    Navigating the Existential Crisis from Literature to Real Life: A Text-to-Self Pedagogical Approach and Its Potential for Existential Literature Instruction

    Amidst the existential crisis in contemporary society, young individuals worldwide find themselves susceptible to self-inflicted harm or even self-destruction in response to their psychological ruptures and injuries, while lacking the requisite self-awareness and the patience to persevere on the journey of self-understanding. In this article, we draw upon Rosenblattā€™s (1995) notion of Literature as Exploration to propound a text-to-self pedagogical approach to reading Haruki Murakami and Can Xue and to teaching contemporary existentialist literature. Comprising three strategies, namely The Art World in My Eyes, The Mind Film, and The Literary Conversation, this approach encourages students to engage deeply with the portrayal of death in the writersā€™ flagship novels that view death as an unwavering companion to young individualsā€™ life journey and growth. Based on Rosenblatt (1995), the three strategies, and our survey-based analysis of 2,000 Vietnamese high school studentsā€™ existential perceptions, we propose a model for literature instruction through promoting studentsā€™ transition from literary interpretations to self-discovery and self-realization of death associated with the existential crisis. This four-stage model, we expect, will get students poised to reconcile with their lost, solitary, and vulnerable ego, actively befriend their ā€˜inner childā€™, and develop into individuals with healed hearts and positive worldviews

With hardworking process to deal with pragmatics, learners' language power of word usage is extremely widened. Not only do they can use target language flexibly, but they are also more confident in conversational English with less anxieties of unexpected interpretation.     Granulocyte colony-stimulating factor reduces biliary fibrosis and ductular reaction in a mouse model of chronic cholestasis

    Background: Biliary atresia is a rare congenital bile duct disease that is the leading cause of liver fibrosis in neonates. Granulocyte colony-stimulating factor (GCSF) is a potential therapy for hepatocellular diseases, but data on GCSF for cholestatic conditions remain limited. Materials and methods: The current study examines the role of GCSF in improving bile duct obstruction in mice. Two doses were administered: 10.0Ā Ī¼g/kg/day and 61.5Ā Ī¼g/kg/day, which is the animal equivalent dose of 5.0Ā Ī¼g/kg in humans. Seven days (D7) after bile duct ligation (BDL), Swiss mice were treated with phosphate buffered saline or GCSF for 5 days. The intrahepatic adaptive response of BDL mice was evaluated on postsurgical days D12, D19, and D26. Results: Treatment with 61.5Ā Ī¼g/kg of GCSF resulted in a significant increase in circulating leukocytes and neutrophils on D12. Amelioration of liver injury, as shown by reduced aspartate aminotransferase levels, increased albumin levels and survival rate, as well as reduced intrahepatic inflammation and hepatic myeloperoxidase expression, downregulated ductular proliferation, periportal fibroblast activation, and fibrosis, enhanced expressions of hepatocyte growth factor, peroxisome proliferator-activated receptor-alpha, and ki67, and suppressed expression of cleaved caspase-3 protein, was noted after treatment with 61.5Ā Ī¼g/kg of GCSF. Additionally, GCSF treatment was associated with an increased number of intrahepatic cd3-Sca1+c-Kit+ bone marrow cells. Conclusions: Treatment with 61.5Ā Ī¼g/kg of GCSF resulted in liver regeneration and survival in BDL mice was seen, suggesting its potential use for human liver diseases

    Simulated Microgravity Induces the Proliferative Inhibition and Morphological Changes in Porcine Granulosa Cells

    Astronauts are always faced with serious health problems during prolonged spaceflights. Previous studies have shown that weightlessness significantly affects the physiological function of female astronauts, including a change in reproductive hormones and ovarian cells, such as granulosa and theca cells. However, the effects of microgravity on these cells have not been well characterized, especially in granulosa cells. This study aimed to investigate the effects of simulated microgravity (SMG) on the proliferation and morphology of porcine granulosa cells (pGCs). pGC proliferation from the SMG group was inhibited, demonstrated by the reduced O.D. value and cell density in the WST-1 assay and cell number counting. SMG-induced pGCs exhibited an increased ratio of cells in the G0/G1 phase and a decreased ratio of cells in the S and G2/M phase. Western blot analysis indicated a down-regulation of cyclin D1, cyclin-dependent kinase 4 (cdk4), and cyclin-dependent kinase 6 (cdk6), leading to the prevention of the G1-S transition and inducing the arrest phase. pGCs under the SMG condition showed an increase in nuclear area. This caused a reduction in nuclear shape value in pGCs under the SMG condition. SMG-induced pGCs exhibited different morphologies, including fibroblast-like shape, rhomboid shape, and pebble-like shape. These results revealed that SMG inhibited proliferation and induced morphological changes in pGCs

    Kinetics of CD4+ T Helper and CD8+ Effector T Cell Responses in Acute Dengue Patients

    Background: The protective or pathogenic role of T lymphocytes during the acute phase of dengue virus (DENV) infection has not been fully understood despite its importance in immunity and vaccine development. Objectives: This study aimed to clarify the kinetics of T lymphocyte subsets during the clinical course of acute dengue patients. Study design: In this hospital-based cohort study, 59 eligible Vietnamese dengue patients were recruited and admitted. They were investigated and monitored for T cell subsets and a panel of clinical and laboratory parameters every day until discharged and at post-discharge from the hospital. Results: We described for the first time the kinetics of T cell response during the clinical course of DENV infection. Severe cases showed significantly lower levels of effector CD8+ T cells compared to mild cases at day ?1 (p = 0.017) and day 0 (p = 0.033) of defervescence. After defervescence, these cell counts in severe cases increased rapidly to equalize with the levels of mild cases. Our results also showed a decline in total CD4+ T, Th1, Th1/17 cells during febrile phase of dengue patients compared to normal controls or convalescent phase. On the other hand, Th2 cells increased during DENV infection until convalescent phase. Cytokines such as interferon-Ī³, IL-12p70, IL-5, IL-23, IL-17A showed tendency to decrease on day 0 and 1 compared with convalescence and only IL-5 showed significance indicating the production during acute phase was not systemic.Conclusion: With a rigorous study design, we uncovered the kinetics of T cells in natural DENV infection. Decreased number of effector CD8+ T cells in the early phase of infection and subsequent increment after defervescence day probably associated with the T cell migration in DENV infection

    Regulation of microtubule dynamics is required to properly control various steps of neurodevelopment. In this study, we identified granule cell antiserum-positive 14 (Gcap14) as a microtubule plus-end-tracking protein and as a regulator of microtubule dynamics during neurodevelopment. Gcap14 knockout mice exhibited impaired cortical lamination. Gcap14 deficiency resulted in defective neuronal migration. Moreover, nuclear distribution element nudE-like 1 (Ndel1), an interacting partner of Gcap14, effectively corrected the downregulation of microtubule dynamics and the defects in neuronal migration caused by Gcap14 deficiency. Finally, we found that the Gcap14ā€“Ndel1 complex participates in the functional link between microtubule and actin filament, thereby regulating their crosstalks in the growth cones of cortical neurons. Taken together, we propose that the Gcap14ā€“Ndel1 complex is fundamental for cytoskeletal remodeling during neurodevelopmental processes such as neuronal processes elongation and neuronal migration. 11Nsciescopu
