    Clonal spread of SCCmec type IV methicillin-resistant Staphylococcus aureus between community and hospital

    ABSTRACTThe staphylococcal chromosome cassette (SCC)mec types of 382 hospital-acquired methicillin-resistant Staphylococcus aureus (HA-MRSA) isolates in Taiwan were analysed over a 7-year period (1999–2005). There was an abrupt increase in SCCmec type IV in HA-MRSA during 2005. The molecular epidemiology of a subset (n = 69) of HA-MRSA isolates with SCCmec types III, IV or V was characterised and compared with that of community-acquired MRSA (CA-MRSA) (n = 26, collected during 2005). Pulsed-field gel electrophoresis revealed three major pulsotypes (A, B and C) and 15 minor clones. Pulsotypes B and C, which contained isolates carrying SCCmec types IV and V, respectively, included both CA-MRSA and HA-MRSA isolates. Among 24 toxin genes analysed, five genes had significant differential distribution between CA-MRSA and SCCmec type III HA-MRSA. Furthermore, among SCCmec type IV isolates, the seb gene was detected more commonly in HA-MRSA. Analysis of representative members of the three major pulsotypes by multilocus sequence typing revealed two sequence types (STs), namely ST239 (SCCmec III) and ST59 (SCCmec IV or SCCmec V). This suggests that ST59:SCCmec IV, which is usually community-acquired, has become an important nosocomial pathogen in the hospital studied

    Identification of differentially expressed genes induced by Bamboo mosaic virus infection in Nicotiana benthamiana by cDNA-amplified fragment length polymorphism

    Background: The genes of plants can be up- or down-regulated during viral infection to influence the replication of viruses. Identification of these differentially expressed genes could shed light on the defense systems employed by plants and the mechanisms involved in the adaption of viruses to plant cells. Differential gene expression in Nicotiana benthamiana plants in response to infection with Bamboo mosaic virus (BaMV) was revealed using cDNA-amplified fragment length polymorphism (AFLP). Results: Following inoculation with BaMV, N. benthamiana displayed differential gene expression in response to the infection. Isolation, cloning, and sequencing analysis using cDNA-AFLP furnished 90 cDNA fragments with eight pairs of selective primers. Fifteen randomly selected genes were used for a combined virus-induced gene silencing (VIGS) knockdown experiment, using BaMV infection to investigate the roles played by these genes during viral infection, specifically addressing the means by which these genes influence the accumulation of BaMV protein. Nine of the 15 genes showed either a positive or a negative influence on the accumulation of BaMV protein. Six knockdown plants showed an increase in the accumulation of BaMV, suggesting that they played a role in the resistance to viral infection, while three plants showed a reduction in coat protein, indicating a positive influence on the accumulation of BaMV in plants. An interesting observation was that eight of the nine plants showing an increase in BaMV coat protein were associated with cell rescue, defense, death, aging, signal transduction, and energy production. Conclusions: This study reports an efficient and straightforward method for the identification of host genes involved in viral infection. We succeeded in establishing a cDNA-AFLP system to help track changes in gene expression patterns in N. benthamiana plants when infected with BaMV. The combination of both DNA-AFLP and VIGS methodologies made it possible to screen a large number of genes and identify those associated with infections of plant viruses.     100學年度研究獎補助論文[[abstract]]Zinc oxide (ZnO) doped antimony (Sb) nanowires have been synthesized for improving ultraviolet sensing and photocatalytic properties. Upon illumination by UV light (365nm , 2.33mWcm−2 ), the photoelectric current of the ZnO:Sb nanowires exhibited a rapid photoresponse as compared to that of the ZnO nanowires. A highest ratio of photocurrent to dark current of around 48.8-fold was achieved in the as-synthesized ZnO:Sb nanowires. A UV-visible spectrophotometer was used to investigate the absorbance spectrum of the ZnO:Sb nanowires, which exhibited a high absorbance ratio with redshift effect in contrast to that of the ZnO nanowires.     A high performance dye-sensitized solar cell with a novel nanocomposite film of PtNP/MWCNT on the counter electrode

    An imide-functionalized material, poly(oxyethylene)-segmented polymer, was synthesized from the reaction of poly(oxyethylene) diamine of 2000 g mol(-1) M(w) and 4,4'-oxydiphthalic anhydride and used to disperse hybrid nanomaterials of platinum nanoparticles and multi-wall carbon nanotubes (PtNP/MWCNT). The composite material was spin-coated into film and further prepared as the counter electrode (PtNP/MWCNT-CE) for a dye-sensitized solar cell (DSSC). The short-circuit current density (J(SC)) and power-conversion efficiency (eta) of the DSSC with PtNP/MWCNT-CE were found to be 18.01 +/- 0.91 mA cm(-2) and 8.00 +/- 0.23%, respectively, while the corresponding values were 14.62 +/- 0.19 mA cm(-2) and 6.92 +/- 0.07% for a DSSC with a bare platinum counter electrode (Pt-CE). The presence and distribution of PtNP/MWCNT on the CE were characterized by using scanning electron microscopy (SEM), atomic force microscopy (AFM), energy dispersive spectroscopy (EDS), and X-ray diffraction (XRD). The attachment of PtNPs on MWCNTs was observed by transmission electron microscopy (TEM). Cyclic voltammetry (CV), incident-photo-to-current efficiency (IPCE) and electrochemical impedance spectra (EIS) were correlated to explain the efficacy of this nanocomposite system

    The effects of the bacterial interaction with visible-light responsive titania photocatalyst on the bactericidal performance

    Bactericidal activity of traditional titanium dioxide (TiO2) photocatalyst is effective only upon irradiation by ultraviolet light, which restricts the potential applications of TiO2 for use in our living environments. Recently carbon-containing TiO2 was found to be photoactive at visible-light illumination that affords the potential to overcome this problem; although, the bactericidal activity of these photocatalysts is relatively lower than conventional disinfectants. Evidenced from scanning electron microscopy and confocal Raman spectral mapping analysis, we found the interaction with bacteria was significantly enhanced in these anatase/rutile mixed-phase carbon-containing TiO2. Bacteria-killing experiments indicate that a significantly higher proportion of all tested pathogens including Staphylococcus aureus, Shigella flexneri and Acinetobacter baumannii, were eliminated by the new nanoparticle with higher bacterial interaction property. These findings suggest the created materials with high bacterial interaction ability might be a useful strategy to improve the antimicrobial activity of visible-light-activated TiO2

    Nanoscale Processing by Adaptive Laser Pulses

    We theoretically demonstrate that atomically-precise ``nanoscale processing" can be reproducibly performed by adaptive laser pulses. We present the new approach on the controlled welding of crossed carbon nanotubes, giving various metastable junctions of interest.     Urban-rural disparity of preventive healthcare utilisation among children under the universal health insurance coverage in Taiwan: a national birth cohort analysis

    Objective: In the context of universal health insurance coverage, this study aimed to determine whether urbanerural inequality still exists in preventive health care (PHC) amongst children in Taiwan. Study design: Prospective cohort study. Methods: A total of 184,117 mothers and their children born in 2009 were identified as the study cohort. The number of children born in urban, satellite and rural areas was 40,176, 57,565 and 86,805, respec-tively. All children were followed for 7 years, before which a total of seven times PHC were provided by Taiwan's National Health Insurance (NHI) programme. Ordinal logistic regression models were used to associate urbanisation level with the frequency of PHC utilisation. Stratified analyses were further per-formed in accordance with the children's birth weight and the mothers' birthplace. Results: Children from satellite areas had higher utilisation for the first four scheduled PHC visits. Children living in urban areas received more PHC for the fifth and sixth scheduled visits. Compared with those from rural areas, children in satellite areas exhibited a small but significant increase in odds in PHC utilisation, with a covariate-adjusted odds ratio (aOR) of 1.04 and 95% confidence interval (CI) of 1.02 e1.06. By contrast, no significant difference was observed between rural and urban areas (aOR ¼ 1.01). Further stratified analyses suggest more evident urbanerural difference in PHC utilisation amongst children with low birth weight and foreign-born mothers. Conclusions: Given a universal health insurance coverage and embedded mechanisms in increasing the availability of healthcare resources in Taiwan, a slight urbanerural difference is observed in PHC uti-lisation amongst children. Hence, sociodemographic inequality in utilisation of PHC still exists. This issue should be addressed through policy intervention

    Acute anal stretch inhibits NMDA-dependent pelvic-urethra reflex potentiation via spinal GABAergic inhibition in anesthetized rats

    The impact of acute anal stretch on the pelvic-urethra reflex potentiation was examined in urethane-anesthetized rats by recording the external urethra sphincter electromyogram activity evoked by the pelvic afferent stimulation. Test stimulation (1 stimulation/30 s) evoked a baseline reflex activity with a single action potential that was abolished by gallamine (5 mg/kg iv). On the other hand, the repetitive stimulation (1 stimulation/1 s) induced spinal reflex potentiation (SRP) that was attenuated by intrathecal 6-cyano-7-nitroquinoxaline- 2,4-dione (a glutamatergic alpha-amino-3-hydroxy-5-methyl- 4-isoxazoleproprionat receptor antagonist, 100 mu M, 10 mu l) and D-2-amino-5-phosphonovalerate [a glutamatergic N-methyl-D-aspartate (NMDA) antagonist, 100 mu M, 10 mu l]. Acute anal stretch using a mosquito clamp with a distance of 4 mm exhibited no effect, whereas distances of 8 mm attenuated and 12 mm abolished the repetitive stimulation-induced SRP. Intrathecal NMDA (100 mu M, 10 mu l) reversed the abolition on SRP caused by anal stretch. On the other hand, pretreated bicuculline [gamma-aminobutyric acid (GABA) A receptor antagonist, 100 mu M, 10 mu l] but not hydroxysaclofen (GABA(B) receptor antagonist) counteracted the abolition on the repetitive stimulation-induced SRP caused by the anal stretch. All of the results suggested that anal stretch may be used as an adjunct to assist voiding dysfunction in patients with overactive urethra sphincter and that GABAergic neurotransmission is important in the neural mechanisms underlying external urethra sphincter activity inhibited by anal stretch
