    The Role Of Literal Meaning In Proverb Comprehension

    According to the \u27Standard Pragmatic Model of Language\u27, a person initially attempts to understand figurative sentences such as metaphors and proverbs in terms of their literal meaning and only processes the figurative meaning when the literal meaning is found inadequate. Experimental studies have failed to support this model; however, many of these studies confounded literalness with conventionality. Furthermore, the role of literal meaning during figurative language comprehension remains unresolved. The purpose of this thesis is to conduct a more valid test of the Standard Pragmatic Model, and to determine the role of literal meaning during proverb comprehension. In the first experiment it was shown that unfamiliar proverbs used figuratively take longer to read compared to unfamiliar proverbs used literally and compared to literal paraphrases of the proverbs\u27 figurative meaning. In contrast, literal and figurative uses of familiar proverbs were read at the same speed. In the second and third studies it was shown that cues related to the literal meaning of a proverb were more effective recall aids for proverbs than were cues related to the figurative meaning of a proverb; however, cues related to the figurative meaning were effective if the proverb was familiar, or if the proverb was used figuratively. These data suggest that literal meaning plays an important role in proverb processing. Experiments four and five tried to determine if literal meaning leads to the comprehension of the abstract proverbial meaning with some use of either (1) verbal associative processes or (2) mental imagery processes. Participants read the proverbs in context while memorizing either a concurrent verbal or visual-spatial pattern. The results, although weak, suggest that proverb processing is more affected by a visual than a verbal distraction task and that reading a paraphrase of the figurative meaning is more affected by a verbal task. Taken together these data are evidence for a modified version of the Standard Pragmatic Model in which both conventional and literal meanings are processed in an obligatory manner. In addition, the results suggest that a proverb\u27s literal meaning is used to generate a mental image that may play a role in proverb processing

    Interventions for Problem Gamblers in the Correctional System

    The prevalence rates of problem gambling in the adult correctional population are 5 to 10 times higher than those found in the general population. The purpose of this study is to examine the type of interventions used to deal with this population. We conducted a combination of literature review and key informant study to examine the state of programming for problem gambling for clients who experience incarceration. The published literature on this topic is remarkably sparse. In fact only two peer reviewed published studies were identified that formally evaluated a treatment program for problem gambling for clients in these settings. However, we uncovered a small number of programs (10) that have been developed for and delivered to this population including a gambling treatment court in Buffalo, Gamblers Anonymous, outpatient treatment during probation or parole, brief psycho-educational programs, a full treatment program, and inpatient treatment after release. Although some programs have been delivered within correctional institution, others have been offered either after release or prior to sentencing. A major issue is the lack of awareness of problem gambling in the criminal justice system among judges, lawyers, wardens, corrections workers, parole officers and the gamblers themselves. The results are discussed in terms of stigma, barriers, issues, and opportunities for programming for this population

    Type 2 diabetes is a metabolic disorder characterized by the inability of beta-cells to secrete enough insulin to maintain glucose homeostasis. MIN6 cells secrete insulin in response to glucose and other secretagogues, but high passage (HP) MIN6 cells lose their ability to secrete insulin in response to glucose. We hypothesized that metabolism of glucose and lipids were defective in HP MIN6 cells causing impaired glucose stimulated insulin secretion (GSIS). HP MIN6 cells had no first phase and impaired second phase GSIS indicative of global functional impairment. This was coupled with a markedly reduced ATP content at basal and glucose stimulated states. Glucose uptake and oxidation were higher at basal glucose but ATP content failed to increase with glucose. HP MIN6 cells had decreased basal lipid oxidation. This was accompanied by reduced expressions of Glut1, Gck, Pfk, Srebp1c, Ucp2, Sirt3, Nampt. MIN6 cells represent an important model of beta cells which, as passage numbers increased lost first phase but retained partial second phase GSIS, similar to patients early in type 2 diabetes onset. We believe a number of gene expression changes occurred to produce this defect, with emphasis on Sirt3 and Nampt, two genes that have been implicated in maintenance of glucose homeostasis.These authors have no support or funding to report

    Functionalized micro-capillary film for the rapid at-line analysis of IgG aggregates in a cell culture bioreactor.

    A micro-capillary film has been developed that offers the potential for an at-line analytical tool for rapid aggregate analysis during biopharmaceutical antibody production. A non-porous walled micro-capillary film (NMCF) with cation exchange functionality was demonstrated to act as a chromatography medium that could be operated with high linear fluid velocities and was highly resistant to blockage by entrained particulates, including cells. The NMCF containing 19 parallel microcapillaries was prepared using a melt extrusion process from poly(ethylene-vinyl alcohol) copolymer (EVOH). The NMCF-EVOH was modified to have cation-exchange functionality (NMCF-EVOH-SP) and shown to differentially bind monomer and aggregated species of IgG antibody directly from a bioreactor. The use of NMCF-EVOH-SP to quantify aggregate concentrations in monoclonal antibody preparations in less than 20 minutes was demonstrated.The authors would like to thank the EPSRC for the provision of a CASE Award.     IRS1-Independent Defects Define Major Nodes of Insulin Resistance

    SummaryInsulin resistance is a common disorder caused by a wide variety of physiological insults, some of which include poor diet, inflammation, anti-inflammatory steroids, hyperinsulinemia, and dyslipidemia. The common link between these diverse insults and insulin resistance is widely considered to involve impaired insulin signaling, particularly at the level of the insulin receptor substrate (IRS). To test this model, we utilized a heterologous system involving the platelet-derived growth factor (PDGF) pathway that recapitulates many aspects of insulin action independently of IRS. We comprehensively analyzed six models of insulin resistance in three experimental systems and consistently observed defects in both insulin and PDGF action despite a range of insult-specific defects within the IRS-Akt nexus. These findings indicate that while insulin resistance is associated with multiple deficiencies, the most deleterious defects and the origin of insulin resistance occur independently of IRS

    Identification of fatty acid binding protein 4 as an adipokine that regulates insulin secretion during obesity.

    A critical feature of obesity is enhanced insulin secretion from pancreatic β-cells, enabling the majority of individuals to maintain glycaemic control despite adiposity and insulin resistance. Surprisingly, the factors coordinating this adaptive β-cell response with adiposity have not been delineated. Here we show that fatty acid binding protein 4 (FABP4/aP2) is an adipokine released from adipocytes under obesogenic conditions, such as hypoxia, to augment insulin secretion. The insulinotropic action of FABP4 was identified using an in vitro system that recapitulates adipocyte to β-cell endocrine signalling, with glucose-stimulated insulin secretion (GSIS) as a functional readout, coupled with quantitative proteomics. Exogenous FABP4 potentiated GSIS in vitro and in vivo, and circulating FABP4 levels correlated with GSIS in humans. Insulin inhibited FABP4 release from adipocytes in vitro, in mice and in humans, consistent with feedback regulation. These data suggest that FABP4 and insulin form an endocrine loop coordinating the β-cell response to obesity

    Mathematical modelling long-term effects of replacing Prevnar7 with Prevnar13 on invasive pneumococcal diseases in England and Wales

    England and Wales recently replaced the 7-valent pneumococcal conjugate vaccine (PCV7) with its 13-valent equivalent (PCV13), partly based on projections from mathematical models of the long-term impact of such a switch compared to ceasing pneumococcal conjugate vaccination altogether. A compartmental deterministic model was used to estimate parameters governing transmission of infection and competition between different groups of pneumococcal serotypes prior to the introduction of PCV13. The best-fitting parameters were used in an individual based model to describe pneumococcal transmission dynamics and effects of various options for the vaccination programme change in England and Wales. A number of scenarios were conducted using (i) different assumptions about the number of invasive pneumococcal disease cases adjusted for the increasing trend in disease incidence prior to PCV7 introduction in England and Wales, and (ii) a range of values representing serotype replacement induced by vaccination of the additional six serotypes in PCV13. Most of the scenarios considered suggest that ceasing pneumococcal conjugate vaccine use would cause an increase in invasive pneumococcal disease incidence, while replacing PCV7 with PCV13 would cause an overall decrease. However, the size of this reduction largely depends on the level of competition induced by the additional serotypes in PCV13. The model estimates that over 20 years of PCV13 vaccination, around 5000–62000 IPD cases could be prevented compared to stopping pneumococcal conjugate vaccination altogether. Despite inevitable uncertainty around serotype replacement effects following introduction of PCV13, the model suggests a reduction in overall invasive pneumococcal disease incidence in all cases. Our results provide useful evidence on the benefits of PCV13 to countries replacing or considering replacing PCV7 with PCV13, as well as data that can be used to evaluate the cost-effectiveness of such a switch
