A new variety of Plocama calabrica (Rubiaceae) from Denizli (Turkey) onfirmed by morphological and molecular ISSR markers
Plocama calabrica (L.f.) M.Backlund & Thulin var. alba Göktürk, O.D.Düţen, B.Gürcan & U. Sarpkaya variety nova is described from South-West Anatolia. The new variety grows on limestone slopes between Akpýnar and Yaylapýnar villages in the Çameli district in Denizli. It is closely related to P. calabrica var. calabrica, and can be readily distinguished by morphological and molecular characters from the related variety. Taxonomic comments such as descriptive and diagnostic characters, distribution and ecology, phenology and proposed conservation status for this new variety are given in the current study. A taxonomic revision of the genus potentilla l. (group d) (rosaceae) in Turkey
Bu çalışma kapsamında, Türkiye’de yayılış gösteren Potentilla L. cinsinin
D grubuna ait 16 tür ve 18 takson üzerinde morfolojik ve mikromorfolojik
araştırmalar ile birlikte taksonomik açıdan problemli bazı türler üzerinde
moleküler ve bir tür üzerinde de fitokimyasal çalışmalar yapılmıştır.
Morfolojik çalışmalar kapsamında, farklı herbaryumlarda bulunan yerli ve
yabancı botanikçilerce toplanan örnekler ile tez aşaması sırasında tarafımızdan
toplanan çok sayıda örnekten yararlanılmıştır. Çalışma sonucunda, Türkiye
Florası’nda bu cinse ait türlerin betimlerinde yer verilmeyen ve türler arasında
farklılıklar gösteren yeni özellikler tespit edilmiş ve bu özelliklerin de ilave
edilmesi ile D grubu Potentilla taksonlarına ait detaylı morfolojik betimler ve
daha kullanışlı tür teşhis anahtarı hazırlanmıştır. Bununla beraber türlerin mevcut
yayılış alanlarına ek olarak tarafımızca yeni yayılış alanları da eklenmiştir.
Mikromorfolojik çalışmalar kapsamında polen, tüy ve meyve
mikromorfolojileri çalışılmıştır. D grubu Potentilla taksonlarına ait polen, tüy ve
meyve yapıları Taramalı Elektron Mikroskobu (SEM) kullanılmak suretiyle
detaylı olarak tanımlanmıştır.
Ayrıca tez kapsamında taksonomik açıdan problemli olan ve morfolojik
olarak net ayrımı yapılamayan P. libanotica ve P. isaurica türleri üzerinde
moleküler çalışmalar yapılmıştır. Bu çalışma sonucunda P. isaurica türünün ayrı
bir tür olamayacağı ve P. libanotica türünün alttürü olduğu sonucuna varılmıştır.
Bunların yanı sıra Denizli’ye özgü bir endemik olan P. davisii türünden
elde edilen aseton ve metanol ekstraktlarının antioksidan kapasitesi, toplam
sekonder metabolitleri ve yüksek performanslı sıvı kromatografisi (HPLC)
kullanılmak suretiyle fenolik bileşimleri tespit edilmiştir.In this study, Group D of Potentilla L. genus of which includes 16 species
and 18 taxa in Turkey on morphology and micromorphology studies with
molecular studies on the some taxonomic problematic species and phytochemical
studies on one species were conducted.
In the morphological studies, samples collected by local and foreign
botanists in different herbariums and many samples collected by us during the
thesis phase were used. As a result of this study new features were determined
which had previously not existed in the descriptions of Potentilla species in Flora
of Turkey. With the addition of these features, detailed morphological
descriptions of Potentilla taxa and a more useful species identification key were
prepared. However, in addition to the existing distribution areas of the species,
new distribution areas have been added by us.
In micromorphological studies, pollen, hair and fruit micromorphologies
were studied. Pollen, hair and fruit structures of Potentilla taxa of group D were
described in detail using Scanning Electron Microscope (SEM).
In addition, molecular studies were carried out on P. libanotica and P.
isaurica species, which are taxonomically problematic and cannot be clearly
distinguished morphologically. As a result of this study, it was concluded that P.
isaurica cannot be a separate species and is a subspecies of P. libanotica.
Also, antioxidant capacity, total secondary metabolites and phenolic
compositions of acetone and methanol extracts obtained from P. davisii which is
endemic to Denizli were determined by using high performance liquid
