    An unstructured CD-grid variational formulation for sea ice dynamics

    For the numerical simulation of earth system models, Arakawa grids are largely employed. A quadrilateral mesh is assumed for their classical definition, and different types of grids are identified depending on the location of the discretized quantities. The B-grid has both velocity components at the center of a cell, the C-grid places the velocity components on the edges in a staggered fashion, and the D-grid is a ninety-degree rotation of a C-grid. Historically, B-grid formulations of sea ice dynamics have been dominant because they have matched the grid type used by ocean models. In recent years, as ocean models have increasingly progressed to C-grids, sea ice models have followed suit on quadrilateral meshes, but few if any implementations of unstructured C-grid sea ice models have been developed. In this work, we present an unstructured CD-grid type formulation of the elastic-viscous-plastic rheology, where the velocity unknowns are located at the edges, rather than at the vertices, as in the B-grid. The notion of a CD-grid has been recently introduced and assumes that the velocity components are co-located at the edges. The mesh cells in our analysis have nn sides, with nn greater than or equal to four. Numerical results are included to investigate the features of the proposed method. Our framework of choice is the Model for Prediction Across Scales (MPAS) within E3SM, the climate model of the U.S. Department of Energy, although our approach is general and could be applied to other models as well. While MPAS-Seaice is currently defined on a B-grid, MPAS-Ocean runs on a C-grid, hence interpolation operators are heavily used when coupled simulations are performed. The discretization introduced here aims at transitioning the dynamics of MPAS-Seaice to a CD-grid, to ultimately facilitate improved coupling with MPAS-Ocean and reduce numerical errors associated with this communication

    Organic and soil material between tills in east‐midland England – direct evidence for two episodes of lowland glaciation in Britain during the Middle Pleistocene

    This paper provides a record and analysis of a site in east-midland England, at which organic and soil material are found between two Middle Pleistocene tills. This is the first discovery of its kind in the area, and demonstrates unequivocally that the region was glaciated on two separate occasions, something that has long been inferred and articulated, but not actually demonstrated. The landforms, sediments and soils are studied with respect to their geomorphological, lithological, pedological, palaeobotanical and structural properties. The organic and soil material along with soil structures indicate, sequentially, a periglacial climate, a long period of warm temperate weathering and a cool temperate climate. Evaluation of this evidence in terms of existing published work identifies a number of problems with existing models and suggests that the most likely model for the glacial history of this part of midland England is an early Middle Pleistocene glaciation which is represented only by trace erratics, a Marine Isotope Stage (MIS) 12 age glaciation which moved across the area from the NW and deposited a chalk-free till, and an MIS 8 age glaciation that transported and deposited an upper chalky till from the NE

    Methods used for successful follow-up in a large scale national cohort study in Thailand

    Background: Ensuring successful follow-up is essential when conducting a prospective cohort study. Most existing literature reviewing methods to ensure a high response rate is based on experience in developed nations. Findings: We report our 4-year follow-up success for a national cohort study examining the health transition underway in Thailand. We began the cohort study in 2005 with a baseline postal questionnaire sent to all 200,000 Thais enrolled as distance learning students at Sukhothai Thammathirat Open University and residing all over Thailand; 87,134 or 44% of the students responded. Subsequently we used University and national media to inform cohort members of study progress. Also, we prepared a health book with study results and health advice which was distributed to all cohort members. After 4 years we repeated the survey and achieved a 71% response rate. In this paper we report the methods used to achieve this response The initial follow-up mail-out generated a response rate of about 48% reflecting the extensive preparatory work between baseline and follow-up. After 4 rounds of telephone contact (more than 100,000 phone calls) and 4 related mail-out rounds progressively over 16 months an overall response rate was achieved of just over 71% (n = 60,774). The total cost was US$4.06/respondent - 19% for printing, 21% for postage, 14% for tape measures (included in mail-out), 18% for data processing 22% for prizes and 6% for telephone. Conclusions: Many of the methods reported as effective for mail questionnaire and cohort response rates held true for Thailand. These included being associated with a university, incentivating cooperation, follow-up contact, providing a second copy of questionnaire where necessary, and assurance of confidentiality. Telephone contact with the cohort and the small prizes given to responders were particularly important in the Thai context as was Thai leadership of the research team

    Gross solids from combined sewers in dry weather and storms, elucidating production, storage and social factors

    Variation in rates of sanitary hygiene products, toilet tissue and faeces occurring in sewers are presented for dry and wet weather from three steep upstream urban catchments with different economic, age and ethnic profiles. Results show, for example, that total daily solids per capita from the low income and ageing populations are almost twice that from high income or ethnic populations. Relative differences are verified through independent questionnaires. The relationship between solids stored in sewers prior to storms, antecedent dry weather period and the proportion of roof to total catchment area is quantified. A full solids' flush occurs when storm flows exceed three times the peak dry weather flow. The data presented will assist urban drainage designers in managing pollution caused by the discharge of sewage solids

    Influence of Comorbidities on Therapeutic Progression of Diabetes Treatment in Australian Veterans: A Cohort Study

    BACKGROUND: This study assessed whether the number of comorbid conditions unrelated to diabetes was associated with a delay in therapeutic progression of diabetes treatment in Australian veterans. METHODOLOGY/PRINCIPAL FINDINGS: A retrospective cohort study was undertaken using data from the Australian Department of Veterans' Affairs (DVA) claims database between July 2000 and June 2008. The study included new users of metformin or sulfonylurea medicines. The outcome was the time to addition or switch to another antidiabetic treatment. The total number of comorbid conditions unrelated to diabetes was identified using the pharmaceutical-based comorbidity index, Rx-Risk-V. Competing risk regression analyses were conducted, with adjustments for a number of covariates that included age, gender, residential status, use of endocrinology service, number of hospitalisation episodes and adherence to diabetes medicines. Overall, 20134 veterans were included in the study. At one year, 23.5% of patients with diabetes had a second medicine added or had switched to another medicine, with 41.4% progressing by 4 years. The number of unrelated comorbidities was significantly associated with the time to addition of an antidiabetic medicine or switch to insulin (subhazard ratio [SHR] 0.87 [95% CI 0.84–0.91], P<0.001). Depression, cancer, chronic obstructive pulmonary disease, dementia, and Parkinson's disease were individually associated with a decreased likelihood of therapeutic progression. Age, residential status, number of hospitalisations and adherence to anti-diabetic medicines delayed therapeutic progression. CONCLUSIONS / SIGNIFICANCE: Increasing numbers of unrelated conditions decreased the likelihood of therapeutic progression in veterans with diabetes. These results have implications for the development of quality measures, clinical guidelines and the construction of models of care for management of diabetes in elderly people with comorbidities.Agnes I.     An interactome-centered protein discovery approach reveals novel components involved in mitosome function and homeostasis in giardia lamblia

    Protozoan parasites of the genus Giardia are highly prevalent globally, and infect a wide range of vertebrate hosts including humans, with proliferation and pathology restricted to the small intestine. This narrow ecological specialization entailed extensive structural and functional adaptations during host-parasite co-evolution. An example is the streamlined mitosomal proteome with iron-sulphur protein maturation as the only biochemical pathway clearly associated with this organelle. Here, we applied techniques in microscopy and protein biochemistry to investigate the mitosomal membrane proteome in association to mitosome homeostasis. Live cell imaging revealed a highly immobilized array of 30–40 physically distinct mitosome organelles in trophozoites. We provide direct evidence for the single giardial dynamin-related protein as a contributor to mitosomal morphogenesis and homeostasis. To overcome inherent limitations that have hitherto severely hampered the characterization of these unique organelles we applied a novel interaction-based proteome discovery strategy using forward and reverse protein co-immunoprecipitation. This allowed generation of organelle proteome data strictly in a protein-protein interaction context. We built an initial Tom40-centered outer membrane interactome by co-immunoprecipitation experiments, identifying small GTPases, factors with dual mitosome and endoplasmic reticulum (ER) distribution, as well as novel matrix proteins. Through iterative expansion of this protein-protein interaction network, we were able to i) significantly extend this interaction-based mitosomal proteome to include other membrane-associated proteins with possible roles in mitosome morphogenesis and connection to other subcellular compartments, and ii) identify novel matrix proteins which may shed light on mitosome-associated metabolic functions other than Fe-S cluster biogenesis. Functional analysis also revealed conceptual conservation of protein translocation despite the massive divergence and reduction of protein import machinery in Giardia mitosomes
