The use of a conjugate of colloidal gold with monotypic antibodies against a neutral Mn(2+)-dependent DNAse has revealed the enzyme localization in ultrathin Epon sections of glutaraldehyde fixed tissues. Technical procedures involved in fixation, embedding, and immune reactions are described. DNAse has been established in the nuclei in both normal and regenerating liver. A protein, immunologically close to DNAse, has been identified in the nuclei of a lymph node, thymus, spleen and cerebellar cortex

Neutral Mn(2+)-dependent DNAse is localized on isolated chromatin structures in both normal and regenerating rat liver. The enzyme was revealed located along the whole length of nucleosomal chain and in hypernucleosomal structures. However, as concerns the quantity of the enzyme, it was distributed unevently along the chromatin, thus reflecting the pattern of different functional states of native chromatin. According to biochemical and immunohistochemical data, DNAse can hydrolyse in vitro only one-stranded DNA. One of possible explanations of the observed differences in DNAse binding with native DNA chromatin and its inability to adsorb on native DNA in vitro may be the presence of hypothetical DNA-binding proteins in native chromatin making complexes with DNAse and thereby responsible for immobilization of the enzyme on chromatin structures in vivo

Use of approximations of Hamilton-Jacobi-Bellman inequality for solving periodic optimization problems

We show that necessary and sufficient conditions of optimality in periodic optimization problems can be stated in terms of a solution of the corresponding HJB inequality, the latter being equivalent to a max-min type variational problem considered on the space of continuously differentiable functions. We approximate the latter with a maximin problem on a finite dimensional subspace of the space of continuously differentiable functions and show that a solution of this problem (existing under natural controllability conditions) can be used for construction of near optimal controls. We illustrate the construction with a numerical example.Comment: 29 pages, 2 figure

The Patient’s and the Therapist’s Evaluation of Bridges of Different Materials and Age

The aim of this study was to find out patients\u27 satisfaction with their bridges made of different materials (metal-ceramics, Au/resin, Ag-Pd/resin). One hundred and sixty four patients were examined at the Dental School, University of Zagreb, Croatia. They assessed their bridges – the overall quality, aesthetics, speech, chewing and the health of the gingiva by the scale from 1–5. The same categories were also assessed by a trained prosthodontist. The majority of the patients was really satisfied and gave the highest grades (quality, aesthetics, speech, etc.) and therefore the results were skewed and asymmetrical towards the biggest scores (biggest grades). The best gingival health was evaluated by the group of patients with ceramic crowns and bridges (p < 0.05) and the worst by the patients with Ag-Pd bridges. Speech was scored higher for the lateral than for the frontal bridges. Patients evaluated the health of the tissue surrounding their bridges, overall quality of fixed prosthodontic appliance and aesthetics with significantly higher scores than the prosthodontist (p < 0.01). The results point at a difference between the patient’s and the therapist\u27s evaluations and to the patient\u27s insufficient care about the gingiva around the bridge abutments

Properties of deoxyribonuclease of rat liver nuclei and changes in its activity during induced synthesis of nucleic acids

DNAase from the proteins of chromatin obtained from rat livers preferentially hydrolyzes denatured DNA on the 3' phosphodiester bonds, forming oligonucleotides consisting of 6 to 8 monomers. The DNAase is not specific with respect to the bases. During the induced synthesis of DNA and RNA in rat liver cells a decrease was noted in the DNAase activity of the chromatin proteins

Towards two-dimensional metallic behavior at LaAlO3/SrTiO3 interfaces

Using a low-temperature conductive-tip atomic force microscope in cross-section geometry we have characterized the local transport properties of the metallic electron gas that forms at the interface between LaAlO3 and SrTiO3. At low temperature, we find that the carriers do not spread away from the interface but are confined within ~10 nm, just like at room temperature. Simulations taking into account both the large temperature and electric-field dependence of the permittivity of SrTiO3 predict a confinement over a few nm for sheet carrier densities larger than ~6 10^13 cm-2. We discuss the experimental and simulations results in terms of a multi-band carrier system. Remarkably, the Fermi wavelength estimated from Hall measurements is ~16 nm, indicating that the electron gas in on the verge of two-dimensionality.Comment: Accepted for publication in Physical Review Letter

Special features of the DNA-hydrolyzing activity of the antibodies in systemic lupus erythematosus

    Get PDF
Two types of IgG anti-DNA antibodies exhibiting DNA-hydrolyzing activity have been isolated from blood serum of patients with systemic lupus erythematosus. This DNase activity of antibodies differs from serum DNases by the non-processive mode, temperature resistance, pH optimum, and the rate of DNA hydrolysis. It is suggested that the anti-DNA antibody molecule possessing DNase activity contains two sites: one site determines specificity of antibody-DNA interaction, whereas the other is responsible for manifestation of the catalytic activity

Rabbit antibodies against a neutral Mn(2+)-dependent rat liver DNAse were obtained, whose specificity towards DNAse was ascertained by suppression of the enzyme activity both in vitro system and immunoblotting assays. Procedures of synthesis of ferritin and colloidal gold conjugates with antibodies are described. The biological activity of the conjugates proved to be similar to that of the original antibodies
