    The effectiveness of axial flow fans in a-frame plenums

    Thesis (PhD (Mechanical and Mechatronic Engineering)--University of Stellenbosch, 1990.ENGLISH ABSTRACT: The ultimate goal of this project is to ensure a better understanding of the governing mechanisms present when flow distorting components are installed in close proximity of an axial flow fan. The effect of different parameters on the operation of axial flow fans is investigated. These parameters are divided into flow enhancing and flow reduction effects. The performance of an axial flow fan can be enhanced by changing the tip clearance, by adding a solid disc to the hub of the fan or by varying the number of fan blades. Flow reductions are caused by components such as inlet grids, walkways and their supporting structures, heat exchangers and windwalls. The effects of flow enhancing components are measured and compared to the results of other authors. The sensitivity of these effects to parameters such as the type of fan rotor and the specific system in which the rotor is installed is highlighted. The system effect (the interaction between the fan rotor and flow resistances in close proximity of each other) of individual components, as well as the combination of different components, is predicted both theoretically and experimentally. These predictions are compared to measured data relevant to the components in an installation where the system effects are present. The results are correlated to the kinetic energy flux coefficient of the flow at different locations within the installation. Experimental data obtained from a full scale unit (inlet shroud diameter of 9,216 m) are used to compare to scaled data from the model (inlet shroud diameter of 1,542 m). The hub to tip ratio of the axial flow fans investigated is 0,15. The most important conclusions are that the performance of the type of axial flow fan under investigation can be improved by reducing its tip clearance and by installing a solid disc to the downstream side of the rotor. An increase in the number of blades of the fan leads to only marginal improvements in the fan performance. The overall performance of the system can also be improved by removing some of the flow resisting components, or by changing their relative positions. All these conclusions are based on the assumption that the power input to the fan rotor remains constant.AFRIKAANSE OPSOMMING: Die uiteindelike doel van hierdie projek is om te verseker dat die beherende meganismes wat teenwoordig is wanneer vloeiversteurende komponente in die nabyheid van 'n aksiaalwaaier geinstalleer word, beter verstaan word. Die effek van verskillende parameters op die werkverrigting van aksiaalwaaiers word ondersoek. Hierdie parameters word verdeel in vloeiverbeterings- en vloeiverminderingseffekte. Die werkverrigting van 'n aksiaalwaaier kan verbeter word deur die lempuntspeling te verstel, deur 'n soliede skyf aan die naaf van die waaierrotor te installeer, of deur die aantal lemme te verander.     The effect of three bufadienolide cardiac glycosides on contraction of isolated rat jejunum

    Three cardiac glycosides were screened for pharmacological effects on isolated rat jejunum. The contraction of rat jejunum with epoxyscillirosidin, a non-cumulative bufadienolide, and cotyledoside and tyledoside D, both cumulative neurotoxic bufadienolides were compared with methacholine. The results indicate that all three bufadienolides cause contraction of jejunal smooth muscle. When combined with atropine (1X10ˉ⁶ M) the response of epoxyscillirosidin and tyledoside D decreased, indicating suppression of a cholinergic response caused by the cardiac glycosides.The articles have been scanned in colour with a HP Scanjet 5590; 600dpi.     Preclinical evaluation of 68Ga-labeled 1,4,7-triazacyclononane-1,4,7-triacetic acid-ubiquicidin as a radioligand for PET infection imaging

    Antimicrobial peptides such as ubiquicidin (UBI) are believed to differentiate between mammalian and bacterial or fungal cells. (99m)Tc-UBI29-41 was previously tested for detecting infection in humans using SPECT. For the present study, the UBI fragment UBI29-41 (TGRAKRRMQYNRR) was conjugated to 1,4,7-triazacyclononane-triacetic acid (NOTA), radiolabeled with (68)Ga, and investigated in a rabbit infection model. METHODS : (68)Ga was obtained from a 1.85-GBq (68)Ge/(68)Ga generator. New Zealand White rabbits were anesthetized with ketamine/medetomidine before tracer administration and placed in a clinical PET/CT scanner. (68)Ga-1,4,7-triazacyclononane-1,4,7-triacetic-acid-ubiquicidin29-41 ((68)Ga-NOTA-UBI29-41) was formulated in saline solution, and 101 ± 41 MBq were administered intravenously. The tracer distribution was studied by PET/CT imaging in animals (a) that were healthy, (b) bearing muscular Staphylococcus aureus infections and turpentine oil-induced muscular inflammations, and (c) bearing ovalbumin-induced lung inflammations. Static PET/CT imaging was performed at different time intervals up to 120 min after injection. For calculation of target-to-nontarget ratios, standardized uptake values were normalized against healthy thigh muscle, representing nontargeted tissue. RESULTS : PET/CT images of healthy animals showed predominant distribution in the kidneys, liver, and bladder; heart and spleen showed moderate, declining uptake, only. The biologic half-life in blood was 29 min. Urinary accumulation of (68)Ga-NOTA-UBI29-41 peaked at 3.8 ± 0.91 percentage injected dose per gram (%ID) at 120 min, and 88 ± 5.2 %ID was recovered in total urine. (68)Ga-NOTA-UBI29-41 imaging in (b) selectively visualized the muscular infection site and was differentiated from sterile inflammatory processes. Standardized uptake value ratios for muscles (infected/inflamed) were 2.9 ± 0.93, 2.9 ± 0.50, 3.5 ± 0.86, and 3.8 ± 0.90 at 5, 30, 60, and 90 min after injection, respectively. Rabbit lungs with asthma showed insignificant uptake. CONCLUSION : (68)Ga-NOTA-UBI29-41 was strongly localized in bacteria-infected areas and minimally detected in a sterile inflammation area in rabbit muscles. The findings propose this compound to be an excellent first-line PET/CT tracer to allow the distinguishing of infection from inflammation.http://jnm.snmjournals.org/hb201

    A reassessment of argasid tick salivary gland ultrastructure from an immuno-cytochemical perspective

    Previous morphological and histochemical studies of argasid tick salivary glands indicated that they were less complex than ixodid salivary glands, with only three granular cell types. The present study shows that there exist at least four different granular cell types in the salivary glands of the argasid tick Ornithodoros savignyi, based on immuno-localization of the anti-hemostatic factors, apyrase and savignygrin. Both anti-hemostatic factors were localized to dense core granule type \u27a\u27 and to granule type \u27b\u27, that shares a similar homogenous morphology with non-labeled granule type \u27d\u27. Furthermore, the major tick salivary gland proteins (TSGPs), previously implicated in granule biogenesis, were localized to all the granular cell types. This indicates that granular cell types with different morphologies can express the same proteins, while cell types that show similar morphologies may not express the same proteins. Argasid tick salivary glands seem to be more complex than previously thought and might not be amenable to morphological classification alone. Alternative classification methodologies that rely on physical expression patterns of the salivary gland proteome might be more reliable as markers for a specific granular cell type

    In vivo uptake and acute immune response to orally administered chitosan and PEG coated PLGA nanoparticles

    Nanoparticulate drug delivery systems offer great promise in addressing challenges of drug toxicity, poor bioavailability and non-specificity for a number of drugs. Much progress has been reported for nano drug delivery systems for intravenous administration, however very little is known about the effects of orally administered nanoparticles. Furthermore, the development of nanoparticulate systems necessitates a thorough understanding of the biological response post exposure. This study aimed to elucidate the in vivo uptake of chitosan and polyethylene glycol (PEG) coated Poly, DL, lactic-co-glycolic Acid (PLGA) nanoparticles and the immunological response within 24 h of oral and peritoneal administration. These PLGA nanoparticles were administered orally and peritoneally to female Balb/C mice, they were taken up by macrophages of the peritoneum. When these particles were fluorescently labelled, intracellular localisation was observed. The expression of pro-inflammatory cytokines IL-2, IL-6, IL-12p70 and TNF-α in plasma and peritoneal lavage was found to remain at low concentration in PLGA nanoparticles treated mice as well as ZnO nanoparticles during the 24 hour period. However, these were significantly increased in lipopolysaccharide (LPS) treated mice. Of these pro-inflammatory cytokines, IL-6 and IL-12p70 were produced at the highest concentration in the positive control group. The anti-inflammatory cytokines IL-10 and chemokines INF-γ, IL-4, IL-5 remained at normal levels in PLGA treated mice. IL-10 and INF-γ were significantly increased in LPS treated mice. MCP-1 was found to be significantly produced in all groups in the first hours, except the saline treated mice. These results provide the first report to detail the induction of cytokine production by PLGA nanoparticles engineered for oral applications
