphologic traits.
The results have shown the presence of a considerable genetic variability in the local population, which is made
up of a miture of numerous genotypes. The lines showed an early stem extension and heading time compared to the
controls (5/3/89 vs 12/3/89 and 12/4/89 vs 17/4/89, respectively). The average values relative to Iodging index (3.2
vs 1.2), plant height (122.8 vs 108.4 cm) and 1,000 kernel weight (39.5 vs 27.2 g) were higher in thelines than in the
controls. The average yield of the lines was similar to that of the controls (3.46 vs 3.52 metric tons ha-1).
Scopo della ricerca è stato la valutazione di una popolazione locale di orzo coltivata in Sardegna. Nel 1989, su
41 linee estratte a caso dalla popolazione locale e su 5 varietà di controllo («Arma», «Barberousse», «Gerbel», «Jaidon»
e «Pirate») sono stati rilevati ad Ottava (SS) una serie di caratteri agronomici e morfologici.
I risultati hanno evidenziato la presenza di una variabilità genetica notevole entro la varietà agronomica che risulta
costituita da un miscuglio di numerosi genotipi. Rispetto alle varietà di controllo le linee sono state caratterizzate in
media da levata e spigatura anticipate, indice di allettamento, altezza del culmo e peso di 1000 cariossidi superiori. La
produzione media delle linee è risultata simile a quella delle varietà
Miglioramento genetico per la produzione di seme in <i>Lolium perenne</i> L.
Attraverso due cicli di selezione fenotipica, condotti a piante spaziate, sono state ottenute, dalla varietà di Lolium perenne L. 'Vejo' due popolazioni migliorate
per produzione di seme e peso di 1.000 semi.
Scopo della ricerca è stato quello di valutare, in condizioni agronomiche appropriate, le popolazioni migliorate.
La valutazione del materiale sperimentale ha interessato due distinte prove adiacenti: una a file è servita per valutare la produzione di seme, una a spaglio per
valutare la produzione di foraggio.
I risultati hanno dimostrato che il lavoro di selezione è risultato efficace soltanto per l'incremento del peso di 1.000 semi (+23%) mentre nessun guadagno è stato osservato nella popolazione selezionata per la produzione di seme.
Two cycles of phenotypic selection of L. perenne cv. 'Vejo' were applied to improve seed prodution in one population and 1,000 seed weight in another. Selection
was made on a spaced plants trial. The aim of the present research has been to evaluate the selected populations under appropriate agronomic conditions.
Evaluation of both populations has been carried out in a specialized crop for seed prodution and in a sward for forage production.
Results showed that seleetion was affective with respect to increase in 1,000 seed weight (+23%), while no results were achieved in selecting for high seed yield

On the basis of a botanical survey of naturally established pastures in Sardinia (Italy), ball clover (Trifolium nigrescens
p.p.) appeared to be one of the most widespread annual forage legumes. Thirteen natural populations collected in Sardinia
in late spring 1987 were evaluated in 1987-88 at Sassari for chromosome number, fertility indexes, 1,000 seed weight,
plant diameter 90 day after sowing (sowing date Oct. 30, 1987), flowering time, average stem length, number of stems
and dry matter yield per plant at harvest (harvesting date May 19, 1988). All the populations were diploid (2n = 2x = 16),
allogamous, self-incompatible and belonged to T. nigrescens Viv. subsp. nigrescens.
The wide variability of the agronomic traits under study permits to obtain positive results with breeding programs
aimed at developing ball clover varieties for pasture improvement in Sardinia. In particular, at population level 1,000
seed weight ranged from 0.163 g to 0.307 g, flowering time from Mar 13 to Apr 4 and dry matter yield per plant from
7.9 g to 17.9 g.
In conclusion, the multiple regression analysis and the path coefficients analysis showed that average stem lenght could
be used to assess the dry matter potentialities of the plants in the evaluation of large germplasm collections of ball clover.
Allo scopo di iniziare la valutazione del germoplasma sardo di Trifolium nigrescens p.p., una delle leguminose foraggere
annuali più diffuse nei pascoli naturali dell'isola, nella tarda primavera 1987 tredici popolazioni naturali sono state
raccolte in differenti aree della Sardegna. Le popolazioni sono state valutate, a Sassari, nel corso del 1987-88 per numero
cromosomico, indici di fertilità per autofecondazione e libera impollinazione, peso di 1.000 semi, diametro della pianta
a 90 giorni dalla semina, epoca di fioritura, lunghezza degli steli, numero di steli e peso secco della pianta alla raccolta.
Tutte le popolazioni sono risultate diploidi (2n = 2x = 16), allogarne, autoincompatibili, ad impollinazione entomofila e
appartenenti alla specie T. nigrescens Viv. subsp. nigrescens.
La variabilità osservata per i caratteri agronomici in esame suggerisce la possibilità di ottenere risultati positivi con
programmi di miglioramento genetico volti allo sviluppo di varietà di T. nigrescens da utilizzare nel miglioramento dei
pascoli in aree marginali della Sardegna. Di notevole interesse è la variabilità tra le medie delle popolazioni messa in
luce per il peso di 1.000 semi (tra 0,163 g e 0,307 g), l'epoca di fioritura (tra il 13 marzo e il 4 aprile) e la produzione
di sostanza secca per pianta (tra 7,9 g e 17,9 g). L'analisi della regressione multipla e l'analisi dei "path coefficients"
hanno evidenziato che la produzione di sostanza secca per pianta è fortemente influenzata dalla lunghezza media degli
steli. Poiché la determinazione di questo secondo carattere è più rapida di quella relativa alla produzione di sostanza
secca, almeno nelle prime fasi di valutazione del germoplasma di T. nigrescens sarebbe possibile aumentare il numero
di accessioni studiate utilizzando la lunghezza media degli steli per valutare indirettamente le potenzialità produttive delle
piante
Risposta dell'orzo (<i>Hordeum vulgare</i> L.) a differenti livelli di intensificazione colturale
Two source of barley germplasm (pure lines selected from local populations and improved varieties) were compared at
3 locations in Sardinia (Ottava, SS; Cabras, OR; Ussana, Ca) under different levels of agronomic treatments: with and without
chemical weed control and at two levels of nitrogen (80 g ha-1 e 40 kg ha-1 N).
The results of commerciaI barley varieties show different responses according to the input level.
Within Sardinian germplasm several interesting genotypes, for low-input conditions have been identified. The comparison
between the genotypes selected from the local populations and the commerciaI barley varieties underline that plant breeding for
low-input conditions could be improved by the utilization of germplasm adapted to extensive practices.
In tre località della Sardegna (Ottava, SS; Cabras, OR; Ussana, Ca) è stata effettuata una serie di prove comparative fra il
1990 e il 1993 utilizzando due gruppi di genotipi: varietà commerciali e linee pure selezionate all'interno di alcune popolazioni
locali sarde.
Gli interventi tecnici considerati sono stati la concimazione azotata e il controllo chimico delle infestanti; in particolare sono
stati utilizzati due livelli di concimazione azotata (80 kg ha-1 e 40 kg ha-1 di N) mentre per quanto riguarda il diserbo è stata
presa in considerazione la presenza o l'assenza del trattamento.
I risultati relativi alle sole varietà commerciali indicano la presenza di risposte differenziate in relazione alla riduzione degli
input. All'interno del germoplasma sardo sono state messe in evidenza delle linee di notevole interesse per la loro capacità
produttiva.
In base ai risultati ottenuti sembra possibile affermare che alcuni genotipi sono più adatti di altri ad essere coltivati in
presenza di bassi livelli di intensificazione colturale. Il confronto fra il comportamento delle linee selezionate dalle popolazioni
locali e le varietà commerciali sottolinea come il miglioramento genetico per l'ottenimento di varietà adattate ad
agroecosistemi caratterizzati dall'impiego di bassi input possa utilmente giovarsi di germoplasma adattato a condizioni
estensive di coltivazione
Total Value of Ownership and Overall Equipment Effectiveness analysis to evaluate the impact of automation on time and costs of therapeutic drug monitoring
Abstract Total Value of Ownership (TVO) and Overall Equipment Effectiveness (OEE) analysis are novel tools capable of monitoring and analyzing industrial processes by assessing the efficiency of the entire instrumental equipment and calculating instrument capacity utilization. Such integrated analysis, measuring quality indicators of the testing process, could also provide new perspectives and methodologies for the workflow organization of clinical laboratories. In this study, TVO and OEE were employed for the evaluation of two different configurations of a therapeutic drug monitoring sector, comparing the results obtained for immunosuppressant (ISD) and anti-epileptic drugs (AED) analysis as well as checking their quantitative performance in terms of limit of quantification, accuracy and precision. TVO analysis was performed for ISDs, including the Total Direct Labor Time, Total Cycle Time and Turnaround Time as well as cost of testing. Instruments' performance and workload were assessed using OEE indicator, studying Availability, Performance and Quality factors. Total Cycle Time for a batch was 3.55 h, decreasing of 1.5 h in the new setting where personnel are engaged for 0.98 h, 25% of total testing time. The calculated cost per sample was 6.60 euro. Availability values were significantly higher for automated sample-handling system and ISDs analysis by LC-MS. Higher Performance values were obtained for LC-MS system for AED and other TDM. Quality values were >0.94 for all instruments. TVO and OEE proved to be applicable to clinical laboratory environment, quantifying benefits and costs of newly developed semi-automated therapeutic drug monitoring sector. This novel approach based on an integrated analysis may help activity planning and quality improvement and could be used in the future for benchmarking progress as a product/process comparison tool in other laboratory fields
In planta production of two peptides of the Classical Swine Fever Virus (CSFV) E2 glycoprotein fused to the coat protein of potato virus X
BACKGROUND: Classical Swine Fever (CSFV) is one of the most important viral infectious diseases affecting wild boars and domestic pigs. The etiological agent of the disease is the CSF virus, a single stranded RNA virus belonging to the family Flaviviridae. All preventive measures in domestic pigs have been focused in interrupting the chain of infection and in avoiding the spread of CSFV within wild boars as well as interrupting transmission from wild boars to domestic pigs. The use of plant based vaccine against CSFV would be advantageous as plant organs can be distributed without the need of particular treatments such as refrigeration and therefore large areas, populated by wild animals, could be easily covered. RESULTS: We report the in planta production of peptides of the classical swine fever (CSF) E2 glycoprotein fused to the coat protein of potato virus X. RT-PCR studies demonstrated that the peptide encoding sequences are correctly retained in the PVX construct after three sequential passage in Nicotiana benthamiana plants. Sequence analysis of RT-PCR products confirmed that the epitope coding sequences are replicated with high fidelity during PVX infection. Partially purified virions were able to induce an immune response in rabbits. CONCLUSION: Previous reports have demonstrated that E2 synthetic peptides can efficiently induce an immunoprotective response in immunogenized animals. In this work we have showed that E2 peptides can be expressed in planta by using a modified PVX vector. These results are particularly promising for designing strategies for disease containment in areas inhabited by wild boars
Prime osservazioni su specie perenni ed annue autoriseminanti in vista della organizzazione di catene di foraggiamento in ambienti mediterranei
Nel corso del periodo 1987-89 sono state valutate le potenzialità produttive e la distribuzione della produzione di 32
tra ecotipi e varietà di graminacee e leguminose foraggere annue e perenni provenienti prevalentemente dall'Italia centrale.
La prova è stata condotta contemporaneamente in tre ambienti appartenenti a diverse zone bioclimatiche dell'area mediterranea
(Perugia, Grosseto e Sassari) utilizzando uno schema sperimentale a blocchi randomizzati con quattro ripetizioni.
I rilievi effettuati hanno riguardato il ricopri mento specifico e la produzione di sostanza secca determinata con criteri differenti:
mensilmente per le leguminose annue; al 50% della fioritura e quindi mensilmente per le leguminose perenni; all'inizio
della spigatura e quindi mensilmente per le graminacee.
Per le specie annuali oltre alla sostanza secca è stata determinata la produzione di seme.
L'obiettivo è stato quello di caratterizzare materiali che possano essere impiegati scalarmente per dilatare il periodo di utilizzazione diretta al pascolo.
Dai risultati è stato possibile individuare, a seconda delle località, alcune popolazione di Medicago polymorpha L., Trifolium subterraneum L. e Lotus sp. pl. che hanno permesso di anticipare il periodo di utilizzazione primaverile e prolungare quello autunnale. Su questa base sono stati ipotizzati esempi di catene di foraggiamento semplificate (a tre anelli).
Per quanto concerne le specie annue autoriseminanti, esse sembrano poter svolgere un importante ruolo nella regolazione
della distribuzione stagionale della produzione foraggera nelle due località caratterizzate da un clima più tipicamente mediterraneo (Grosseto e Sassari) mentre la produzione di seme è risultata più che soddisfacente per assicurare l'autorisemina in tutte e tre le località.
During the period 1987-1989, the yield and the distribution of forage production of 32 ecotypes and varieties of annual
and perennial forage Iegumes and grasses were evaluated. The origin of plant materials was CentraI Italy. The trial was
carried out in three environments characterized by different Mediterranean subclimates (Perugia, Grosseto and Sassari).
The experimental design was a randomized complete block with four replicates.
Specific ground cover was assessed in fall 1987, 1988 and 1989. Dry matter yield was assessed: monthly for annual
legumes; at 50% of flowering and then monthly for perennial legumes; at early heading and then monthly for grasses.
Seed yield of annual species was also recorded. Aim of the trial was to characterize plant materials that could be utilized subsequently in order to extend the period of direct utilization of herbage by grazing animals. It was possible depending
on localities, to identify some populations of Medicago polymorpha L., Trifolium subterraneum L. and Lotus sp. pl.. that allowed to bring forward the spring utilization and to extend the autumn utilization.
On the basis of the resu1ts obtained, it was possible to set up some simplified forage chains. Annual self reseeding species seemed to be more important to improve seasonal distribution of herbages in the typical Mediterranean locations (Sassari and Grosseto). Seed production was more than appreciable in order to allow self reseeding at the three locations
Three-dimensional graphene on a nano-porous 4H-SiC backbone: a novel material for food sensing applications
Sensors which are sensitive to volatile organic compounds and thus able to
monitor the conservation state of food, are precious because they work
non-destructively and allow to avoid direct contact with the food, ensuring
hygienic conditions. In particular, the monitoring of rancidity would solve a
widespread issue in food storage. The sensor discussed here is produced
utilizing a novel three-dimensional arrangement of graphene, which is grown on
a crystalline silicon carbide (SiC) wafer previously porousified by chemical
etching. This approach allows a very high surface-to.volume ratio. Furthermore,
the structure of the sensor surface features a large amount of edges, dangling
bounds, and active sites, which make the sensor, on a chemically robust
skeleton, chemically active, particularly to hydrogenated molecules. The
interaction of the sensor with such compounds is read out by measuring the
sensor resistance in a four wire configuration. The sensor performance has been
assessed on three hazelnut samples: sound hazelnuts, spoiled hazelnuts, and
stink bug hazelnuts. A resistance variation of about DeltaR = 0.13 (0.02) Ohm
between sound and damaged hazelnuts has been detected. Our measurements confirm
the ability of the sensor to discriminate between sound and damaged hazelnuts.
The sensor signal is stable for days, providing the possibility to use this

Effects of grapevine applications of fosetyl-aluminium formulations for downy mildew control on “esca” and associated fungi
Esca of grapevine is a fungal disease with a complex aetiology that is common in almost all regions of the world where grapes are cultivated. Despite much research, no effective control of the disease has been found. We investigated possible activity of fosetyl-aluminium (fosetyl-Al), an active ingredient in many fungicides against downy mildew, to inhibit development of esca in grapevine and the main pathogens linked to the disease, Phaeomoniella chlamydospora (Pch) and Phaeoacremonium aleophilum (Pal). In greenhouse experiments conducted on vines artificially inoculated with Pch or Pal, we found a reduction in necrosis in the woody tissue. In field experiments conducted over several years statistically significant reductions in the annual and cumulative incidence of the disease symptoms, and in cumulative vine mortality, were seen. The effect of fosetyl-Al treatments on leaf gas exchange, levels of resveratrol and å-viniferin in the wood, and enzymatic activities were also studied. Hypotheses regarding the mechanism of action of fosetyl-Al against esca are outlined, and the possible use of products containing the chemical, applied on foliage to control downy mildew, is discussed as a strategy for control of esca
