Oxidation of cod microsomal lipids in situ and in vitro as affected by processing parameters
Muscle foods are a major source of food protein in many parts of the world including the United States. Acid and/or alkali solubilization is a recent method developed to separate muscle proteins with good functional properties. The unsaturated lipids of the fish muscle are highly susceptible to oxidative degradation, especially in the presence of hemoglobin. The goal of this research work was to study structural and physicochemical changes of fish membranes that occur during the acid or alkali solubilization procedures. The effect on lipid oxidation and ways to stabilize the membrane lipids against oxidation were also studied. Washed and unwashed cod, as well as cod microsomes were used as model systems for the study. Peroxide value, TBARS and sensory were employed to follow lipid oxidation. Citric acid and calcium chloride were effective in protecting the muscle from lipid oxidation either when used in washing the muscle or when added directly to the minced muscle. The maximum effect was achieved when citric acid and calcium chloride were added in the first washing step. Solubilizing the muscle at pH 3 or 10.8 in the absence of hemoglobin protected the protein isolates from lipid oxidation. However, when the muscle and hemoglobin were together exposed to low pH, oxidation was promoted. Protein isolates prepared by citric acid/calcium chloride-treated washed cod solubilized at pH 3 in the presence of hemoglobin exhibited very good oxidative stability. Part of the protection that citric acid/calcium chloride offered against lipid oxidation was attributed to the ability of citric acid to break down unstable peroxides. Hydrochloric acid was also able to break down formed peroxides to a similar degree as citric acid. At pH 5.3 or lower, 99% of isolated cod membranes sediment at low centrifugation speeds. Isolated membranes that were exposed to pH 3.0 or 10.8 were less susceptible to hemoglobin-mediated lipid oxidation. Cod hemoglobin exposed for 20sec or 20min to pH 3 was rendered less pro-oxidative than the untreated cod hemoglobin. However, when microsomes and hemoglobin were together exposed to low pH, oxidation was promoted. Microsomes prepared in the presence of citric acid and calcium chloride were less susceptible to hemoglobin-mediated lipid oxidation compared to untreated microsomal suspensions. The soluble fraction of the cod muscle (press juice) was able to retard lipid oxidation of washed cod and protein isolate, even when washed cod was solubilized at low pH in the presence of hemoglobin. Press juice retained its anti-oxidative activity even after exposure to low pH. Press juice was able to prevent hemoglobin-mediated, NADH-dependent and Fe-ascorbate mediated lipid oxidation of isolated cod microsomes
Removal of lipids and diarrhetic shellfish poisoning toxins (DSP) from blue mussels (Mytilus edulis) by an acid or alkaline protein solubilization technique
Diarrhetic shellfish poisoning (IDSP) toxins pose a serious health risk for consumers of bivalves and other shellfish, as well as a huge economic burden for the bivalve-producing farmers. In this work, the aim was to utilize a solubilization-based protein-isolation method to produce a low-DSP toxin protein isolate from toxic blue mussels that are unsuitable for the whole shellfish market. A homogenate of whole mussel meat was solubilized at low pH (2.8) or high pH (11.1), followed by centrifugation and reprecipitation of the solubilized mussel proteins at the isoelectric pH. In a second centrifugation, precipitated proteins were collected. These processes resulted in 81 (acid solubilization) and 72% (alkaline solubilization) reduction in the initial DTX-1 toxin content of the mussel meat. No other DSP toxins were-found in the protein isolates. Acid processing of mussel meat resulted in 50% reduction in the total lipid content, while alkaline treatment did not significantly affect the) lipid content. The effect of citric acid and calcium chloride addition to the mussel meat-water homegenate on lipid and toxin content was also investigated. A poor correlation factor was surprisingly obtained between reductions in DTX-1 toxin and lipids in protein isolates from,processed toxic mussels. Results from an analytical mass balance of the DTX-1 toxin during acid processing showed that 61% of this toxin ended up in the aqueous supernatant after the second centrifugation. The present study presents a promising alternative way of utilizing mussels for food production in periods when they are toxic
Σύγκριση σωματοδομής και κινηματικής ρίψης της μπάλας μεταξύ νεαρών ηλικιακών ομάδων της χειροσφαίρισης
Η ρίψη της μπάλας στη χειροσφαίριση αποτελεί μια βασική κινητική δραστηριότητα που έχει ως σκοπό να επιδειχθεί η αποτελεσματικότητα μέσω της επίτευξης τέρματος ή και της σωστής μεταβίβασης. Ως μια σύνθετη βαλλιστική ενέργεια πραγματοποιείται κυρίως διαμέσου της περιστροφικής κίνησης επιμέρους μελών του σώματος και με συνεχώς αυξανόμενη και ελεγχόμενη γωνιακή ταχύτητα (Atwater, 1979). Η εκτέλεσή της βασίζεται σε μια κινηματική αλληλουχία όλων των επιμέρους αρθρώσεων του σώματος, η οποία αρχίζει από τα κάτω άκρα και τον κορμό και τελειώνει στις φάλαγγες των δακτύλων του χεριού ρίψης (Joris et al., 1985).
Η μελέτη της κινηματικής ακολουθίας της ρίψης έχει γίνει κυρίως σε αθλητές του μπέιζμπολ, ενώ στη χειροσφαίριση, οι πληροφορίες είναι λιγοστές και αφορούν στην ταχύτητα της μπάλας. Επιπροσθέτως, φαίνεται ότι υπάρχει διάσταση απόψεων στη σχέση των ανθρωπομετρικών χαρακτηριστικών με την κινηματική της ρίψης, όπως και απουσία μελετών που να εξηγούν την επίδραση της χρονολογικής ηλικίας ή και εμπειρίας στη ρίψη με το κινηματικό πρότυπο ή και την αποτελεσματικότητα αυτής. Σκοπός της μελέτης είναι να εξεταστεί η επίδραση της χρονολογικής ηλικίας στο κινηματικό πρότυπο της ρίψης σε αγόρια 8-17 ετών που ασχολούνται με τη χειροσφαίριση. Ειδικότερα σ’ αυτή τη μελέτη εξετάζονται ερευνητικά ερωτήματα, όπως: α) Εάν διαφοροποιείται η ταχύτητα της μπάλας κατά τη διάρκεια της ρίψης μεταξύ επιμέρους ηλικιακών ομάδων στο εύρος των 8 έως 17 χρόνων; β) Εάν μεταβάλλονται τα κινηματικά χαρακτηριστικά των επιμέρους αρθρώσεων που συμμετέχουν στη ρίψη (ταχύτητα, επιτάχυνση) μεταξύ των διαφορετικών ηλικιακών ομάδων; και γ) Εάν επιμέρους χαρακτηριστικά της σωματομορφίας, όπως οι επιμήκεις διαστάσεις επηρεάζουν το κινηματικό πρότυπο της ρίψης;
Στη μελέτη θα συμμετέχουν 80 αγόρια ηλικίας 8 έως 17 χρόνων, επιλεγμένα με τυχαία δειγματοληψία από σχολεία του Νομού Αττικής. Όλοι οι δοκιμαζόμενοι θα ασχολούνται αθλητικά με τη χειροσφαίριση και θα αγωνίζονται στα αντίστοιχα ηλικιακά πρωταθλήματα ή θα λαμβάνουν μέρος σε οργανωμένα προγράμματα φυσικής αγωγής με κατεύθυνση τη χειροσφαίριση. Οι δοκιμαζόμενοι θα χωριστούν σε τέσσερις διαφορετικές ομάδες (των 20 ατόμων). Τα παιδιά ηλικίας 8-9 και 10-11 χρόνων, θα επιλεγούν από αθλητικά σωματεία, που καλλιεργούν τη χειροσφαίριση σ’ αυτές τις ηλικίες. Οι ηλικιακές ομάδες των 13-14 και 16-17 χρόνων θα επιλεγούν από σχολικές μονάδες οι οποίες έχουν Τμήματα Αθλητικής Διευκόλυνσης χειροσφαίρισης στα Γυμνάσια (ΤΑΔ) και τα Λύκεια (ΕΤΑΔ) του νομού.
Για τη συλλογή του υλικού θα χρησιμοποιηθούν οι μέθοδοι της σωματομετρίας και της βιντεογράφησης. Ειδικότερα, θα γίνουν επιλεγμένες μετρήσεις σωματομορφίας με αναφορά σε επιμήκεις σωματικές διαστάσεις, περιφέρειες οστών, διακονδυλικά πλάτη οστών και δερματικές πτυχές (Heyward & Stolarczyk, 1996). Επιπλέον, ο κάθε δοκιμαζόμενος θα εκτελέσει τρεις έως πέντε ρίψεις της μπάλας με συγκεκριμένο στόχο, οι οποίες θα καταγραφούν από 2 βιντεοκάμερες RedLake με συχνότητα δειγματοληψίας 125 Hz. Η συλλογή και η επεξεργασία των κινηματικών δεδομένων θα γίνει μέσω λογισμικού προγράμματος Peak Motus (Peak Performance Technοlogies, Inc.), προκειμένου να γίνει ανάλυση και των γραμμικών συνισταμένων των ταχυτήτων του ισχίου, ώμου, αγκώνα και καρπού. Για τη στατιστική ανάλυση των δεδομένων θα χρησιμοποιηθούν δείκτες μέσου επιπέδου και διασποράς, ενώ ο σχεδιασμός θα βασιστεί κυρίως α) στην απλή ανάλυση διακύμανσης με ανεξάρτητη την ηλικία και έλεγχος αυτής, όπως και β) στην ανάλυση συσχέτισης κατά Pearson (r), με επίπεδο σημαντικότητας p<.05
Vitamin D<sub>3</sub> Bioaccessibility from Supplements and Foods—Gastric pH Effect Using a Static In Vitro Gastrointestinal Model
Vitamin D3 deficiency is a global phenomenon, which can be managed with supplementation and food fortification. However, vitamin D3 bioaccessibility may depend on factors such as matrix composition and interactions throughout the gastrointestinal (GI) tract. This research focused on the effect of different matrices on vitamin D3 content during digestion, as well as the effect of pH on its bioaccessibility. The INFOGEST protocol was employed to simulate digestion. Three different types of commercial supplements, two foods naturally rich in vitamin D3, and three fortified foods were investigated. High-Performance Liquid Chromatography was used to determine the initial vitamin D3 content in the supplements and foods, as well as after each digestion stage. The results indicate that the foods exhibited higher bioaccessibility indices compared to the supplements and a higher percentage retention at the end of the gastric phase. The pH study revealed a positive correlation between an increased gastric pH and the corresponding content of vitamin D3. Interestingly, exposing the matrix to a low pH during the gastric phase resulted in an increased intestinal content of D3. Vitamin D3 is more bioaccessible from foods than supplements, and its bioaccessibility is susceptible to changes in gastric pH. Fasting conditions (i.e., gastric pH = 1) enhance the vitamin’s bioaccessibility
