RHQT Nb3Al 15-Tesla magnet design study
Feasibility study of 15-Tesla dipole magnets wound with a new copper stabilized RHQT Nb{sub 3}Al Rutherford cable is presented. A new practical long copper stabilized RHQT Nb{sub 3}Al strand is presented, which is being developed and manufactured at the National Institute of Material Science (NIMS) in Japan. It has achieved a non-copper J{sub c} of 1000A/mm{sup 2} at 15 Tesla at 4.2K, with a copper over non-copper ratio of 1.04, and a filament size less than 50 microns. For this design study a short Rutherford cable with 28 Nb{sub 3}Al strands of 1 mm diameter will be fabricated late this year. The cosine theta magnet cross section is designed using ROXIE, and the stress and strain in the coil is estimated and studied with the characteristics of the Nb{sub 3}Al strand. The advantages and disadvantages of the Nb{sub 3}Al cable are compared with the prevailing Nb{sub 3}Sn cable from the point of view of stress-strain, J{sub c}, and possible degradation of stabilizer due to cabling. The Nb{sub 3}Al coil of the magnet, which will be made by wind and react method, has to be heat treated at 800 degree C for 10 hours. As preparation for the 15 Tesla magnet, a series of tests on strand and Rutherford cables are considered
Cathepsin E Deficiency Impairs Autophagic Proteolysis in Macrophages
Cathepsin E is an endosomal aspartic proteinase that is predominantly expressed in immune-related cells. Recently, we showed that macrophages derived from cathepsin E-deficient (CatE-/-) mice display accumulation of lysosomal membrane proteins and abnormal membrane trafficking. In this study, we demonstrated that CatE-/- macrophages exhibit abnormalities in autophagy, a bulk degradation system for aggregated proteins and damaged organelles. CatE-/- macrophages showed increased accumulation of autophagy marker proteins such as LC3 and p62, and polyubiquitinated proteins. Cathepsin E deficiency also altered autophagy-related signaling pathways such as those mediated by the mammalian target of rapamycin (mTOR), Akt, and extracellular signal-related kinase (ERK). Furthermore, immunofluorescence microscopy analyses showed that LC3-positive vesicles were merged with acidic compartments in wild-type macrophages, but not in CatE-/- macrophages, indicating inhibition of fusion of autophagosome with lysosomes in CatE-/- cells. Delayed degradation of LC3 protein was also observed under starvation-induced conditions. Since the autophagy system is involved in the degradation of damaged mitochondria, we examined the accumulation of damaged mitochondria in CatE-/- macrophages. Several mitochondrial abnormalities such as decreased intracellular ATP levels, depolarized mitochondrial membrane potential, and decreased mitochondrial oxygen consumption were observed. Such mitochondrial dysfunction likely led to the accompanying oxidative stress. In fact, CatE-/- macrophages showed increased reactive oxygen species (ROS) production and up-regulation of oxidized peroxiredoxin-6, but decreased antioxidant glutathione. These results indicate that cathepsin E deficiency causes autophagy impairment concomitantly with increased aberrant mitochondria as well as increased oxidative stress
Impaired chemotaxis and cell adhesion due to decrease in several cell-surface receptors in cathepsin E-deficient macrophages.
Cathepsin E is an endo-lysosomal aspartic proteinase exclusively present in immune system cells. Previous studies have shown that cathepsin E-deficient (CatE(-/-)) mice display aberrant immune responses such as atopic dermatitis and higher susceptibility to bacterial infection. However, the mechanisms underlying abnormal immune responses induced by cathepsin E deficiency are still unclear. In this study, we found that the cell-surface levels of chemotactic receptors, including chemokine receptor (CCR)-2 and N-formyl peptide receptors (FPRs), were clearly diminished in CatE(-/-)macrophages compared with those in wild-type cells. Consistently, chemotaxis of CatE(-/-)macrophages to MCP-1 and N-formyl-methionyl-leucyl-phenylalanine was also decreased. Similar to the chemotactic receptors, the surface expressions of the adhesion receptors CD18 (integrin beta(2)) and CD 29 (integrin beta(1)) in CatE(-/-) macrophages were significantly decreased, thereby reducing cell attachment of CatE(-/-) macrophages. These results indicate that the defects in chemotaxis and cell adhesion are likely to be involved in the imperfect function of CatE(-/-)macrophages

On the Physics Case of a Super Flavour Factory
We summarize the physics case of a high-luminosity e{sup +}e{sup -} flavor factory collecting an integrated luminosity of 50 - 75 ab{sup -1}. Many New Physics sensitive measurements involving B and D mesons and {tau} leptons, unique to a Super Flavor Factory, can be performed with excellent sensitivity to new particles with masses up to {approx} 100 (or even {approx} 1000) TeV. Flavor- and CP-violating couplings of new particles that may be discovered at the LHC can be measured in most scenarios, even in unfavorable cases assuming minimal flavor violation. Together with the LHC, a Super Flavor Factory, following either the SuperKEKB or the SuperB proposal, could be soon starting the project of reconstructing the New Physics Lagrangian

Characteristics of Cu stabilized Nb3Al strands with low Cu ratio
Characteristics of recently developed F4-Nb{sub 3}Al strand with low Cu ratio are described. The overall J{sub c} of the Nb{sub 3}Al strand could be easily increased by decreasing of the Cu ratio. Although the quench of a pulse-like voltage generation is usually observed in superconducting unstable conductor, the F4 strand with a low Cu ratio of 0.61 exhibited an ordinary critical transition of gradual voltage generation. The F4 strand does not have magnetic instabilities at 4.2 K because of the tantalum interfilament matrix. The overall J{sub c} of the F4 strand achieved was 80-85% of the RRP strand. In the large mechanical stress above 100 MPa, the overall J{sub c} of the F4 strand might be comparable to that of high J{sub c} RRP-Nb{sub 3}Sn strands. The Rutherford cable with a high packing factor of 86.5% has been fabricated using F4 strands. The small racetrack magnet, SR07, was also fabricated by a 14 m F4 cable. The quench current, I{sub q}, of SR07 were obtained 22.4 kA at 4.5 K and 25.2 kA at 2.2 K. The tantalum matrix Nb{sub 3}Al strands are promising for the application of super-cooled high-field magnets as well as 4.2 K operation magnets
