    Sex-specific changes in gene expression and delayed sex differentiation in response to estrogen pollution in grayling (Salmonidae)

    The synthetic 17α-ethinylestradiol (EE2) is an estrogenic compound of oral contraceptives and therefore a common pollutant that has been suspected to affect the demography of river-dwelling salmonids. We study a population of European grayling (Thymallus thymallus) that suffers from sex ratio distortions. Here we test how ecologically relevant concentrations of EE2 affect sex-specific gene expression around early stages of sex differentiation. We collected gametes from F1s of wild spawners, used them for in vitro fertilizations, and raised the resulting embryos singly under experimentally controlled conditions. Embryos were either exposed to 1ng/L EE2 or sham-exposed. RNA was collected from samples taken 10 days before hatching, at the day of hatching, and towards the end of the yolk-sac stage, to study gene expression and relate it to genetic sex (sdY genotype). We found that EE2 affects gene expression of a very large number of genes especially at the day of hatching. The effects of EE2 on gene expression is strongly sex-specific. At the day of hatching, EE2 affected about twice as many genes in females than in males, and towards the end of the yolk-sac larval stage, EE2 effects were nearly exclusively observed in females. Among the many effects was, for example, a surprising EE2-induced molecular masculinization in the females’ heads. Histological examination of gonadal development of EE2-treated or sham-exposed juveniles during the first 4.5 months after hatching revealed a delaying effect of EE2 on sex differentiation. Because grayling sex determination goes through an all-male stage (a rare case of undifferentiated gonochorism), the rate of EE2-induced sex reversal could not be unequivocally determined during the observational period. However, two EE2-treated genetic males had ovarian tissues at the end of the study. We conclude that common levels of EE2 pollution affect grayling from very early stages on by interfering with male and female gene expression around the onset of sex differentiation, by delaying sex differentiation, and by feminizing some males

    Acute ingestion of a novel whey-derived peptide improves vascular endothelial responses in healthy individuals: a randomized, placebo controlled trial

    <p>Abstract</p> <p>Background</p> <p>Whey protein is a potential source of bioactive peptides. Based on findings from <it>in vitro </it>experiments indicating a novel whey derived peptide (NOP-47) increased endothelial nitric oxide synthesis, we tested its effects on vascular function in humans.</p> <p>Methods</p> <p>A randomized, placebo-controlled, crossover study design was used. Healthy men (n = 10) and women (n = 10) (25 ± 5 y, BMI = 24.3 ± 2.3 kg/m<sup>2</sup>) participated in two vascular testing days each preceded by 2 wk of supplementation with a single dose of 5 g/day of a novel whey-derived peptide (NOP-47) or placebo. There was a 2 wk washout period between trials. After 2 wk of supplementation, vascular function in the forearm and circulating oxidative stress and inflammatory related biomarkers were measured serially for 2 h after ingestion of 5 g of NOP-47 or placebo. Macrovascular and microvascular function were assessed using brachial artery flow mediated dilation (FMD) and venous occlusion strain gauge plethysmography.</p> <p>Results</p> <p>Baseline peak FMD was not different for Placebo (7.7%) and NOP-47 (7.8%). Placebo had no effect on FMD at 30, 60, and 90 min post-ingestion (7.5%, 7.2%, and 7.6%, respectively) whereas NOP-47 significantly improved FMD responses at these respective postprandial time points compared to baseline (8.9%, 9.9%, and 9.0%; <it>P </it>< 0.0001 for time × trial interaction). Baseline reactive hyperemia forearm blood flow was not different for placebo (27.2 ± 7.2%/min) and NOP-47 (27.3 ± 7.6%/min). Hyperemia blood flow measured 120 min post-ingestion (27.2 ± 7.8%/min) was unaffected by placebo whereas NOP-47 significantly increased hyperemia compared to baseline (29.9 ± 7.8%/min; <it>P </it>= 0.008 for time × trial interaction). Plasma myeloperoxidase was increased transiently by both NOP-47 and placebo, but there were no changes in markers inflammation. Plasma total nitrites/nitrates significantly decreased over the 2 hr post-ingestion period and were lower at 120 min after placebo (-25%) compared to NOP-47 (-18%).</p> <p>Conclusion</p> <p>These findings indicate that supplementation with a novel whey-derived peptide in healthy individuals improves vascular function.</p

    Identification of co-expression gene networks, regulatory genes and pathways for obesity based on adipose tissue RNA Sequencing in a porcine model

    Background: Obesity is a complex metabolic condition in strong association with various diseases, like type 2 diabetes, resulting in major public health and economic implications. Obesity is the result of environmental and genetic factors and their interactions, including genome-wide genetic interactions. Identification of co-expressed and regulatory genes in RNA extracted from relevant tissues representing lean and obese individuals provides an entry point for the identification of genes and pathways of importance to the development of obesity. The pig, an omnivorous animal, is an excellent model for human obesity, offering the possibility to study in-depth organ-level transcriptomic regulations of obesity, unfeasible in humans. Our aim was to reveal adipose tissue co-expression networks, pathways and transcriptional regulations of obesity using RNA Sequencing based systems biology approaches in a porcine model. Methods: We selected 36 animals for RNA Sequencing from a previously created F2 pig population representing three extreme groups based on their predicted genetic risks for obesity. We applied Weighted Gene Co-expression Network Analysis (WGCNA) to detect clusters of highly co-expressed genes (modules). Additionally, regulator genes were detected using Lemon-Tree algorithms. Results: WGCNA revealed five modules which were strongly correlated with at least one obesity-related phenotype (correlations ranging from -0.54 to 0.72, P <0.001). Functional annotation identified pathways enlightening the association between obesity and other diseases, like osteoporosis (osteoclast differentiation, P = 1.4E(-7)), and immune-related complications (e. g. Natural killer cell mediated cytotoxity, P = 3.8E(-5); B cell receptor signaling pathway, P = 7.2E(-5)). Lemon-Tree identified three potential regulator genes, using confident scores, for the WGCNA module which was associated with osteoclast differentiation: CCR1, MSR1 and SI1 (probability scores respectively 95.30, 62.28, and 34.58). Moreover, detection of differentially connected genes identified various genes previously identified to be associated with obesity in humans and rodents, e.g. CSF1R and MARC2. Conclusions: To our knowledge, this is the first study to apply systems biology approaches using porcine adipose tissue RNA-Sequencing data in a genetically characterized porcine model for obesity. We revealed complex networks, pathways, candidate and regulatory genes related to obesity, confirming the complexity of obesity and its association with immune-related disorders and osteoporosis

    Reproductive health of brown trout inhabiting Swiss rivers with declining fish catch

    In recent years, brown trout catches have been declining in many Swiss rivers . One hypothesis is that this declining catch is linked to environmental estrogens, known to have the potential to adversely affect fish reproduction. In order to assess if the reproductive health of brown trout is impaired, we sampled fish at three sites along four rivers with a well documented catch decline. These rivers are affected by inputs of treated sewage effluent. The sampling was conducted during two years; we measured plasma vitellogenin (Vtg) concentrations and surveyed gonadal histology. Analysis of plasma Vtg revealed elevated concentrations (up to 8 mu g/mL) in only 10 out of 197 males. Furthermore, there were no site specific patterns to this induction. These results indicate that the exposure to estrogenic compounds is low. Also the incidence of ovarian atresia was low and we found no male intersex fish . In contrast to males, females caught along two rivers had spermatogenic activity in ovarian tissue. However, this intersex condition does not appear to be connected to exposure to environmental estrogens. At one of 12 sites there was a high incidence of gonadal parasites in ovarian tissue, which may affect reproductive output but was not a general problem across sites. In conclusion, the exposure to estrogenic compounds does not appear to significantly affect the reproductive parameters we investigated in Swiss brown trout

    Transfer kinetics of polar organic compounds over polyethersulfone membranes in the passive samplers Pocis and Chemcatcher

    Passive samplers for polar organic compounds often use a polyethersulfone (PES) membrane to retain the particulate sorbent material (e.g., in a POCIS; polar organic chemical integrative sampler) or to reduce the sampling rate and thus extend the kinetic regime (e.g., in a Chemcatcher). The transport kinetics over the PES membrane are evaluated here in a short-term (6 days) and a long-term (32 days) experiment with POCIS and Chemcatchers. Passive samplers were placed in a channel with flowing river water that was spiked with 22 organic chemicals including pharmaceuticals, pesticides and biocides; with logK(ow) (logarithmic octanol-water partitioning coefficient) values between -2.6 and 3.8. Samplers were removed at intervals and membranes and sorbent material were extracted and analyzed with LC-MS/MS. Uptake kinetics of the compounds fell between two extremes: (1) charged chemicals and chemicals of low hydrophobicity did not accumulate in PES and rapidly transferred to the sorbent (e.g., diclofenac) and (2) more hydrophobic chemicals accumulated strongly in the PES and appeared in the sorbent after a lag-phase (e.g., diazinon and diuron). Sorption kinetics were modeled with a three-compartment first-order kinetic model to determine uptake and elimination rate constants and partitioning coefficients. Water PES partitioning coefficients fitted with the model correlated well with experimentally determined values and logK(ow). Sampling rates of Chemcatcher (0.02-0.10 L/d) and POCIS (0.02-0.30 L/d) showed similar patterns and correlated well. Thus the samplers are interchangeable in practical applications. Longer lag-phases may pose problems when calculating time-weighted average aqueous concentrations for short passive sampling windows and for a correct integrative sampling of fluctuating concentrations
