    Involvement of pro-inflammatory cytokines and growth factors in the pathogenesis of Dupuytren's contracture: a novel target for a possible future therapeutic strategy?

    Dupuytren's contracture (DC) is a benign fibro-proliferative disease of the hand causing fibrotic nodules and fascial cords which determine debilitating contracture and deformities of fingers and hands. The present study was designed to characterize pro-inflammatory cytokines and growth factors involved in the pathogenesis, progression and recurrence of this disease, in order to find novel targets for alternative therapies and strategies in controlling DC. The expression of pro-inflammatory cytokines and of growth factors was detected by immunohistochemistry in fibrotic nodules and normal palmar fascia resected respectively from patients affected by DC and carpal tunnel syndrome (CTS; as negative controls). Reverse transcription (RT)-PCR analysis and immunofluorescence were performed to quantify the expression of transforming growth factor (TGF)-β1, interleukin (IL)-1β and vascular endothelial growth factor (VEGF) by primary cultures of myofibroblasts and fibroblasts isolated from Dupuytren's nodules. Histological analysis showed high cellularity and high proliferation rate in Dupuytren's tissue, together with the presence of myofibroblastic isotypes; immunohistochemical staining for macrophages was completely negative. In addition, a strong expression of TGF-β1, IL-1β and VEGF was evident in the extracellular matrix and in the cytoplasm of fibroblasts and myofibroblasts in Dupuytren's nodular tissues, as compared with control tissues. These results were confirmed by RT-PCR and by immunofluorescence in pathological and normal primary cell cultures. These preliminary observations suggest that TGF-β1, IL-1β and VEGF may be considered potential therapeutic targets in the treatment of Dupuytren's disease (DD)

    Stress management interventions among healthcare workers using mindfulness. A systematic review.

    Background: stress among healthcare professionals has gained more and more attention due to the negative consequences on their and patients’ health. As a result of intense working hours, night shifts, responsibilities of care, and emotional contact with patients, healthcare workers experience stressful conditions. Interventions to prevent and manage their wellbeing are needed, in order to reduce the risks of onset of burnout syndrome. Aim: the aim of this systematic review is to analyze how mindfulness courses can improve mental and physical well-being of healthcare workers. Methods: a literature search was conducted in May 2018 using the databases Medline (PubMed), Scopus and Isi Web of Knowledge. Studies were included if they examined mindfulness interventions as possible solutions to manage stress among healthcare workers. Results: fifty-eight articles met the inclusion criteria: 13 of them were clinical trials; 11 were randomized clinical trials; 12 were systematic reviews; 7 were narrative reviews and 15 were observational studies. The studies included showed effectiveness of mindfulness programs in reducing stress, self-compassion, burnout, anxiety and depression. Significant negative association has been observed between MBSR and stress levels (β: −0.60, 95% CI:−5.95 to −4.04, P< 0.001) and mental exhaustion (β:−0.43, 95% CI:−3.30 −1.86, P< 0.001). Clinical trials focused on psychoeducational interventions highlighted decreased burnout scores in intervention group (SDM: -0.38). Conclusion: courses based on mindfulness showed to be effective in improving healthcare workers’ well-being, increasing their quality of life and the productivity outcomes. The evidence derived from this systematic review suggests that these interventions should be included within the work organization in order to be viable tools for promoting self-care and quality of care

    Z' effects and anomalous gauge couplings at LC with polarization

    We show that the availability of longitudinally polarized electron beams at a 500 GeV Linear Collider would allow, from an analysis of the reaction e^+e^-\to W^+W^-, to set stringent bounds on the couplings of a Z' of the most general type. In addition, to some extent, it would be possible to disentangle observable effects of the Z' from analogous ones due to competitor models with anomalous trilinear gauge couplings.Comment: 22 pages LaTex, 6 figures available on request, revised version accepted for publication in Int.     Pseudo Goldstones at Future Colliders from the Extended Bess Model

    We consider the production of the lightest pseudo-Goldstone bosons at future colliders through the vector resonances predicted by the extended BESS model, which consists of an effective lagrangian parametrization with dynamical symmetry breaking, describing scalar, vector and axial-vector bound states in a rather general framework. We find that the detection of pseudo-Goldstone pairs at LHC requires a careful evaluation of backgrounds. For e+e- collisions in the TeV range the backgrounds can be easily reduced and the detection of pseudo-Goldstone pairs is generally easier.Comment: 17 pages and 12 figures (included as a uuencoded tar file), LaTeX (style article), UGVA-DPT 1994/03-84

    Extraction of the pion distribution amplitude from polarized muon pair production

    We consider the production of muon pairs from the scattering of pions on longitudinally polarized protons. We calculate the cross section and the single spin asymmetry for this process, taking into account pion bound state effects. We work in the kinematic region where the photon has a large longitudinal momentum fraction, which allows us to treat the bound state problem perturbatively. Our predictions are directly proportional to the pion distribution amplitude.     An Elusive Z' Coupled to Beauty

    By extending the standard gauge group to SU(3)_c \times SU(2)_L \times U(1)_Y \times U(1)_X with X charges carried only by the third family we accommodate the LEP measurement of R_b and predict a potentially measurable discrepancy in A_{FB}^{b} in e^+e^- scattering and that D^0\bar{D}^0 mixing may be near its experimental limit. The Z', which explicitly violates the GIM mechanism, can nevertheless be naturally consistent with FCNC constraints.     Angiopoietin-like protein 4 overexpression in visceral adipose tissue from obese subjects with impaired glucose metabolism and relationship with lipoprotein lipase

    Angiopoietin-like protein 4 (ANGPTL4) regulates lipid partitioning by inhibiting circulating and tissue lipoprotein lipase (LPL); ANGPTL4 loss-of-function variants improve insulin sensitivity and reduce type 2 diabetes (T2D) risk with mechanisms partially unknown. This study was designed to explore metabolic implications of differential ANGPTL4 and LPL expression in human adipose tissue (AT). We recruited eighty-eight obese individuals, with and without abnormal glucose metabolism (AGM), undergoing bariatric surgery; visceral AT (VAT) fragments were obtained intra-operatively and analyzed by immunohistochemistry and mRNA by rt-PCR. Data on hepatic ANGPTL4 mRNA were available for 40 participants. VAT ANGPTL4 expression was higher in AGM individuals than in those with normal glucose tolerance (NGT) and associated with VAT inflammation, insulin resistance, and presence of adipocyte size heterogeneity. Increased ANGPTL4 was associated with AGM with OR = 5.1 (95% C.I.: 1.2–23; p = 0.02) and AUROC = 0.76 (95% C.I.: 1.2–23; p < 0.001). High LPL was associated with the detection of homogeneous adipocyte size, reduced microvessel density, and higher HIF-1α levels and inversely correlated to blood transaminases. In conclusion, in obese individuals, VAT ANGPTL4 levels are increased in the presence of local inflammation and AGM. Conversely, higher LPL expression describes a condition of increased lipid storage in adipocytes, which may serve as a protective mechanism against ectopic fat accumulation and related metabolic disease in obesity
