    EFFECT OF 12-O-TETRADECANOYLPHORBOL 13-ACETATE ON DNA SYNTHESIS IN PROLIFERATING RAT HEPATOCYTES IN PRIMARY CULTURE

    DNA synthesis in primary culture of rat hepatocytes was initiated over 16h after the beginning of culture and reached a peak between 32h and 40h under the conditions of this study. The addition of 12-O-tetradecanoylphorbol 13-acetate (TPA) (100 ng/ml) during the initial 24h of culture inhibited DNA synthesis, while addition between 16h or 20h and 24h stimulated DNA synthesis. Chelator-extractable and calcium-dependent protein kinase activity in the soluble fraction of hepatocytes with TPA addition during the initial 24h of culture was apparently reduced at 8, 12, 16 and 24h. It is suggested that TPA addition just prior to the initiation of DNA synthesis activates proten kinase C (PKC) and stimulates DNA synthesis, whereas TPA addition at an early stage of culture inhibits DNA synthesis, possibly because of PKC depletion

    Room-temperature reduction at SrRuO₃– metal interface in hydrogenous atmosphere detected by interface-sensitive resistance measurement

    Using interface-sensitive resistance measurement techniques, we detected the reducing reaction precursor at the interface between the metallic oxide SrRuO₃ and the electrodes under a hydrogenous atmosphere at room temperature. The interface resistance between this polycrystalline oxide and the electrodes (metallic pads or wires) clearly increased with the hydrogen present even at room temperature. In contrast, for bulk SrRuO₃, no increase in resistance was found. The rate of increase of the interface resistance depends on the electrode material. For example, that of SrRuO₃- Ag is larger than that of SrRuO₃-Cu, and the rate is related to the propensity for bulk oxide to reduce; Ag₂O is easier to reduce than CuO. The origin of the increase in interface resistance is posited to be the partial deficiency of oxygen in SrRuO₃. Our experiments suggest that the reduction at the interface of SrRuO₃ occurs at relatively low temperatures (room temperature) compared with the bulk reducing temperature of ~200℃ previously reported.     Local conductance spectra of itinerant ferromagnetic SrRuO3 through break junction

    We have measured the local differential conductance spectra (dI/dV-V) of an itinerant ferromagnet composed of polycrystalline SrRuO3 using the mechanically controllable break junction technique. Below the material's Curie temperature (T-C = 160 K), characteristic peak or dip conductance spectra are observed. The characteristic energy scale is comparable to the exchange spin splitting energy that is based on ferromagnetic band calculations. Both the peak and dip spectral shapes are explained based on the itinerant ferromagnetic characteristics of SrRuO3 in terms of spin-dependent transmission, which is similar to the giant magnetoresistance mechanism.     Ultrasound study of the solid-liquid transition and solid-liquid interface of 4He in aerogels

    金沢大学理工研究域数物科学系The freezing and melting of 4He in various aerogels with porosities ranging from 92 to 97% were studied using longitudinal ultrasound. The freezing pressure, detected from changes in velocity and attenuation, was elevated compared with that of the bulk by about 0.3 MPa, and showed a weak dependence on aerogel porosity. Inside the aerogels, there was a liquid phase, a solid phase or a coexisting state depending on temperature and pressure of the sample. We report the measurement of the transmission of sound through the solid-liquid interface in aerogel for the first time, which was independent of temperature and unaffected by a small addition of 3He, unlike that at the bulk interface. This indicates that sound attenuation at the interface is due to the disorder, originating from silica strands. Small amounts of 3He in the solid significantly decreased attenuation, because the resultant pinning of dislocations by 3He suppressed phonon scattering, as is observed in bulk solid 4He. © 2009 The Physical Society of Japan.

    Introduction: Kawasaki disease (KD) is a systemic vasculitis involving coronary arteries, sometimes resulting in aneurysms and myocardial infarction. Hyper-coagulability in the acute-phase of KD is indicated in some circumstances based on changes of individual clotting factors. Comprehensive coagulation assays, clot waveform analysis (CWA) and thrombin/plasmin generation assay (T/P-GA), have been developed to assess physiological hemostasis, but these techniques have not been applied in KD. Methods: We utilized both assays to analyze coagulation function in KD children (n = 42) prior to intravenous-immunoglobulin (IVIG) treatment (Pre), 1-week (1W) and 1-month (1M) post-IVIG. Results: In CWA, the clot time (CT) pre-treatment was prolonged, and was significantly shortened at 1W and 1M. However, the maximum coagulation velocity (|min1|) and acceleration (|min2|) were ~2-fold greater relative to controls, indicating an overall hypercoagulable tendency. These parameters were related to fibrinogen concentration, and were decreased at 1W and declined to normal at 1M. In T/P-GA, the endogenous potentials of thrombin and plasmin were greater relative to control at each of three time-points, and measurements at 1W were greater than those Pre-treatment. The ratios of TG and PG relative to control were similar, however, suggesting well-balanced dynamic coagulation and fibrinolysis. In non-responders to IVIG, the |min1| and |min2| measurements were greater than those in responders at 1W and 1M, suggesting that non-responders remained hypercoagulable after primary treatment. Conclusion: The coagulation data observed in KD were consistent with hypercoagulability, although fibrinolytic function appeared to be well-balanced.     EFFICACY OF INTERFERON THERAPY FOR CHRONIC HEPATITIS C : A COOPERATIVE STUDY IN ELEVEN HOSPITALS

    We investigated the influences of liver histology,serum levels of hepatitis C virus (HCV) and HCV genotypes on responsiveness to interferon (IFN) therapy in 342 patients with chronic hepatitis C. Either 9 million units (MU) of lymphoblastoid alpha IFN or 3 MU of recombinant IFN-alpha was administered daily for 2 weeks and then three times a week for 22 weeks. IFN responses were divided into three groups on the basis of the results of polymerase chain reaction (PCR) assay detecting HCV-RNA in serum. Complete response (CR) was defined as sustained elimination of HCV for at least 6 months after treatment,partial response (PR) as HCV elimination for a limited period,non-response (NR) as continuously positive for HCV-RNA in serum. Quantitation of pre-treament HCV-RNA amount in serum was determined by competitive PCR assay in 47 patients. HCV genotyping was performed in 114 patients by PCR with genotype-specific primers. CR was obtained in 97 patients (28.4%),PR in 104 (30.4%) and NR in 141 (41.2%). IFN responses,represented by CR/PR/NR,were 15/18/11 in 44 patients with chronic persistent hepatitis (CPH),72/65/73 in 210 patients with chronic aggressive hepatitis (CAH) 2a,and 10/21/57 in 88 patients with CAH2b. CR rate was lower in patients with CAH2b (11.4%) compared to those with CPH (34.1%) or CAH2a (34.3%). Averages of pre-treatment serum HCV-RNA amount (copies/50μl) were 10³·⁵⁵ in 13 CRs,10⁴·⁵⁶ in 17 PRs,and 10⁵·⁹⁵ in 17 NRs. There was a positive correlation between pre-treatment HCV-RNA levels and IFN unresponsiveness. HCV genotyping in 114 patients revealed that HCV type Ⅰ infection was observed in one,type Ⅱ in 94,type Ⅲ in 11,type Ⅳ in 6 and mixed (types Ⅱ and Ⅳ) in 2 patients,and their IFN responses (CR/PR/NR) were 0/0/1,28/26/40,3/5/3,1/3/2 and 0/1/1,respectively

    Five-year quality of life assessment after carbon ion radiotherapy for prostate cancer

    The aim of this study was to prospectively assess 5-year health-related quality of life (HRQOL) of patients treated with carbon ion radiotherapy (C-ion RT) for clinically localized prostate cancer. A total of 417 patients received carbon ion radiotherapy at a total dose of 63–66 Gray-equivalents (GyE) in 20 fractions over 5 weeks, and neoadjuvant and adjuvant androgen deprivation therapy (ADT) were administered for intermediate and high-risk patients. A HRQOL assessment was performed at five time points (immediately before the initiation of C-ion RT, immediately after, and at 12, 36 and 60 months after completion of C-ion RT) using Functional Assessment of Cancer Therapy (FACT) questionnaires. FACT-G and FACT-P scores were significantly decreased; however, the absolute change after 60 months was minimal. The transient decreases in the Trial Outcome Index (TOI) score returned to their baseline levels. Use of ADT, presence of adverse events, and biochemical failure were related to lower scores. Scores of subdomains of FACT instruments indicated characteristic changes. The pattern of HRQOL change after C-ion RT was similar to that of other modalities. Further controlled studies focusing on a HRQOL in patients with prostate cancer are warranted
